Metal-mediated intracellular reactions are becoming invaluable tools in chemical and cell biology, and hold promise for strongly impacting the field of biomedicine. Most of the reactions reported so far involve either uncaging or redox processes. Demonstrated here for the first time is the viability of performing multicomponent alkyne cycloaromatizations inside live mammalian cells using ruthenium catalysts. Both fully intramolecular and intermolecular cycloadditions of diynes with alkynes are feasible, the latter providing an intracellular synthesis of appealing anthraquinones. The power of the approach is further demonstrated by generating anthraquinone AIEgens (AIE=aggregation induced emission) that otherwise do not go inside cells, and by modifying the intracellular distribution of the products by simply varying the type of ruthenium complex.  相似文献

The ionization effects on 28 conformations of N-glycylglycine are analyzed by means of the hybrid B3LYP and the hybrid meta-MPWB1K density functionals and by single-point calculations at the CCSD(T) level of theory. The most favorable process observed corresponds to the ionization of the only neutral conformation that presents a OH...NH2 intramolecular hydrogen bond, which leads to CO2 elimination after a spontaneous proton transfer from -COOH to NH2. The remaining neutral structures evolve to 20 different conformations of N-glycylglycine radical cation, which lie about 25-40 kcal/mol higher than the decarboxylated [NH3CH2CONHCH2]+*...[CO2] complex. Structural changes induced by ionization depend on the intramolecular hydrogen bonds of the initial conformation, since they determine the nature of the electron hole formed. In most cases, ionization takes place at the terminal -NH2 and -CO of the amide bond, which produces a strengthening of the peptide bond and the formation of new -NH2...OC(amide) and -NH2...OCOH hydrogen bonds. However, if -NH2 and -CO(amide) simultaneously act as proton acceptor in the neutral conformation, ionization is mainly localized at the carboxylic group, which produces a strengthening of the -COOH...OC(amide) bond. Both functionals lead to similar trends and compare well with CCSD(T) results except for a few cases for which B3LYP provides a too delocalized picture of the electron hole and consequently leads to artificial geometry reorganization.  相似文献

Four succinato-bridged complexes of copper(II) have been synthesized. Complex 1, [Cu(2)(mu-OH(2))(2)L(bpy)(2)(NO(3))(2)](n) and 2, [Cu(2)(mu-OH(2))(2)L(phen)(2)(NO(3))(2)](n)(bpy = 2,2[prime or minute]-bipyridine; phen = 1,10-phenanthroline and LH(2)= succinic acid) exhibit 1D coordination polymer structures where both the nitrate ions are directly linked to the copper(ii) producing synthons in a 2D sheet. A novel 2D grid-like network, ([Cu(4)L(2)(bpy)(4)(H(2)O)(2)](ClO(4))(4)(H(2)O))n3, is obtained upon changing the nitrate by perchlorate anion in complex 1, where the channels are occupied by the anions. On changing the nitrate by tetrafluoroborate anion in complex 2, a novel octanuclear complex, [Cu(8)L(4)(phen)(12)](BF(4))(8).8H(2)O 4, is isolated. The coligand bpy and phen in these complexes show face-to-face (in 1,2,3,4) or edge-to-face (in 4 )pi-pi interactions forming the multidimensional supramolecular architectures. Interestingly, the appearance of edge-to-face pi-pi interactions in complex facilitates the formation of discrete octanuclear entities. Variable-temperature (300-2 K) magnetic measurements of complexes have been done. Complexes 1 and 2 show very weak antiferromagnetic (OOC-CH(2)-CH(2)-COO) and ferromagnetic coupling (mu-H(2)O). Complex 3 also shows antiferromagnetic (syn-syn mu-OCO), and ferromagnetic coupling (mu-O of the -COO group). Complex 4 with two types (syn-syn and syn-anti) of binding modes of the carboxylate group shows strong antiferromagnetic interaction.  相似文献

The design, construction and evaluation of a versatile cyclic olefin copolymer (COC)-based continuous flow potentiometric microanalyzer to monitor the presence of ammonium ion in recycling water processes for future manned space missions is presented. The microsystem integrates microfluidics, a gas-diffusion module and a detection system in a single substrate. The gas-diffusion module was integrated by a hydrophobic polyvinylidene fluoride (PVDF) membrane. The potentiometric detection system is based on an all-solid state ammonium selective electrode and a screen-printed Ag/AgCl reference electrode. The analytical features provided by the analytical microsystem after the optimization process were a linear range from 0.15 to 500 mg L−1 and a detection limit of 0.07 ± 0.01 mg L−1. Nevertheless, the operational features can be easily adapted to other applications through the modification of the hydrodynamic variables of the microfluidic platform.  相似文献

