The importance of imaging mass spectrometry (MS) for visualizing the spatial distribution of molecular species in biological tissues and cells is growing. We have developed a new system for imaging MS using MeV ion beams, termed MeV-secondary ion mass spectrometry (MeV-SIMS) here, and demonstrated more than 1000-fold increase in molecular ion yield from a peptide sample (1154 Da), compared to keV ion irradiation. This significant enhancement of the molecular ion yield is attributed to electronic excitation induced in the near-surface region by the impact of high energy ions. In addition, the secondary ion efficiency for biologically important compounds (>1 kDa) increased to more than 10(10) cm(-2), demonstrating that the current technique could, in principle, achieve micrometer lateral resolution. In addition to MeV-SIMS, peptide compounds were also analyzed with cluster-SIMS and the results indicated that in the former method the molecular ion yields increased substantially compared to the latter. To assess the capability of MeV-SIMS to acquire heavy-ion images, we have prepared a micropatterned peptide surface and successfully obtained mass spectrometric imaging of the deprotonated peptides (m/z 1153) without any matrix enhancement. The results obtained in this study indicate that the MeV-SIMS technique can be a powerful tool for high-resolution imaging in the mass range from 100 to over 1000 Da.  相似文献

The Ag electrode surface of a solid electrolyte in its working state has been studied by X-ray photoelectron spectroscopy. The nature of the AgO system on the electrode is dependent on the temperature. There are three types of AgO systems at 400°C. Type 1 with an O is binding energy of 532.6 eV only appeared at 325°C. However, when the temperature was raised to 400°C, types II and III (with BEs 531.1 and 529.2 eV) appeared. These O is signals are discussed in detail.  相似文献

Abstract— The action spectrum and dose-rate dependence for photoreactivation of mutation to prototrophy in late-lag-phase cultures of Escherichia coli H3r30 (which lacks active photo-reactivating enzyme) are roughly similar to those for photoprotection from killing in other strains. It is suggested that photoreactivation of this mutation in H/r30 is an indirect effect, similar in mechanism to photoprotection. The action spectrum and dose-rate dependence for photoreactivation of mutation to prototrophy in late-lag-phase cultures of E. coli H3r30-R (which possesses active photoreactivating enzyme) are roughly similar to those for photoreactivation of killing in most other strains. It is suggested that photoreactivation of this mutation in H/r30-R is a direct effect at long wavelengths, but that there is an indirect component at short wavelengths. A quite different interpretation of these data is noted. Finally, it is found that, under the conditions of these experiments, indirect photoreactivation of killing in H/r30 and H/r30-R is weak or nonexistent.  相似文献

The kinetic behavior on the polymerization of formaldehyde with and without acidic catalyst, in liquid carbon dioxide, in the temperature range of 30 to 50°C. was investigated. In the polymerization without catalyst both the polymer yield and the degree of polymerization increased with reaction time and also with rising temperature. With acidic catalyst, such as acetic acid and dichloroacetic acid, both the polymer yield and the degree of polymerization increased more than that in the polymerization without catalyst. The overall rate of polymerization with and without acidic catalyst was expressed by the first-order rate equation with respect to monomer concentration. From the results it was concluded that the polymerizations belonged to a type of successive polymerization with rapid initiation and no termination. The rate constant and the activation energy of each elementary process of polymerization were estimated on the basis of the results.  相似文献
