Cylindrospermopsin (CYN) belongs to a group of toxins produced by several strains of freshwater cyanobacteria. It is a compact zwitterionic molecule composed of a uracil section and a tricyclic guanidinium portion with a primarily hepatotoxic effect. Using low multi-stage and high-resolution mass spectrometry, the gas-phase reactions of this toxin have been investigated. Our data show that collision-induced dissociation (CID) spectra of CYN are dominated by neutral losses, and three major initial fragmentation pathways are clearly distinguishable. Interestingly, comparative analysis of protonated and cationizated molecules showed a significant difference in the balance of the SO3 and terminal ring elimination. These data indicate that the differential ion mobility of H+, Li+, Na+ and K+ leads to different fragmentation pathways, giving rise to mass spectra with different profiles.  相似文献

We describe in full the first synthesis of the potent insect antifeedant azadirachtin through a highly convergent approach. An O-alkylation reaction is used to unite decalin ketone and propargylic mesylate fragments, after which a Claisen rearrangement constructs the central C8-C14 bond in a stereoselective fashion. The allene which results from this sequence then enables a second critical carbon-carbon bond forming event whereby the [3.2.1] bicyclic system, present in the natural product, is generated via a 5-exo-radical cyclisation process. Finally, using knowledge gained through our early studies into the reactivity of the natural product, a series of carefully designed steps completes the synthesis of this challenging molecule.  相似文献

UDP-galactopyranose mutase (UGM) is the key enzyme involved in the biosynthesis of Galf. UDP-Galp and UDP-Galf are two natural substrates of UGM. A protocol that combines the use of STD-NMR spectroscopy, molecular modeling, and CORCEMA-ST calculations was applied to the investigation of the binding of UDP-Galf and its C3-fluorinated analogue to UGM from Klebsiella pneumoniae. UDP-Galf and UDP-[3-F]Galf were bound to UGM in a manner similar to that of UDP-Galp. The interconversions of UDP-Galf and UDP-[3-F]Galf to their galactopyranose counterparts were catalyzed by the reduced (active) UGM with different catalytic efficiencies, as observed by NMR spectroscopy. The binding affinities of UDP-Galf and UDP-[3-F]Galf were also compared with those of UDP-Galp and UDP by competition STD-NMR experiments. When UGM was in the oxidized (inactive) state, the binding affinities of UDP-Galf, UDP-Galp, and UDP-[3-F]Galf were of similar magnitudes and were lower than that of UDP. However, when UGM was in the reduced state, UDP-Galp had higher binding affinity compared with UDP. Molecular dynamics (MD) simulations indicated that the "open" mobile loop in UGM "closes" upon binding of the substrates. Combined MD simulations and STD-NMR experiments were used to create models of UGM with UDP-Galf and UDP-[3-F]Galf as bound ligands. Calculated values of saturation-transfer effects with CORCEMA-ST (complete relaxation and conformational exchange matrix analysis of saturation transfer) were compared to the experimental STD effects and permitted differentiation between two main conformational families of the bound ligands. Taken together, these results are used to rationalize the different rates of catalytic turnover of UDP-Galf and UDP-[3-F]Galf and shed light on the mechanism of action of UGM.  相似文献

Polyanilines doped with different acids (HCl, H2SO4 and dodecylbenzenesulfonic acid, DBSA) were prepared and their surface electrical conductivities were characterized in a four-probe device, connected to a real-time data acquisition board. Collected data were synchronized and conductivity calculations were performed. The conductive behavior of the polyanilines was investigated along the electrification time. This allowed for introduction of a non-Gaussian technique for determination of the confidence intervals of surface resistivity data. It is shown that the distribution of experimental surface resistivity data does not follow a normal probability distribution function (PDF). Thus, ordinary assumptions related to normal distribution of the experimental errors are wrong and must be corrected for proper determination of the confidence limits of measured resistivity values. It is shown here that confidence limits of resistivity values are asymmetrical and that distribution of experimental values can follow multimodal distributions.  相似文献