Dynamic combinatorial libraries are powerful systems for studying adaptive behaviors and relationships, as models of more complex molecular networks. With this aim, we set up a chemically diverse dynamic library of pseudopeptidic macrocycles containing amino‐acid side chains with differently charged residues (negative, positive, and neutral). The responsive ability of this complex library upon the increase of the ionic strength has been thoroughly studied. The families of the macrocyclic members concentrating charges of the same sign showed a large increase in its proportion as the ionic strength increases, whereas those with residues of opposite charges showed the reverse behavior. This observation suggested an electrostatic shielding effect of the salt within the library of macrocycles. The top‐down deconvolution of the library allowed us to obtain the fundamental thermodynamic information connecting the library members (exchange equilibrium constants), as well as to parameterize the adaptation to the external stimulus. We also visualized the physicochemical driving forces for the process by structural analysis using NMR spectroscopy and molecular modeling. This knowledge permitted the full understanding of the whole dynamic library and also the de novo design of dynamic chemical systems with tailored co‐adaptive relationships, containing competing or cooperating species. This study highlights the utility of dynamic combinatorial libraries in the emerging field of systems chemistry.  相似文献

The energy barrier to magnetisation relaxation in single‐molecule magnets (SMMs) proffers potential technological applications in high‐density information storage and quantum computation. Leading candidates amongst complexes of 3d metals ions are the hexametallic family of complexes of formula [Mn6O2(R‐sao)6(X)2(solvent)y] (saoH2=salicylaldoxime; X=mono‐anion; y=4–6; R=H, Me, Et, and Ph). The recent synthesis of cationic [Mn6][ClO4]2 family members, in which the coordinating X ions were replaced with non‐coordinating anions, opened the gateway to constructing families of novel [Mn6] salts in which the identity and nature of the charge balancing anions could be employed to alter the physical properties of the complex. Herein we demonstrate initial experiments to show that this is indeed possible. By replacing the diamagnetic ClO4? anions with the highly anisotropic ReIV ion in the form of [ReIVCl6]2?, the energy barrier to magnetisation relaxation is increased by up to 30 %.  相似文献

Highly enantioselective epoxidation of α‐substituted styrenes with aqueous H2O2 is described by using a chiral iron complex as the catalyst and N‐protected amino acids (AAs) as coligands. The amino acids synergistically cooperate with the iron center in promoting an efficient activation of H2O2 to catalyze epoxidation of this challenging class of substrates with good yields and stereoselectivities (up to 97 % ee) in short reaction times.  相似文献

Formation of the thiochroman ring system is achieved by a two step synthesis that involves heating 3‐thiophenyl‐1‐propanols or 4‐thiophenyl‐2‐butanols in toluene with catalytic amounts of p‐toluenesulfonic acid. The propanols are made by the addition of sulfur stabilized carbanions to styrene oxide, ethylene oxide, propylene oxide, and isobutylene oxide. The carbanions are generated by treatment of either benzyl phenyl sulfide or thioanisole with butyllithium. The effect of substitution at the 1 and 3 positions of the propanols on the reaction yields is discussed. The mechanism of the reaction apparently involves intramolecular electrophilic aromatic substitution rather than a Claisen or thio‐Claisen rearrangement.  相似文献

A rapid, robust and economical method for the analysis of persistent halogenated organic compounds in small volumes of human serum and umbilical cord blood is described. The pollutants studied cover a broad range of molecules of contemporary epidemiological and legislative concern, including polychlorobiphenyls (PCBs), polychlorobenzenes (CBs), hexachlorocyclohexanes (HCHs), DDTs, polychlorostyrenes (PCSs) and polybromodiphenyl ethers (PBDEs). Extraction and clean-up with n-hexane and concentrated sulphuric acid was followed with analysis by gas chromatography coupled to electron capture (GC-ECD) and GC coupled to negative ion chemical ionisation mass spectrometry (GC-NICI-MS). The advantages of this method rest in the broad range of analytes and its simplicity and robustness, while the use of concentrated sulphuric acid extraction/clean-up destroys viruses that may be present in the samples. Small volumes of reference serum between 50 and 1000 μL were extracted and the limits of detection/quantification and repeatability were determined. Recoveries of spiked compounds for the extraction of small volumes (≥300 μL) of the spiked reference serum were between 90% and 120%. The coefficients of variation of repeatability ranged from 0.1–14%, depending on the compound. Samples of 4-year-old serum and umbilical cord blood (n = 73 and 40, respectively) from a population inhabiting a village near a chloro-alkali plant were screened for the above-mentioned halogenated pollutants using this method and the results are briefly described.  相似文献

A new pyridylpyrazole-containing tetradentate ligand, namely N,N-bis(3,5-dimethylpyrazol-1-ylmethyl)aminomethylpyridine (L), and two of its binuclear azido-bridged complexes, [Ni2(L)2(N3)2](ClO4)2·2EtOH (1) and [Cu2(L)2(N3)2](ClO4)2 (2), have been synthesized and characterized by physico-chemical and spectroscopic methods. The crystal structures of both complexes are reported. Each metal atom in the complexes has a MN6 coordination environment with distorted octahedral geometry. Variable-temperature magnetic susceptibility measurements for complex (1) show typical antiferromagnetic behavior with J value −84.5 ± 1.3 cm−1, whereas complex (2) has no magnetic interactions.  相似文献
