Metabolites can be an important read-out of disease. The identification and validation of biomarkers in the cancer metabolome that can stratify high-risk patients is one of the main current research aspects. Mass spectrometry has become the technique of choice for metabolomics studies, and mass spectrometry imaging (MSI) enables their visualization in patient tissues. In this study, we used MSI to identify prognostic metabolite biomarkers in high grade sarcomas; 33 high grade sarcoma patients, comprising osteosarcoma, leiomyosarcoma, myxofibrosarcoma, and undifferentiated pleomorphic sarcoma were analyzed. Metabolite MSI data were obtained from sections of fresh frozen tissue specimens with matrix-assisted laser/desorption ionization (MALDI) MSI in negative polarity using 9-aminoarcridine as matrix. Subsequent annotation of tumor regions by expert pathologists resulted in tumor-specific metabolite signatures, which were then tested for association with patient survival. Metabolite signals with significant clinical value were further validated and identified by high mass resolution Fourier transform ion cyclotron resonance (FTICR) MSI. Three metabolite signals were found to correlate with overall survival (m/z 180.9436 and 241.0118) and metastasis-free survival (m/z 160.8417). FTICR-MSI identified m/z 241.0118 as inositol cyclic phosphate and m/z 160.8417 as carnitine.

The relaxation of slightly poly-disperse linear flexible polymers has been expressed in a simplified blending rule which is presumed to be a weighted linear superposition of the relaxation spectra of mono-disperse components which constitute the blend. Discrete components are characterized by their molecular weight M i,weight fraction w i,and relaxation time spectrum H i(). ). In contrast to broadly distributed blends in which the small molecules mobilize the large ones and vice versa, we introduce the term slightly polydisperse for blends with molecular weight distributions narrow enough to have very little change in the longest relaxation times of each molecular weight component. The properties of this blending rule are analyzed and dynamic data is calculated for slightly poly-disperse polystyrene. As an application, the blending rule is used to determine the characteristic mono-disperse parameters (BSW parameters) of two materials, poly (vinyl methyl ether) and polycarbonate, for which we could not determine their BSW parameters directly since they were not available in nearly monodisperse form. Diversity oriented synthesis of fused pyrimidines leads to scaffolds with many biological activities. In the case of the preparation of pyrido[2,3-d]pyrimidines from 2-alkylthiopyrimidines, the formation of a new carbon-carbon bond at C5 is required, a reaction, that is, very limited in scope. However Claisen type rearrangement of simple 4-allylic ethers affords C5 substituted pyrimidines readily; in cases with an ester substituent, rearrangement occurs at room temperature. Subsequent cyclisation to afford 6-methylpyrido[2,3-d]pyrimidin-7(8H)-ones was achieved in high yield. Using allylic ethers derived from 3-chloromethyl-4-arylbut-3-en-2-ones as substrates, a new titanium[IV]chloride catalysed reaction affording 6-arylmethyl-7-methylpyrido[2,3-d]pyrimidines was discovered. In contrast, 2-alkylthiopteridines are readily available. In both cases, substitution at C2 and C4 to generate diversity has been carried out and the reactivity compared; yields of substitution products were generally higher with pteridine substrates. In biological assays unexpected hits were found for activity against the Gram positive bacterium, Nocardia farcinia, and against the parasite Trypanosoma brucei brucei, illustrating the value of diversity oriented synthesis in the discovery of biologically active compounds.  相似文献

The sequestration of industrially emitted CO(2) in gas hydrate reservoirs has been recently discussed as an option to reduce atmospheric greenhouse gas. This CO(2) contains, despite much effort to clean it, traces of impurities such as SO(2) and NO(2) . Here, we present results of a pilot study on CO(2) hydrates contaminated with 1% SO(2) or 1% NO(2) and show the impact on hydrate formation and stability. Microscopic observations show similar hydrate formation rates, but an increase in hydrate stability in the presence of SO(2). Laser Raman spectroscopy indicates a strong enrichment of SO(2) in the liquid and hydrate phase and its incorporation in both large and small cages of the hydrate lattice. NO(2) is not verifiable by laser Raman spectroscopy, only the presence of nitrate ions could be confirmed. Differential scanning calorimetry analyses show that hydrate stability and dissociation enthalpy of mixed CO(2)-SO(2) hydrates increase, but that only negligible changes arise in the presence of NO(2) impurities. X-ray diffraction data reveal the formation of sI hydrate in all experiments. The conversion rates of ice+gas to hydrate increase in the presence of SO(2), but decrease in the presence of NO(2). After hydrate dissociation, SO(2) and NO(2) dissolved in water and form strong acids.  相似文献

