(1) Background: The obesity epidemic has been drastically progressing in both children and adults worldwide. Pharmacotherapy is considered necessary for its treatment. However, many anti-obesity drugs have been withdrawn from the market due to their adverse effects. Instead, natural products (NPs) have been studied as a source for drug discovery for obesity, with the goal of limiting the adverse effects. Zebrafish are ideal model animals for in vivo testing of anti-obesity NPs, and disease models of several types of obesity have been developed. However, the evidence for zebrafish as an anti-obesity drug screening model are still limited. (2) Methods: We performed anti-adipogenic testing using the juvenile zebrafish obesogenic test (ZOT) and mouse 3T3-L1 preadipocytes using the focused NP library containing 38 NPs and compared their results. (3) Results: Seven and eleven NPs reduced lipid accumulation in zebrafish visceral fat tissues and mouse adipocytes, respectively. Of these, five NPs suppressed lipid accumulation in both zebrafish and 3T3-L1 adipocytes. We confirmed that these five NPs (globin-digested peptides, green tea extract, red pepper extract, nobiletin, and Moringa leaf powder) exerted anti-obesity effects in diet-induced obese adult zebrafish. (4) Conclusions: ZOT using juvenile fish can be a high-throughput alternative to ZOT using adult zebrafish and can be applied for in vivo screening to discover novel therapeutics for visceral obesity and potentially also other disorders.  相似文献

Some new coordination polymers have been synthesized by combining adipyl bis-biuret and azelyl bis-biuret with metals of first transition series viz Mn(II), Co(II), Ni(II), Cu(II) and Zn(II). They were characterized by elemental analysis, infrared and reflectance spectral and magnetic studies. A detailed thermal study was carried out and thermal stability compared. We describe herein the preparation of 24 pure asparagine-linked oligosaccharides (Asn-oligosaccharides) from asparagine-linked biantennary complex-type sialylundecasaccharide [(NeuAc-alpha-2,6-Gal-beta-1,4-GlcNAc-beta-1,2-Man-alpha-1,6/1,3-)(2)-Man-beta-1,4-GlcNAc-beta-1,4-GlcNAc-beta-1-asparagine, 2] obtained from egg yolk. Our synthetic strategy aimed at adapting branch specific exo-glycosidases digestion (beta-D-galactosidase, N-acetyl-beta-D-glucosaminidase and alpha-D-mannosidase) of the individual asialo-branch after preparation of monosialyloligosaccharides obtained from 2 by acid hydrolysis of NeuAc. In order to perform branch specific exo-glycosidase digestion, isolation of pure monosialyloligosaccharides obtained was essential. However, isolation of two kinds of monosialyloligosaccharides are difficult by HPLC due to their highly hydrophilic nature. Therefore, we examined chemical protection with hydrophobic protecting (Fmoc and benzyl) groups. These chemical protection enabled us to separate the monosialyloligosaccharides by use of a HPLC column (ODS) on synthetic scales. Using these pure monosialiloligosaccharides enable us to obtain 24 Asn-linked oligosaccharides (100 mg scale) within a few weeks by branch specific exo-glycosidase digestions (alpha-D-neuraminidase, beta-D-galactosidase, N-acetyl-beta-D-glucosaminidase and alpha-D-mannosidase). In addition, solid-phase synthesis of glycopeptide having Asn-linked sialyl-undeca- and asialo-nonasaccharides thus obtained, was also performed on an acid labile HMPA-PEGA resin.  相似文献

The refractive-index contrast in dielectric multilayer structures, optical resonators, and photonic crystals is an important figure of merit that creates a strong demand for high-quality thin films with a low refractive index. A SiO2 nanorod layer with low refractive index of n = 1.08, to our knowledge the lowest ever reported in thin-film materials, is grown by oblique-angle electron-beam deposition of SiO2. A single-pair distributed Bragg reflector employing a SiO2 nanorod layer is demonstrated to have enhanced reflectivity, showing the great potential of low-refractive-index films for applications in photonic structures and devices.  相似文献

p-tert-Butylcalix[5]arene crystallizes from ethyl acetate in the monoclinic space groupC2/c witha=34.793(9),b=13.637(3),c=25.577(7), =114.28(2)°, andD c =1.08 g cm–3 forZ=8 (C59H78O7). Refinement based on 2789 observed reflections led toR=0.071. The complex crystallizes with one molecule of ethyl acetate per calixarene molecule, with the solvent residing between calixarene units instead of within the cavity. The calixarene units neighbor-include to form a polymeric array: a t-butyl group of one calixarene is housed in the cavity of an adjacent calixarene. The packing arrangement of the calixarene molecules bears a remarkable resemblance to a zeolitic array.  相似文献

We consider the problem of efficient estimation of tail probabilities of sums of correlated lognormals via simulation. This problem is motivated by the tail analysis of portfolios of assets driven by correlated Black-Scholes models. We propose two estimators that can be rigorously shown to be efficient as the tail probability of interest decreases to zero. The first estimator, based on importance sampling, involves a scaling of the whole covariance matrix and can be shown to be asymptotically optimal. A further study, based on the Cross-Entropy algorithm, is also performed in order to adaptively optimize the scaling parameter of the covariance. The second estimator decomposes the probability of interest in two contributions and takes advantage of the fact that large deviations for a sum of correlated lognormals are (asymptotically) caused by the largest increment. Importance sampling is then applied to each of these contributions to obtain a combined estimator with asymptotically vanishing relative error.  相似文献

Diffusion spectrum imaging (DSI) is capable of resolving crossing and touching fiber bundles in a given voxel. Acquisition of DSI data involves sampling large number of points in the q-space which significantly increases scan times. The scan times can be reduced by exploiting the symmetry of the q-space. In this study the fiber pathways for five (fornix, cingulum, superior longitudinal fasciculus, corticospinal tract, and crossing fibers in the centrum semiovale region) fiber bundles derived using three subsampled data sets of different sizes derived from the 257 samples in the q-space are compared. The coefficient of variation of the ratio of the number of fiber pathways for each subsample data set to the original data points, averaged over all the 10 subjects, was used for quantitatively investigating the effect of subsampling on the tractography. The effect of threshold angles on tractography is also investigated. The effect of subsampling on the orientation distribution function (ODF) was quantitatively evaluated using both scalar and vector measures derived from the ODF. A streamline tractography method that improves the curvature problem and reduces the local truncation error to further improve the mapping of fiber pathways is adapted. Analysis of the fiber pathways in ten normal subjects, based on qualitative and quantitative methods, shows that the 129 and 198 q-space points provide very similar result with angle of threshold between 41° and 45°. Based on the scan time advantage, 129 subsampled points appear to be adequate for tractography.  相似文献

Abstract

Delipidated egg yolk (DEY) was digested with protease. The digest was ultrafiltered using a molecular weight cut-off of 10,000, and the UF permeate was treated with a reverse osmosis membrane. The resulting sialylglycopeptides-rich fraction was purified by the combination of anion exchange and gel filtration chromatography. Two major sialylglycopeptides, A-I and A-II, were characterized as biantennary complex type sialylglycopeptides having one or two N-acetylneuraminic acid and 1 - 3.5 amino acids including one asparagine residue by composition analyses and 1H NMR spectroscopy.  相似文献
