The rate of specific hydrogen ion catalyzed hydrolysis of 2-(p-heptoxyphenyl)-1,3-dioxolane and equilibrium protonation of 1-hydroxy-2-naphthoate ion in zwitterionic micelles of SB3-14, C14H29NMe2+(CH2)3SO3-, are increased markedly by NaClO4 which induces anionic character and uptake of H3O+ in the micelles. Other salts, for example, NaNO3, NaBr, and NaCl, have similar but much smaller effects on this uptake of H3O+.  相似文献

[reaction: see text] p-Nitromandelic acid as a safety-catch linker for Boc/Bzl-SPPS of base-labile compounds like peptides and depsipeptides is described. This linker permits acidic removal of side-chain protection groups from the resin. For cleavage from the solid support, the p-nitro group was reduced with tin(II) chloride. After washing off the reducing agents, the (depsi)peptide acids with or without the side-chain protection schemes were obtained by microwave irradiation at 50 degrees C with 5% TFA in dioxane.  相似文献

Today a high degree of "false" appendicitis diagnoses are occurring. In this study, a screening experiment of biomarkers of two different kinds of appendicitis, gangrenous and phlegmonous, were conducted with CE and CEC coupled to MS. Plasma samples were obtained from patients pre- and post-surgery. A large amount of data was generated to be able to compare them, and chemometrics tools were utilized to visualize the differences. Indicative patterns were found for both pre- and post-surgery of the two types of inflammation as well as between them. The divergences were traced back to the MS peaks obtained in the CE- and CEC-MS setups as possible biomarkers for the two forms of appendicitis.  相似文献

Cyclodextrin glucanotransferase production from Bacillus clausii E16, a new bacteria isolated from Brazilian soil samples was optimized in shake-flask cultures. A 2(4) full-factorial central composite design was performed to optimize the culture conditions, using a response surface methodology. The combined effect among the soluble starch concentration, the peptone concentration, the yeast extract concentration, and the initial pH value of the culture medium was investigated. The optimum concentrations of the components, determined by a 2(4) full-factorial central composite design, were 13.4 g/L soluble starch, 4.9 g/L peptone, 5.9 g/L yeast extract, and initial pH 10.1. Under these optimized conditions, the maximum cyclodextrin glucanotransferase activity was 5.9 U/mL after a 48-h fermentation. This yield was 68% higher than that obtained when the microorganism was cultivated in basal culture medium.  相似文献

Spherical micellar aggregates have been obtained in chloroform by mixing poly(styrene)-block-poly(4-vinylpyridine) (PS-b-P4VP) diblock copolymers with perfluorinated surfactants (FS) bearing a carboxylic acid head. These micellar aggregates are resulting from the self-assembly of the insoluble P4VP/fluorinated complexes into a core surrounded by the soluble PS coronal chains. Their characteristic features have been studied as a function of various parameters including the composition of the PS-b-P4VP copolymer, the tail length of the fluorinated surfactant, the 4VP/FS molar ratio, the number of carboxylic acid group (1 or 2) on the surfactant, the presence of the PS block and of the fluorine atoms on the surfactant. Dilution of these initial micellar aggregates triggers a morphological reorganization resulting in the formation of more stable vesicles. The extent of this morphological transition is related to the solubility of the P4VP/fluorinated complexes during the dilution process. This transition is complete for short P4VP/FS complexes, incomplete for long P4VP/FS complexes, and not observed whenever an alpha,omega-difunctional FS is used in P4VP/FS complexes, leading to a cross-linked core. Finally, the spheres-to-vesicles transition has been advantageously used in order to encapsulate molecules, as demonstrated by confocal fluorescence microscopy.  相似文献

Matrix-assisted laser desorption ionization (MALDI), Peptide Mass Fingerprinting (PMF) and MALDI-MS/MS ion search (using MASCOT) have become the preferred methods for high-throughput identification of proteins. Unfortunately, PMF can be ambiguous, mainly when the genome of the organism under investigation is unknown and the quality of spectra generated is poor and does not allow confident identification. The post-source decay (PSD) fragmentation of singly charged tryptic peptide ions generated by MALDI-TOF/TOF typically results in low fragmentation efficiency and/or complex spectra, including backbone fragmentation ions (series b and y), internal fragmentation etc. Interpreting these data either manually and/or using de novo sequencing software can frequently be a challenge. To overcome this limitation when studying the proteome of adult Angiostrongylus costaricensis, a nematode with unknown genome, we have used chemical N-terminal derivatization of the tryptic peptides with 4-sulfophenyl isothiocyanate (SPITC) prior to MALDI-TOF/TOF MS. This methodology has recently been reported to enhance the quality of MALDI-TOF/TOF-PSD data, allowing the obtainment of complete sequence of most of the peptides and thus facilitating de novo peptide sequencing. Our approach, consisting of SPITC derivatization along with manual spectra interpretation and Blast analysis, was able to positively identify 76% of analyzed samples, whereas MASCOT analysis of derivatized samples, MASCOT analysis of nonderivatized samples and PMF of nonderivatized samples yielded only 35, 41 and 12% positive identifications, respectively. Moreover, de novo sequencing of SPITC modified peptides resulted in protein sequences not available in NCBInr database paving the way to the discovery of new protein molecules.  相似文献

The fractionation of a statistical copolyester made of poly(ethylene terephthalate- isophthalate) has been performed using Temperature Rising Elution Fractionation (TREF). TREF fractions were analyzed by size exclusion chromatography, thermal analysis, 1H-nuclear magnetic resonance and small angle X-Ray diffractometry. Results show that the fractions exhibit both interchain and intrachain defect distribution. As the intrachain defect distribution can not be determined by experimental techniques, a computer simulation based on the determination of terephthalic sequences length in a virtual polymeric distribution was performed. The simulated chains were constructed using a stochastic process. The analysis of the results obtained on the distribution of the simulated sequences in the polymer correlates quantitatively well between the new fractionation mechanism proposed for copolyesters and the experimental results.  相似文献