The NiII complexes [Ni([9]aneNS2‐CH3)2]2+ ([9]aneNS2‐CH3=N‐methyl‐1‐aza‐4,7‐dithiacyclononane), [Ni(bis[9]aneNS2‐C2H4)]2+ (bis[9]aneNS2‐C2H4=1,2‐bis‐(1‐aza‐4,7‐dithiacyclononylethane) and [Ni([9]aneS3)2]2+ ([9]aneS3=1,4,7‐trithiacyclononane) have been prepared and can be electrochemically and chemically oxidized to give the formal NiIII products, which have been characterized by X‐ray crystallography, UV/Vis and multi‐frequency EPR spectroscopy. The single‐crystal X‐ray structure of [NiIII([9]aneNS2‐CH3)2](ClO4)6?(H5O2)3 reveals an octahedral co‐ordination at the Ni centre, while the crystal structure of [NiIII(bis[9]aneNS2‐C2H4)](ClO4)6?(H3O)3? 3H2O exhibits a more distorted co‐ordination. In the homoleptic analogue, [NiIII([9]aneS3)2](ClO4)3, structurally characterized at 30 K, the Ni? S distances [2.249(6), 2.251(5) and 2.437(2) Å] are consistent with a Jahn–Teller distorted octahedral stereochemistry. [Ni([9]aneNS2‐CH3)2](PF6)2 shows a one‐electron oxidation process in MeCN (0.2 M NBu4PF6, 293 K) at E1/2=+1.10 V versus Fc+/Fc assigned to a formal NiIII/NiII couple. [Ni(bis[9]aneNS2‐C2H4)](PF6)2 exhibits a one‐electron oxidation process at E1/2=+0.98 V and a reduction process at E1/2=?1.25 V assigned to NiII/NiIII and NiII/NiI couples, respectively. The multi‐frequency X‐, L‐, S‐, K‐band EPR spectra of the 3+ cations and their 86.2 % 61Ni‐enriched analogues were simulated. Treatment of the spin Hamiltonian parameters by perturbation theory reveals that the SOMO has 50.6 %, 42.8 % and 37.2 % Ni character in [Ni([9]aneNS2‐CH3)2]3+, [Ni(bis[9]aneNS2‐C2H4)]3+ and [Ni([9]aneS3)2]3+, respectively, consistent with DFT calculations, and reflecting delocalisation of charge onto the S‐thioether centres. EPR spectra for [61Ni([9]aneS3)2]3+ are consistent with a dynamic Jahn–Teller distortion in this compound.  相似文献

Introduction of a β-electron withdrawing group to cycloalkanones allows facile C-C bond fragmentation. The reaction has been demonstrated with a large range of ring sizes, bearing various leaving and electron withdrawing groups, and using a variety of nitrogen and oxygen containing nucleophiles (>30 examples). The application of fragmentation products to the preparation of substituted γ-lactones has been demonstrated. Mechanistic studies are reported which are suggestive of a Grob/Eschenmoser type reaction.  相似文献

An analytical method for determination of the xanthate group distribution on viscoses based on liquid-state NMR spectroscopy was developed. Sample preparation involves stabilization of the xanthate group by allylation followed by derivatization of the remaining free hydroxyl groups at the glucose unit. The method was applied for studying (1) the γ-value (number of xanthate groups per 100 glucose units) of viscose, (2) the distribution of the xanthate groups on the anhydroglucose unit (AGU), and (3) changes of the xanthate group distribution during ripening. Results of the γ-value determination are well comparable with reference methods. Elucidation of the xanthate group distribution on the AGU gives the percentage at the C-6 position and a cumulative share of the positions C-2 and C-3. During ripening, xanthate groups at C-2 and C-3 degrade first, while xanthates at C-6 decompose at a slower rate.  相似文献

The thermo-responsive behaviour of poly-(N-isopropylacrylamide) (PNiPAM) microgels embedded in covalently cross-linked non-temperature-sensitive polyacrylamide (PAam) hydrogel matrixes with different compositions was investigated by using small angle neutron scattering (SANS). The composition of the composite hydrogel was varied by (a) increasing the cross-linker and acrylamide concentration leading to strong hydrogel matrixes and (b) by increasing the microgel concentration to obtain composite gels with an internal structure. Additionally we synthesized composite hydrogels by using γ-irradiation as initiation for the polymerisation. This leads to the formation of chemical bonds between the PNiPAM microgels and the surrounding polyacrylamide matrix. Thus it is possible to synthesize hydrogels without an additional cross-linker, as well as pure particle networks. Some samples were prepared at two different temperatures, below and above the volume phase transition temperature of PNiPAM, resulting in highly swollen or totally collapsed microgels during the incorporation step. The volume phase transition of microgels is not influenced by a hydrogel matrix with high acrylamide concentration independent of the preparation temperature. However, an increased cross-linker concentration leads to a corset like constraint on microgel swelling. Microgels, which are embedded in the collapsed state (at 50 °C), are not able to swell upon cooling, whereas microgels embedded in the swollen state can collapse upon heating. For samples with an increased microgel concentration, the close microgel packing was disturbed by the formation of the polyacrylamide matrix. The hydrogel matrix squeezes the microgels together and leads to partial aggregation. The experiments demonstrate how composite hydrogels with stimuli-sensitive heterogeneities can be prepared such that the full responsiveness of the embedded microgels is retained while the macroscopic dimensions of the gel are not affected by the volume phase transition of the microgels.  相似文献

The title compound, cis‐dichloridobis(triethylphosphane)platinum(II) chloroform monosolvate, [PtCl2(C6H15P)2]·CHCl3, has been obtained from ligand scrambling in the cis‐[PtCl2(Cyp2PCl)(PEt3)] (Cyp = cyclopentyl) system in CHCl3 solvent. Unlike the two previously reported unsolvated polymorphs, which are both monoclinic, the compound crystallizes in an orthorhombic setting. Furthermore, the system exhibits a reversible temperature‐dependent structural phase transition, coupling a reduction in anisotropic displacement parameters and a reduction in crystallographic symmetry on cooling. The high‐temperature phase adopts space group Pnma with the complex and solvent molecules sitting across a crystallographic mirror plane (Z′ = 0.5). The low‐temperature phase adopts the space group P212121 with Z′ = 1.  相似文献
