Phytoestrogens occur naturally in plants and are structurally similar to mammalian estrogens. The lignans are a class of phytoestrogen and can be metabolized to the biologically active enterolignans, enterodiol, and enterolactone by a consortium of intestinal bacteria. Secoisolariciresinol diglucoside (SDG), a plant lignan, is metabolized to enterodiol and, subsequently, enterolactone. Matairesinol, another plant lignan, is metabolized to enterolactone. Other dietary enterolignan precursors include lariciresinol, pinoresinol, syringaresinol, arctigenin, and sesamin. Enterolignan exposure is determined in part by intake of these precursors, gut bacterial activity, and host conjugating enzyme activity. A single SDG dose results in enterolignan appearance in plasma 8-10 h later--a timeframe associated with colonic bacterial metabolism and absorption. Conjugation of enterolignans with sulfate and glucuronic acid occurs in the intestinal wall and liver, with the predominant conjugates being glucuronides. Controlled feeding studies have demonstrated dose-dependent urinary lignan excretion in response to flaxseed consumption (a source of SDG); however, even in the context of controlled studies, there is substantial interindividual variation in plasma concentrations and urinary excretion of enterolignans. The complex interaction between colonic environment and external and internal factors that modulate it likely contribute to this variation. Knowledge of this field, to date, indicates that understanding the sources of variation and measuring the relevant panel of compounds are important in order to use these measures effectively in evaluating the impact of lignans on human health. 相似文献

Strong electrogenerated chemiluminescence (ECL) of fluorescein is generated during cathodic pulse polarization of oxide-covered
aluminum electrodes and the resulting decay of emission is so sluggish that time-resolved detection of fluorescein is feasible.
The present ECL in aqueous solution is based on the tunnel emission of hot electrons into the aqueous electrolyte solution,
which probably results in the generation of hydrated electrons and hydroxyl radicals acting as redox mediators. The successive
one-electron redox steps with the primary radicals result in fluorescein in its lowest excited singlet state. The method allows
the detection of fluorescein (or its derivatives containing usable linking groups to biomolecules) over several orders of
magnitude of concentration with detection limits well below nanomolar concentration level. The detection limits can still
be lowered, e.g., by addition of azide or bromide ions as coreactants. The results suggest that the derivatives of fluorescein,
such as fluorescein isothiocyanate (FITC), can be detected by time-resolved measurements and thus be efficiently used as electrochemiluminescent

Constitutive androstane receptor (CAR), along with pregnane x receptor (PXR), is an important metabolic sensor in the hepatocytes. Like all other nuclear receptors (NRs), CAR works in concert with coregulator proteins, coactivators, and corepressors which bind to the NRs. The main basis for the receptor to distinguish between coactivators and corepressors is the position of the C-terminal helix 12 (H12), which is determined by the bound NR ligand. CAR, having constitutive activity, can be repressed or further activated by its ligands. Crystal structure of human CAR bound to an agonist and a coactivator peptide is available, but no structural information on an inverse agonist-bound human CAR and a corepressor exists. In our previous molecular dynamics (MD) studies, no corepressor peptide was included. Therefore, probably due to the strong interactions which keep the relatively short H12 of CAR in the active position, the structural changes elicited by inverse agonists were very subtle, and H12 of CAR seemed to more or less retain its active conformation. Here, we have run a series of MD simulations to study the movement of H12 in the presence of both activating and repressing ligands as well as a corepressor peptide. The presence of the corepressor on the coregulator surface of CAR induced a clear shift of H12 of the inverse agonists-bound CAR. In general, H12 moved toward H10 and not away from the ligand binding domain, as seen in some other NRs. Cell adhesion processes take place through mechanotransduction mechanisms where stretching of proteins results in biological responses. In this work, we present the first cyto-mechanoresponsive surface that mimics such behavior by becoming cell-adhesive through exhibition of arginine-glycine-aspartic acid (RGD) adhesion peptides under stretching. This mechanoresponsive surface is based on polyelectrolyte multilayer films built on a silicone sheet and where RGD-grafted polyelectrolytes are embedded under antifouling phosphorylcholine-grafted polyelectrolytes. An elementary supramolecular conducting system was constructed using a novel (±)‐thioctic acid‐functionalized β‐cyclodextrin host deposited on a gold (Au) surface and an iridium‐bearing guest molecule with biphenyl tails to insert specifically into the cyclodextrin cavity. The resulting supramolecular system was used to investigate remote electron communication between the flat Au surface and the platinum (Pt)/iridium (Ir) tip of a scanning tunnelling microscope. The morphology of the surfaces after successive deposition of host molecules followed by guest molecules was investigated. Formation of features of 2 nm size was shown on the Au surface functionalised with the supramolecular system. I–V spectroscopic analysis of the tunnelling current through this supramolecular layer revealed the relation between the effective barrier height and tunnelling distance. Thus, in the supramolecular host–metallo‐guest system, a small increase of conductance is observed, compared to the layer without the guest. This can be attributed to the presence of the Ir‐guest, which eventually creates intermediate energy states between the Au substrate and the Pt/Ir tip. Copyright © 2011 John Wiley & Sons, Ltd. 相似文献

Ultrastrong coupling is studied in a modulation-doped parabolic potential well coupled to an inductance-capacitance resonant circuit. In this system, in accordance to Kohn's theorem, strong reduction of the energy level separation caused by the electron-electron interaction compensates the depolarization shift. As a result, a very large ratio of 27% of the Rabi frequency to the center resonance frequency as well as a polariton gap of width 2π × 670 GHz are observed, suggesting parabolic quantum wells as the system of choice in order to explore the ultrastrong coupling regime. 相似文献

Although predictors of violent relationships have been identified, we are only beginning to understand the day-to-day dynamics of domestic violence. The objective of this study was to identify commonly seen patterns and strings of consecutive days involving verbal or physical abuse, and their preceding and subsequent events. Adult women (n=20) seen in a primary care clinic who experienced violence within the past month were enrolled. Subjects completed a daily telephone assessment of household environment and marital relationship for two months using Interactive Verbal Response (IVR). Results were analyzed using orbital decomposition, an analytic technique based on symbolic dynamics, in which categorical time series data are used to identify recurrent patterns of strings and quantify their complexity. While days without abuse had varied patterns involving arguments, stress levels, daily hassles, husband's alcohol intake, and sense of closeness (27 unique patterns), days involving verbal or physical abuse included a narrower range of patterns (15 patterns for verbal and 16 patterns for physical abuse). Daily patterns appear to cluster in triplets (3 consecutive days) of activity and show nonlinearity with triplets involving verbal abuse and triplets involving physical violence. Triplets involving either verbal or physical abuse were associated with arguments and high stress, but differed in the consistency of association with hassles, alcohol intake, and closeness. Finally, physical and verbal abuse tended to self-propagate. However, days involving verbal abuse did not precede days involving physical violence. In conclusions, while patterns of violence and household environments followed a nonlinear trajectory, only a limited set of patterns were observed. Although violence led to more violence, verbal abuse did not necessarily lead to physical aggression.
