Melanin in the long-lived melanosomes of the retinal pigment epithelium (RPE) may undergo photobleaching with aging, which appears to diminish the antioxidant function of melanin and could make photobleached melanosomes less efficient in protecting biomolecules from oxidative modification. Here we analyzed whether photobleaching of melanosomes affects their ability to modify the oxidation state of nearby protein. As conventional methods developed to study soluble antioxidants are not well suited for analysis of granules such as melanosomes, we developed a new analytic method to focus on particle surfaces that involves experimentally coating granules with the cytoskeletal protein beta-actin to serve as a reporter for local protein oxidation. Isolated porcine RPE melanosomes were photobleached with visible light to simulate aging, then photobleached melanosomes, untreated melanosomes and control particles (black latex beads) were actin coated and illuminated in a photosensitized cell free system. Protein was re-stripped from particles and analyzed for carbonylation by Western blotting. Quantitative densitometry showed no reproducible differences for protein associated with untreated melanosomes when compared with control particles. Melanin has both anti- and pro-oxidant functions when light irradiated, but neither of these functions predominated in the protein oxidation assay when untreated melanosomes were used. However, protein extracted from photobleached melanosomes showed markedly increased carbonylation, both of associated actin and of endogenous melanosomal protein(s), and the effect increased with extent of granule photobleaching. Photobleaching of RPE melanosomes therefore changes the oxidation state of protein endogenous to the organelle and reduces the ability of the granule to modify the oxidation of exogenous protein near the particle surface. The results support the growing body of evidence that photobleaching of RPE melanosomes, which is believed to occur with aging, changes the physicochemical properties of the organelle and reduces the likelihood that the granules perform an antioxidant function.  相似文献

The present study aimed to prepare and characterize amylose inclusion complexes with rhynchophorol aggregation pheromone. The potato and cassava V-amylose–rhynchophorol complexes were successfully obtained, but the B-type polymorph was also formed. The potato (64.6%) and cassava (40.9%) amylose complexes have presented high crystalline indexes, confirmed by the XRD technique. No endothermic peaks associated with the melting of crystalline arrays were noticed between 30 and 100°C. The dynamic TG runs demonstrated that both the degree of order of crystalline arrays and molecular weights influence the thermal stabilities of the V-amyloses-rhynchophorol. The isothermal TG showed that the retention of rhynchophorol seems to be directly proportional to the molecular weights of starch polymers. The recovered rhynchophorol contents were in the order of 10?7 grams per 50 milligrams of dried powders, which would be useful for insect attraction as demonstrated by the GC–MS technique. The starch polymers studied have presented suitable for retention of rhynchophorol guest molecules.

By virtue of the soft x-ray angle-resolved photoelectron spectroscopy, the three-dimensional bulk fermiology has been successfully performed for a strongly correlated Ce compound, ferromagnet CeRu2Ge2 in the paramagnetic phase. A clear difference of the Fermi surface topology from either band calculation or de Haas-van Alphen results in the ferromagnetic phase is observed and interpreted by considering the difference of the 4f contribution to the Fermi surfaces in the paramagnetic phase.  相似文献

The precipitation of phenytoin sodium injection provoked by mixing with infusion fluids renders its use in clinical practice difficult, as rapid intravenous (i.v.) push and i.v. infusion are supposed to be avoided. As some of its aspects remain unclear, this study tried to elucidate this precipitation mechanism. In particular, this study focused on the significant precipitation induced by glucose infusion fluid. The precipitation provoked by 5% glucose infusion fluid was obviously different from the precipitation that accompanied simple pH reduction, in terms of the growth mode and morphology of crystals. In addition, the effect of glucose was partially unrelated to pH reduction. NMR measurements including a two-dimensional nuclear Overhauser effect spectroscopy (2D-NOESY) spectrum indicated the specific interaction between glucose and propylene glycol, which is incorporated into phenytoin sodium injection as a solubilizing agent. These results led to the conclusion that this interaction was crucial for the precipitation of phenytoin, as it diminished the solubilizing effect of propylene glycol, resulting in the enhancement of the crystallization of phenytoin. The determination of phenytoin solubility in aqueous solutions at different pH values revealed that phenytoin incorporated in the admixture could be dissolved completely, as long as the injection was diluted with saline or water. These findings offer a profound insight into the formulation design of phenytoin sodium injection and its use in clinical practice.  相似文献

The field-orientation dependent thermal conductivity of the heavy-fermion superconductor UPt3 was measured down to very low temperatures and under magnetic fields throughout the distinct superconducting phases: B and C phases. In the C phase, a striking twofold oscillation of the thermal conductivity within the basal plane is resolved reflecting the superconducting gap structure with a line of node along the a axis. Moreover, we find an abrupt vanishing of the oscillation across a transition to the B phase, as a clear indication of a change of gap symmetries. We also identify extra two line nodes below and above the equator in both B and C phases. From these results together with the symmetry consideration, the gap function of UPt3 is determined as a E(1u) representation characterized by a combination of two line nodes at the tropics and point nodes at the poles.  相似文献

Polypropylene block copolymer (PPco) is easier to process in extrusion-based additive manufacturing compared to isotactic PP homopolymer because it shrinks and warps less during printing. This study investigated the thermal properties of PPco and spray-dried CNF (SDCNF)-PPco composite pellet formulations. Thermogravimetric analysis data showed that the addition of SDCNF lowered the thermal degradation onset temperature and increased the residual mass content (at 450 °C) of PPco pellets. The peak degradation temperatures of SDCNF and PPco remained the same. The peak crystallization temperature and relative crystallinity of PPco were increased by the addition of SDCNF and MAPP. The peak melting temperature of PPco was not significantly changed. Printed specimens showed three melting peaks (β, β′ and α crystals) while injection molded PPco only showed one (α crystal) melting peak. Dynamic mechanical analysis results showed that the heat deflection temperatures of printed SDCNF-PPco composites were higher than injection molded PPco. SEM micrographs showed that the addition of SDCNF increased the number of voids inside the printed parts.

We observe several non-Fermi liquid behaviors in the normal-state transport properties of CeMIn5 (M: Rh and Co) under pressure at low temperatures: (1) The dc-resistivity shows T-linear dependence, ρxxT. (2) The magnitude of Hall coefficient |RH| increases rapidly with decreasing temperature, and reaches a value much larger than |1/ne| at low temperatures. (3) The magnetoresistance displays T- and H-dependence that strongly violate Kohler's rule, and is well scaled by the tangent of the Hall angle, . These non-Fermi liquid properties in the electron transport are remarkably pronounced when the AF fluctuations are enhanced in the vicinity of the quantum critical point. Since all of these salient features have been also reported for high-Tc cuprates, we infer that the non-Fermi liquid transport properties capture universal features of strongly correlated electron systems in the presence of strong antiferromagnetic fluctuations.  相似文献