The spinel-type ferrites NiFe2O4 and Zn0.5Ni0.5Fe2O4 modified by lanthanide ions Eu3+ and Tb3+ were prepared by a sol—gel process with propylene oxide as a gelating agent. The phase homogeneity of the samples was tested by XRD and Mössbauer spectroscopy. Transmission electronic microscopy used for characterisation of the morphology of the samples revealed nanosized powdered samples with a narrow distribution of particle sizes. It was noted that the presence of Ln3+ ions influenced the magnetic properties of nanosized NiFe2O4 and Ni0.5Zn0.5Fe2O4 ferrites. The dependence of the magnetic properties of the samples on the rare-earth doping may be explained by the different grain sizes. The saturation magnetisation tends to decrease with increasing rare-earth doping and decreasing crystallite size. A similar trend was observed for the coercive field, with the exception of the Tb3+-doped Zn0.5Ni0.5Fe2O4 where it remained the same as in the pure ferrite.  相似文献

The overall adsorption rate of Reactive Black 5 dye (RB5) on chitosan based materials was elucidated using diffusional models. Fundamental aspects, such as, geometry of the adsorbents and swelling effects were considered. Chitosan based materials (powder and film) were prepared from shrimp wastes and characterized regarding to the fundamental features for adsorption. Experimental decay curves were obtained under different conditions of stirring rate and initial dye concentration. The data were modeled according to the external mass transfer and diffusional models. The kL (external mass transfer coefficient), Dep (effective pore diffusion coefficient) and Ds (surface diffusion coefficient) values were estimated. For both adsorbents, it was found that the surface diffusion was the intraparticle diffusion mechanism governing the adsorption rate of RB5, since its contribution was higher than 92 % regardless the position and time. The Ds values ranged from 2.85 × 10?11 to 5.78 × 10?11 for chitosan powder and from 4.15 × 10?11 to 12.12 × 10?11 cm2 s?1 for chitosan films. The RB5 adsorption was faster when chitosan powder was used, mainly at higher stirring rates and initial dye concentrations. The swelling effect was most pronounced for the chitosan films, where, provided an increase of about 65 times in the Ds value.  相似文献

The aim of this paper is to present a general algebraic formulation for the decoherence-free subspaces (DFSs). In order to build the DFSs we consider the tensor product of Clifford algebras and left minimal ideals. States, error operators and projection operators are defined in a purely algebraic point of view. For this purpose, we initially generalize some results of Pauli and Artin about semisimple algebras. Then we derive orthogonality theorems for associative algebras analogous to theorems for finite groups. Some advantages and perspectives are also discussed.  相似文献

Odorous organic compounds from liquid and gas samples of animal wastes were studied by headspace (HS)-solid-phase microextraction (SPME)-GC-MS. 1-Pirenyldiazomethane (PDAM) was adsorbed/absorbed on the SPME fibre in order to obtain the corresponding ester derivatives during the preconcentration step. The SPME fibre was immersed into a PDAM solution. Then, the SPME fibre was withdrawn and exposed to the HS of the liquid cow slurry. This way derivatisation of VFAs took place in the SPME fibre together with the preconcentration of the rest of the analytes of interest. The analytes were desorbed in the hot injection port (300 degrees C) of a GC-MS for 3 min. Four different fibre types and different immersion periods of the fibre in the PDAM solution were studied in order to obtain the best sensitivity with the selected fibre. Accuracy, precision and the LODs were calculated using spiked liquid and gas samples. The possibility of storing liquid samples after sampling by preconcentration on the fibre was also considered. Storage time and temperature were studied. The optimised method was applied to the determination of the analytes in liquid and gas samples from cow slurries from an intensive production farm.  相似文献

Organobentonites were synthesized by treating Na-bentonites (Wyomingand Re?adiye) with cetylpyridinium chloride (CPC) using ion exchangemethod. The difference in the basal spacing by 0.65 Å and a higher massloss by 13.32% of organobentonite (Wyoming) in the temperature range 200–550°Ccomparing with that of organobentonite (Re?adiye) were in conformitywith the CEC values of organobentonites. The HOH stretching and bending peaksof bentonites became sharper following the treatment with the organo-cation.The intensity decrease of the Al–OH band for organobentonite (Re?adiye)and in particular, the significantly higher amount of m-cresolretained by the organobentonite of lower CEC than that by the organobentonite(Wyoming) indicate the influence of the CP cation on the adsorption process.  相似文献
