The acid-catalysed rearrangement of the cyclohex-2-en-1-ols 15 , d3- 15 , 16 , 17 and 19 , the cyclohexa-2,5-dien-1-ols 20 and 21 , and also the allyl alcohols 22 and 23 (Scheme 3), using 98-percent sulfuric acid/acetic anhydride 1:99 at room temperature, was investigated. From the rearrangement of 4-allyl-4-phenyl-cyclohex-2-en-1-ol ( 15 ), with reaction times greater than 2 hours a single product is obtained, 4-allyl-biphenyl ( 50 ) in 33% yield (Scheme 9). With reaction times below 2 hours the acetate 53 from 15 was isolated, and this could be converted into 50 . The reaction of 2′,3′,3′-d3-15 in Ac2O/H2SO4 lead to 1′,1′,2′-d3-50 (Scheme 11). The rearrangement of 4-allyl-4-methyl-cyclohex-2-en-1-ol (16) (Scheme 14) yielded 39% of the corresponding acetate 60 and 30% of 4-allyl-toluene ( 6 ), which also resulted by a rearrangement of 60 under the reaction conditions. These rearrangements are all [3s,4s]-sigmatropic reactions, which proceed via the cyclohexenyl cation a (Scheme 12, R = C6H5, CH3). In Ac2O/H2SO4 the allyl-cyclohexadienes primarely formed subsequently undergo dehydrogenation to yield the benzene derivatives 6 , 50 and d3- 50 . From the rearrangement of 4,4-diphenyl-cyclohex-2-en-1-ol ( 19 ) at 0° a reaction mixture is obtained which consists of the acetate 55 , 2,3-diphenyl-cyclohexa-1,4-diene ( 57 ) and o-terphenyl ( 56 ) (Scheme 10). Both 55 and 57 are converted under the reaction conditions to o-terphenyl ( 56 ). No 4-(1′-methylallyl)-biphenyl is obtained from the rearrangement of 4-crotyl-4-phenyl-cyclohex-2-en-1-ol ( 17 ). In this case, apart from the corresponding acetate 64 , a single product 5-(1′-acetoxyethyl)-1-phenyl-bicyclo[2.2.2]oct-2-ene ( 65 ) (Scheme 16) was obtained; under the reaction conditions the acetate 64 rearranges to 65 . The rearrangement of 4-allyl-4-phenyl-cyclohexa-2,5-dien-1-ol ( 20 ) gives, as expected, not only 4-allyl-biphenyl ( 50 ) but also 2- and 3-allyl-biphenyl ( 51 and 52 ) and biphenyl (Scheme 13). 4-Benzyl-4-methyl-cyclohexa-2,5-dien-1-ol (syn- and anti- 21 ) gave in Ac2O/H2SO4 at 10° as rearrangement products 93% of 2-benzyltoluene ( 97 ) and 7% of 4-benzyl-toluene ( 98 ) (Scheme 21). Hence [1,4]-rearrangements in cyclohexadienyl cations, seems to occur only to a limited extent. The alicyclic alcohols 22 and 23 (Scheme 18) gave, in Ac2O/H2SO4, as main product the corresponding acetates 73 and 75 , as well as small amounts of olefins 74 and 76 formed by dehydration i.e. [3,4]-rearrangements occur in these systems. Also no [3,4]-rearrangements were observed in solvents reactions of either 4,4-dimethyl-hepta-1, 6-dien-3-yl tosulate (79; see Scheme 19) or its corresponding alcohol 24.  相似文献

Summary The reaction of 3-aroyl-6-aryl-4-hydroxy-2H-pyran-2-ones (Ar=p-tolyl, 1,1-biphenyl-4-yl or thienyl) with aniline and substitutedo-phenylenediamine (R=H, CH3 or Cl) yields a series of new Schiff bases2a–f in 51–72% yield. Bromination of1a gave the 5-bromo derivative1c, while the compounds1a,1b,2b,2e, and2f were converted into 2,6-diaryl-4H-pyran-4-ones3a–c. All products have been fully characterized.

Zusammenfassung Die Reaktion von 3-Aroyl-6-aryl-4-hydroxy-2H-pyran-2-onen (Ar=p-Tolyl, 1,1-Biphenyl-4-yl oder Thienyl) mit Anilin und substituierteno-Phenylendiaminen liefert neue Schiff'sche Basen2a–f/bd in 51–72% Ausbeute. Bromierung von1a gab das 5-Bromderivate1c, während die Verbindungen1a,1b,2b,2e und2f in 2,6-Diaryl-4H-pyran-4-onen3a–c übergeführt wurden. Alle Produkte wurden voll charakterisiert.

