A commercial dissolving pulp was treated with aqueous solutions containing 3, 5 and 7 % of an organometalic complex (nitren) with the aim to selectively extract xylan and study its impact on the conventional physical–chemical properties of the pulp. The influence of these treatments on the pulp dissolution in a moderate solvent (8 % NaOH aqueous solution) was assessed by measuring the dissolution yields and the dissolution mechanisms. The results of this study show that nitren treatment has the effect of removing a large part of the xylan present in a dissolving pulp. It is also removing mannans and most important, it is influencing cellulose in two ways, (1) extracting it with more intensity when the nitren concentration increases, and (2) decreasing its mean molecular mass, also more evident with nitren concentration increase. The nitren extractions are favourable for the dissolution in cold NaOH–water, being more effective with higher concentrations. This chemical modification of the fiber surface leads to the disassembly of the primary wall. This allows an easier access of the NaOH reagent to regions not accessible on the initial fibres, which with the decrease of the cellulose molecular weight allows an easier dissolution and gives different dissolution mechanisms.  相似文献

We experimentally test a precommitment mechanism for the trust game. Before the investor’s decision, the allocator places an amount into escrow, to be forfeited if he keeps the proceeds of investment for himself. We vary the available escrow amounts—in particular, whether there is a high amount that gives rise to an efficient equilibrium—and whether escrow is voluntary or imposed. We find that when chosen, the high escrow amount does lead to efficient outcomes. We also find substantial investment when the high amount is unavailable or not chosen, though well below that when it is chosen, and declining over time. We find only weak evidence for behavioral theories, such as crowding out and signaling. These results are seen when escrow choices are imposed as well as when they are voluntary.   相似文献

More and more attention is being focused on the analysis of post-translational modifications (PTMs) on proteins as researchers are continually learning how essential they are for proper cellular function. As there are hundreds of different types of known PTMs, traditional methods of modification analysis are incapable of comprehensively monitoring for post-translational modifications, a task which is a necessity for truly understanding a cell's biology. This review highlights recent developments in novel multiplexed methods of PTM analysis including: fluorescent stain and immuno-based methods, hardware-based mass spectrometric methods and computational-based mass spectrometric methods. Many of these techniques show great promise and will likely be a valuable resource for the biological community.  相似文献

A practical and efficient synthesis of spirobarbiturates of type III is reported when NH acidity of the imide function of the hydrophilic linker element allowed the introduction of different substituents. Structural characterization, which was based on both X-ray crystallography and spectroscopic investigations, indicated type II beta-turn formation. Introduction of the molecular scaffold into solid phase peptide synthesis gave rise to spirocyclic neuropeptide analogs.  相似文献

The ability to control and pump high ionic strength fluids inside microchannels forms a major advantage for clinical diagnostics and drug screening processes, where high conductive biological and physiological buffers are used. Despite the known potential of AC electro‐thermal (ACET) effect in different biomedical applications, comparatively little is known about controlling the velocity and direction of fluid inside the chip. Here, we proposed to discretize the conventional electrodes to form various asymmetric electrode structures in order to control the fluid direction by simple switching the appropriate electric potential applied to the discretized electrodes. The ACET pumping effect was numerically studied by solving electrical, thermal and hydrodynamic multi‐physic coupled equations to optimize the geometrical dimensions of the discretized system. PBS solutions with different ionic strength were seeded with 1 μm sized fluorescent particles and electrothermally driven fluid motion was observed inside the channel for different electrode structures. Experimental analyses confirm that the proposed micropump is efficient for a conductivity range between 0.1 and 1 S/m and the efficiency improves by increasing the voltage amplitude. Behavior of the proposed electrode–electrolyte system is discussed by lumped circuit model. Frequency response of system illustrated that the optimal frequency range increases by increasing the conductivity of medium. For 0.18 S/m PBS solution, the constant pumping effect was observed at frequency range between 100 kHz and 1 MHz, while frequency range of 100 kHz to 5 MHZ was observed for 0.42 S/m. The characteristics of experimental results were in good agreement with the theoretical model.  相似文献

Post-translational modifications (PTMs) of proteins are essential for proper function, as they regulate many aspects of a protein's activity and interaction with substrates. When analyzing modified peptides derived from such proteins by mass spectrometry, these modifications can dissociate, producing either a marker ion or neutral loss characteristic of the modification, which have conventionally been monitored with a precursor ion scan or neutral loss scan, respectively. Although powerful, both precursor ion scans and neutral loss scans can only screen for one particular modification at a time. This has led to the development of multiple neutral loss monitoring (MNM) for neutral losses and multiple precursor ion monitoring (MPM) for marker ions on electrospray instruments. Here, we report their implementation on a matrix-assisted laser desorption/ionization (MALDI) instrument as well as the inception of a novel scan strategy termed targeted multiple precursor ion monitoring (tMPM). This latter scan strategy has been developed on a MALDI tandem time-of-flight (TOF/TOF) mass spectrometer for the identification of multiple PTMs via their associated marker ions by manipulating certain components of the instrument, notably the timed ion selector and the delayed extraction source 2. Targeted MPM combined with a second approach, multiple neutral loss monitoring (MNM), is shown to be a successful approach in the identification of PTMs, identifying multiple modified peptides in a complex sample matrix.  相似文献

The coordination polymer CPO-27-Mg was rapidly synthesized under microwave irradiation. This material exhibits a sufficiently high drug loading towards aspirin (~8% wt.) and paracetamol (~14% wt.). The binding of these two molecules with the inner surface of the metal-organic framework was studied employing the Gaussian and Plane Wave approach of the Density Functional Theory. The structure of CPO-27-Mg persists after the adsorption of aspirin or paracetamol and their desorption energies, being quite high, decrease under solvent conditions.  相似文献

Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) and thin-layer chromatography (TLC) have been used to study dopamine and iron mediated free-radical transformation of lipids in their hydrophilic parts. It has been shown that the action of the dopamine/Fe2+ system on galactocerebroside or cardiolipin, which are the components of mixed micelles, results in formation of ceramide or phosphatidic acid and phosphatidylhydroxyacetone, respectively. These data, when combined with results obtained using the ascorbate/Fe2+/H2O2 oxidizing system with the same substrates, demonstrate that the formation of these products proceeds via an OH-radical induced fragmentation taking place in polar moiety of the starting lipids.   相似文献
