Cellulose-based carriers Granocel were specially prepared and optimised for covalent immobilization of enzymes. The effects of carrier characteristics such as pore size, chemistry of anchor groups and their density on invertase immobilization efficiency were evaluated. It was found that the preferential adsorption/binding of the enzyme to a carrier during coupling and its activity after immobilization depended on microenvironmental effects created by hydrophilic surface of the carrier, functional groups and their activators. The best preparations (activity approx. 300 U/mL, high storage stability) were obtained for NH2-Granocel activated with glutaraldehyde. It is probably due to Granocel modification with pentaethylenehexamine that gave a 19-atom spacer arm. The enzyme concentration in coupling mixture was optimised as well. The kinetic parameters of sucrose hydrolysis for native and immobilized invertase were evaluated. Compared to the native invertase, K m value of immobilized enzyme was only twice higher with about three times lower substrate inhibition. Reaction runs in a well mixed batch reactors with native and immobilized invertase showed slightly slower reaction rate in the case of the enzyme covalently bound to Granocel. Very good stability of cellulose-based carrier was proved experimentally by 20 successive reaction runs in a batch reactor.  相似文献

The synthesis and evaluation as 5‐HT1A and 5‐HT7 serotonin receptor ligands of the two sets of O‐substituted hydroxybenzamides, structurally related to 2‐{3‐[4‐(2‐methoxyphenyl)piperazin‐1‐yl]propoxy}benzamide ( 1 ), (Ki 5‐HT1A = 21 nM, 5‐HT7 = 234 nM) are reported. To affect the affinity for 5‐HT1A and 5‐HT7 receptors, an amide moiety ( 2 , 3 , 4 , 5 , 6 ) and a hydrocarbon chain length ( 7 , 8 , 9 , 10 ) were modified. The serotonergic activity of compounds 2 , 3 , 4 , 5 , 6 , 7 , 8 , 9 , 10 was generally higher in the case of 5‐HT1A receptors compared with 5‐HT7 ones; the most active 5‐HT1A ligands being meta‐isomer 2 (Ki = 7 nM) and both analogs of 1 with the longest spacer, i.e., penta‐ and hexa‐methylene derivatives 9 and 10 (Ki = 4 and 3 nM, respectively). The observed biological properties of compounds 1 , 2 , 3 , 4 , 5 , 6 , 7 , 8 , 9 , 10 were elucidated using molecular modeling procedures. J. Heterocyclic Chem., (2010).  相似文献

Titania layers obtained by a sol-gel technique doped with redox mediator, Meldola’s Blue, were employed for construction of a new NADH senor. Optimization of preparation process as well as experimental conditions affecting the response of the sensor were examined. Under optimal conditions NADH could be determined in the wide linear range from 90 to 2300 μM with detection limit 12 μM and a high sensitivity 12.5 nA μM−1. The usefulness of developed sensor was preliminarily checked in determination of NADH forming during enzymatic oxidation of ethanol catalyzed by alcohol dehydrogenase (ADH).   相似文献

We present new tags based on the derivatives of phenylboronic acid and apply them for the selective detection of sugars and peptide-sugar conjugates in mass spectrometry. We investigated the binding of phenylboronic acid and its quaternary ammonium salt (QAS) derivatives to carbohydrates and peptide-derived Amadori products by HR-MS and MS/MS experiments. The formation of complexes between sugar or sugar-peptide conjugates and synthetic tags was confirmed on the basis of the unique isotopic distribution resulting from the presence of boron atom. Moreover, incorporation of a quaternary ammonium salt dramatically improved the efficiency of ionization in mass spectrometry. It was found that the formation of a complex with phenylboronic acid stabilizes the sugar moiety in glycated peptides, resulting in simplification of the fragmentation pattern of peptide-derived Amadori products. The obtained results suggest that derivatization of phenylboronic acid as QAS is a promising method for sensitive ESI-MS detection of carbohydrates and their conjugates formed by non-enzymatic glycation or glycosylation.

Abstract  Modular conditions for the formation of thioureas or pyrimidine-4-one-2-thiones connected to the benzo[b]thiophene, benzene and indole structures were performed. A benzo[b]thiophene isothiocyanate derivative was used as a model to study the condensation with simple aromatic amines and amino-l-sorbose derivative. The construction of pyrimidine-4-one-2-thiones using basic conditions afforded efficiently new heterocyclic aromatics, which were further transformed using the alkylated sulfur as a leaving group in palladium-catalyzed cross-coupling reactions. The synthesis of six analogues of the potent thymidylate synthase (TS) inhibitor N -[4-[ N -[(3,4-dihydro-2-methyl-4-oxo-6-quinazolinoyl)-methyl]- N -prop-2-ynylamino]benzoyl]- L -glutamic acid 2 is described in which the glutamic acid residue has been replaced by DL -aminophosphonic acids. New antifolates were tested as inhibitors of TS isolated from mouse L1210 leukemic cells as well as inhibitors of growth mouse leukemic L5178Y cells. In general these modifications result in compounds that are considerably less potent than 2 as TS inhibitors with K i 's 0.17-1.10 w M. Very poor solubility in water limited their proper assay of growth cells inhibition.  相似文献

The reactivity of tosylated glycerol carbonate 2 toward nucleophiles has been exploited to generate glycidol analogues protected with carbonate or carbamate groups. The activated glycerol 2 is a reasonable linking agent with thiol and alcohol nucleophiles and an excellent and selective one with primary amines, allowing efficient bis-functionalizations of glycerol.  相似文献

Four compounds showing moderate antituberculostatic activity have been studied to test the hypothesis that the planarity of the 2‐[amino(pyrazin‐2‐yl)methylidene]dithiocarbazate fragment is crucial for activity. N′‐Anilinopyrazine‐2‐carboximidamide, C11H11N5, D1, and diethyl 2,2′‐[({[amino(pyrazin‐2‐yl)methylidene]hydrazinylidene}methylidene)bis(sulfanediyl)]diacetate, C14H19N5O4S2, B1, maintain planarity due to conjugation and attractive intramolecular hydrogen‐bond contacts, while methyl 3‐[amino(pyrazin‐2‐yl)methylidene]‐2‐methyldithiocarbazate, C8H11N5S2, C1, and benzyl 3‐[amino(pyrazin‐2‐yl)methylidene]‐2‐methyldithiocarbazate, C14H15N5S2, C2, are not planar, due to methylation at one of the N atoms of the central N—N bond. The resulting twists of the two molecular halves (parts) of C1 and C2 are indicated by torsion angles of 116.5 (2) and −135.9 (2)°, respectively, compared with values of about 180° in the crystal structures of nonsubstituted compounds. As the methylated derivatives show similar activity against Mycobacterium tuberculosis to that of the nonsubstituted derivatives, maintaining planarity does not seem to be a prerequisite for activity.  相似文献

Propyl gallate (PG) adsolubilisation in the cationic, anionic and nonionic surfactant micelles formed in the bulk solution and at the silica/solution interface has been investigated. It was found that in the absence of surfactant, propyl gallate does not adsorb on the silica surface from aqueous solution. However, in the presence of hexyltrimethylammonium bromide (CTAB), its uptake by silica significantly increases. Alumina is quite an effective adsorbent for SDS and propyl gallate and does not adsorb nonionic TX-100. The addition of PG promotes adsorption of SDS and TX-100.  相似文献
