Oriented immobilization of Desulfovibrio gigas hydrogenase onto carbon electrodes by covalent bonds for nonmediated oxidation of H2

The orientation of hydrogenase bound covalently to a pyrolytic graphite edge electrode modified with a 4-aminophenyl monolayer can be modulated via electrostatic interactions during the immobilization step. At low ionic strength and when the amino groups of the electrode surface are mostly protonated, the hydrogenase is immobilized with the negatively charged region that surrounds its 4Fe4S cluster nearer to the protein surface facing the electrode. This allows direct electron transfer between the immobilized hydrogenase and the electrode, which is observed by the strong catalytic currents measured in the presence of the H2 substrate. Therefore, a very stable enzymatic electrode is produced that catalyzes nonmediated H2 oxidation.     

Vascular endothelial and smooth muscle cell culture on NaOH-treated poly(epsilon-caprolactone) films: a preliminary study for vascular graft development

Tissue engineering offers the potential of providing vessels that can be used to replace diseased and damaged native blood vessels. The endothelization of a synthetic vascular graft minimizes the failures associated with blood clotting and platelet activation. The aim of this study was to culture vascular-derived endothelial and smooth muscle cells on both untreated and NaOH-treated poly(epsilon-caprolactone) (PCL) films, a biocompatible and bio-resorbable polymer, and to evaluate the behavior of both cell types as a preliminary study for vascular graft development. PCL films were prepared by hot pressing; characterized by DSC, IR, SEM, and scanning force microscopy; and treated with NaOH to increase the surface hydrophilicity before cell culture. Endothelial and smooth muscle cells, isolated from pig cava vein, were characterized by immunofluorescence and confocal microscopy studies of endothelial nitric oxide synthase and alpha-smooth muscle actin. Good adhesion, growth, viability and morphology of both the endothelial and smooth muscle cells on PCL films were obtained, but a light stimulation of mitochondrial activity was observed during short culture times. NaOH treatment improved the adhesion and enhanced the proliferation in both cell types. This verified the possible use of this modified polymer as a support in the preparation of a synthetic vascular graft. Indirect spectrophotometric determination of cyanide

A spectrophotometric method for cyanide based on its inhibition of the colour formation reaction between nickel(II) and 3-(4,5-dimethyl-2-thiazolylazo)-2,6-dihydroxybenzoic acid has been developed, and allows the determination of down to 0.1 g of cyanide. Most of the interferences can be avoided by displacement of the hydrogen cyanide using an arsine generator.     

Unusual volumetric and hydration behavior of the catanionic system sodium undecenoate: sodecyltrimethylammonium bromide

Following the studies on the effect of double bonds in the surfactant hydrophobic tail on the formation of mixed surfactant aggregates, we studied the viscosity and density of the system Sodium 10-undecenoate (SUD)–decyltrimethylammonium bromide (DTAB)–water. We found that the partial molar volume (pmv) and intrinsic viscosity of both, micellised and unmicellised mixtures, are non-ideal, dependent on the mixture composition and related to structural changes in micelles. These phenomena are caused by the presence of the double bond at the distal extreme of the SUD molecule, which has some affinity with water by formation of hydrogen bonds. In particular, as far as we know, this is the first report on non-ideal behavior of the pmv in mixed micelles.     

Metabonomics: the use of electrospray mass spectrometry coupled to reversed-phase liquid chromatography shows potential for the screening of rat urine in drug development

The application of liquid chromatography/mass spectrometry (LC/MS) followed by principal components analysis (PCA) has been successfully applied to the screening of rat urine following the administration of three candidate pharmaceuticals. With this methodology it was possible to differentiate the control samples from the dosed samples and to identify the components of the mass spectrum responsible for the separation. These data clearly show that LC/MS is a viable alternative, or complementary, technique to proton NMR for metabonomics applications in drug discovery and development.     

Coalescence in three-phase fluid systems

Summary The gravity coalescence of drops at flat fluid/liquid interfaces was studied in three-phase systems. Rest-time distributions were in qualitative agreement with the earlier theory of Charles and Mason for two-phase systems. The probability of coalescence increased inversely with the square of intervening film thickness. Partial coalescence was similar to that previously observed in two-phase systems. The viscosity of the drop affected the mechanism of coalescence more than the viscosities of the other phases.

Zusammenfassung Es wurde die Koaleszenz von Tropfen an ebenen Flüssigkeits-Flüssigkeitsgrenzfl?chen in Dreiphasensystemen untersucht. Die beobachtete Verteilung der Verweilzeiten ist in qualitativer übereinstimmung mit der Theorie von Charles und Mason für Zweiphasensysteme. Die Wahrscheinlichkeit dafür, da? Koaleszenz auftritt, nimmt umgekehrt proportional mit dem Quadrat der Filmdicke zwischen Tropfen und Phasengrenze zu. Die partielle Koaleszenz war ?hnlich, wie sie früher in Zweiphasensystemen beobachtet worden war. Die Viskosit?t des Tropfens beeinflu?t den Mechanismus der Koaleszenz mehr als die Viskosit?t der anderen Phasen.

1H and 13C assignments of two new triterpenes from Cladocolea grahami

The structure elucidation and 1H and 13C assignments of the new triterpenes 3beta-palmitoxy-7beta-hydroxyolean-12-ene (1) and its hydrolysis product 3beta,7beta-dihydroxyolean-12-ene (2) and 3beta-sn-glyceroyl-(1'-palmitoxy)urs-12-ene (3), isolated from the aerial parts of Cladocolea grahami (Loranthaceae), are reported.     

Time-dependent pump-probe spectra of NeBr2

Time- and frequency-resolved pump-probe measurements on NeBr2 have been performed to better characterize its fragmentation dynamics on the B electronic state for vibrational levels in the energy region of the transition from direct vibrational predissociation to intramolecular vibrational relaxation dynamics. Above nu'=20 of the Br2 stretching mode, it was observed that the dependence of lifetime on the vibrational quantum number deviates from the energy-gap law by leveling off in the range of 10 psE transitions of the complex. These transitions are shifted 20 cm(-1) to lower energy from the free Br2 resonances, indicating an E state Ne-Br2 bond energy of 82 cm(-1). Measurements of NeBr2 vibrational predissociation via the delta nu=-2 channel were also performed for nu'=27, 28, and 29. The closing of the delta nu=-1 channel leads to an increase in the lifetimes of these vibrational levels. A new Nd:yttrium aluminum garnet pumped dual optical parametric oscillator/optical parametric amplifier system is described that allows us to conveniently record time-delayed pump-probe spectra with 2-cm(-1) spectral resolution and 15-ps time resolution.     

Complex emulsion inversion pattern associated with the partitioning of nonionic surfactant mixtures in the presence of alcohol cosurfactant

Commercial ethoxylated nonionic surfactant mixtures containing alcohol cosurfactant exhibit a three-phase behavior whose formulation strongly varies with the water/oil ratio. As a consequence, a change in water/oil ratio can result in a sequence of up to three different emulsion inversion processes, through a combination of formulation and composition effects.