Elastic and dissipative properties of granular assemblies under uniaxial compression are studied both experimentally and by numerical simulations. Following a novel compaction procedure at varying oscillatory pressures, the stress response to a step strain reveals an exponential relaxation followed by a slow logarithmic decay. Simulations indicate that the latter arises from the coupling between damping and collective grain motion predominantly through sliding. We characterize an analogous "glass transition" for packed grains, below which the system shows aging in time-dependent sliding correlation functions.  相似文献

The N-aryl- and N-heteroaryl-2,5-dimethylpyrrole-3-carbaldehydes 2–8 have been synthesized by condensation of hexane-2,5-dione with the appropriate amine and subsequent Vilsmeier-Haack formylation of the pyrrole ring. Diastereomeric association complexes of the racemic pyrrole 8 were studied by pmr chemical shifts and the splittings induced by (+)-1-(9-anthryl)-2,2,2-trifluoroethanol. Enrichment of the enantiomers M and P of 2 and 6 was achieved by liquid chromatography on triacethylcellulose. The barrier to partial rotation about the C-N bond in 6 was determined by thermal racemization and its lower limits in 2 and 8 were estimated.  相似文献

A general approach for the detection and structural elucidation of brain ganglioside species GM1, GD1 and GT1 by nano-electrospray ionization quadrupole time-of-flight (nanoESI-QTOF) mass spectrometry (MS), using combined data from MS and MS/MS analysis of isolated native ganglioside fractions in negative ion mode and their permethylated counterparts in the positive ion mode is presented. This approach was designed to detect and sequence gangliosides present in preparatively isolated ganglioside fractions from pathological brain samples available in only very limited amounts. In these fractions mixtures of homologue and isobaric structures are present, depending on the ceramide composition and the position of the sialic acid attachment site. The interpretation of data for the entire sequence, derived from A, B, C and Y ions by nanoESI-QTOFMS/MS in the negative ion mode of native fractions, can be compromised by ions arising from double and triple internal cleavages. To distinguish between isobaric carbohydrate structures in gangliosides, such as monosialogangliosides GM1a and GM1b, disialogangliosides GD1a, GD1b and GD1c or trisialogangliosides GT1b, GT1c and GT1d, the samples were analysed after permethylation in the positive ion nanoESI-QTOFMS/MS mode, providing set of data, which allows a clear distinction for assignment of outer and inner fragment ions according to their m/z values. The fragmentation patterns from native gangliosides obtained by low-energy collision induced dissociation (CID) by nanoESI-QTOF show common behaviour and follow inherent rules. The combined set of data from the negative and positive ion mode low-energy CID can serve for the detection of structural isomers in mixtures, and to trace new, not previously detected, components.  相似文献

Bovine ribonuclease B (RNAse B) and asialofetuin (FETUA) were subjected to in-capillary tryptic digest (Pohlentz et al. Proteomics. 2005, 5, 1758-1763) and the obtained glycopeptides were analyzed, respectively, by nanoelectrospray ionization mass spectrometry and collision-induced dissociation (CID) during the ongoing digest. For RNAse, B glycans of the high-mannose type (Man(4) to Man(9)) attached to either a tetra- or a hexapeptide containing the sole N-glycosylation site of the protein were detected. Glycopeptides derived from all three N-glycosylation sites of FETUA were observed, and the corresponding CID spectra proved the respective glycans to be oligosaccharides of the triantennary complex type. Moreover, an O-glycopeptide carrying Gal-GalNAc at T(280) could be unambiguously identified. An in-solution tryptic/chymotryptic digest of human transferrin (TRFE) was analyzed directly for glycopeptides subsequent to the addition of methanol and formic acid. Disialylated diantennary glycans were observed in glycopeptides of both N-glycosylation sites of TRFE. These results demonstrate the feasibility of direct structure determination of glycopeptides in proteolytic mixtures without any further refurbishment.  相似文献

A novel kinetic method for micro-quantitative determinations of morphine (MH) is proposed and validated. The method is based on the potentiometric monitoring of the concentration perturbations of the oscillatory reaction system being in a stable non-equilibrium stationary state close to the bifurcation point between stable and oscillatory state. The response of the Bray-Liebhafsky (BL) oscillatory reaction as a matrix system, to the perturbations by different concentrations of morphine, is followed by a Pt-electrode. The proposed method relies on the linear relationship between maximal potential shift, ΔEm, and the logarithm of the added morphine amounts in the range of 0.004-0.18 μmol. Under optimum conditions, the sensitivity of the proposed method (as the limit of detection) is 0.001 μmol and the method is featured by good precision (R.S.D. = 1.6%) as well as the excellent sample throughput (45 samples h−1). In addition to standard solution analysis, this approach was successfully applied for quantitative determination of morphine in a typical pharmaceutical dosage form. Some aspects of the possible mechanism of morphine action on the BL oscillating chemical system are discussed in detail.  相似文献

The generation of synthetic compounds with exclusive target specificity is an extraordinary challenge of molecular recognition and demands novel design strategies, in particular for large and homologous protein families such as protein kinases with more than 500 members. Simple organic molecules often do not reach the necessary sophistication to fulfill this task. Here, we present six carefully tailored, stable metal-containing compounds in which unique and defined molecular geometries with natural-product-like structural complexity are constructed around octahedral ruthenium(II) or iridium(III) metal centers. Each of the six reported metal compounds displays high selectivity for an individual protein kinase, namely GSK3α, PAK1, PIM1, DAPK1, MLCK, and FLT4. Although being conventional ATP-competitive inhibitors, the combination of the unusual globular shape and rigidity characteristics, of these compounds facilitates the design of highly selective protein kinase inhibitors. Unique structural features of the octahedral coordination geometry allow novel interactions with the glycine-rich loop, which contribute significantly to binding potencies and selectivities. The sensitive correlation between metal coordination sphere and inhibition properties suggests that in this design, the metal is located at a "hot spot" within the ATP binding pocket, not too close to the hinge region where globular space is unavailable, and at the same time not too far out toward the solvent where the octahedral coordination sphere would not have a significant impact on potency and selectivity. This study thus demonstrates that inert (stable) octahedral metal complexes are sophisticated structural scaffolds for the design of highly selective chemical probes.  相似文献
