Computational investigations were performed to examine the effects of the addition of 2-acetamido-2-deoxy-β-D-galactopyranosylamine or 1-amino-1-deoxy-D-glucitol connected to the C-terminus of vancomycin with different linkers. The purpose of this modification was to find more effective vancomycin derivatives by providing alternative interactions between vancomycin moiety and the peptidoglycan precursor. Each prepared vancomycin–peptidoglycan complex was optimized and submitted to the molecular dynamics study and analysis. The analysis of overall root mean square deviation, changes in position and interactions involving modified part of vancomycin as well as cluster analysis were carried out. One of the proposed vancomycin analogues seems to be efficient vancomycin substitute.  相似文献

This article describes numerous attempts of the SN reaction of chlorine atom in chloromethyl group with nucleophilic compounds, such as the following: phenols, thiophenols, and amino compounds. The influences of ratios of nitroimidazodihydrooxazoles, nucleophiles and basic agent, and the polarity of solvent on the kind of formed products were established. Also, the comparison of reactivity with nucleophiles of close structural isomers of nitroimidazodihydrooxazoles has been made. A series of new analogs of nitrogen mustards (4a–4h) containing the 1,3,5-triazine ring substituted with dipeptide residue were synthesized and evaluated for the inhibition of both acetylcholinesterase (AChE) and β-secretase (BACE1) enzymes. The AChE inhibitory activity studies were carried out using Ellman’s colorimetric method, and the BACE1 inhibitory activity studies were carried out using fluorescence resonance energy transfer (FRET). All compounds displayed considerable AChE and BACE1 inhibition. The most active against both AChE and BACE1 enzymes were compounds A and 4a, with an inhibitory concentration of AChE IC50 = 0.051 µM; 0.055 µM and BACE1 IC50 = 9.00 µM; 11.09 µM, respectively.  相似文献

A series of silicate–phosphate glass materials from the SiO2-P2O5-K2O-MgO system (pure and doped with sulfur ions) were synthesized by melting raw material mixtures that contained activated carbon as a reducer. The bulk composition of glass was confirmed with X-ray fluorescence spectroscopy. The homogeneity of the glass was confirmed through elemental mapping at the microstructural level with scanning electron microscopy combined with an analysis of the microregions with energy-dispersive X-ray spectroscopy. The structural and optical properties of the glass were studied by using spectroscopic techniques. The infrared spectroscopy studies that were conducted showed that the addition of sulfur caused changes in the silicate–phosphate networks, as they became more polymerized, which was likely related to the accumulation of potassium near the sulfur ions. By using irradiation with an optical parametric oscillator (OPO) nanosecond laser system operating at the second harmonic wavelength, the glass samples emitted a wide spectrum of luminescence, peaking at about 700 nm when excited by UV light (210–280 nm). The influence of the glass composition and the laser-processing parameters on the emission characteristics is presented and discussed. This work also referred to the density, molar volume, and theoretical optical basicity of pure and sulfur-doped glass.  相似文献

Functionalisation of synthetic polymers by using enzymes has been recently demonstrated. The major advantage of enzymes over chemical processes lies in their surface specific and endo-wise mode of action. Surface hydrophilisation of PET with lipases and cutinases leads to a dramatic increase of the surfacial acid and hydroxyl group content while conventional chemical treatment does not cause any change. However, this PET-hydrolysing activity by enzymes from distinct classes has not yet been correlated to activity on natural polyesters. Here, we show that lipases, cutinases and a PHA-depolymerase are all capable of hydrolysing PET, while only lipases and cutinases also hydrolysed cutin to various degrees. Lipases showed a higher specificity for terminal fatty acids while the cutinases preferred hydroxy fatty acids during cutin hydrolysis.  相似文献

The effect of pressure on the luminescence spectrum of the weak field system Cr3+ : LiTaO3 is presented. By using pressure to induce a low to high crystal field strength transition, we demonstrate the ability of pressure to identify distinct dopant bonding environments in luminescent materials.  相似文献
