Comprehensive two-dimensional liquid chromatography-size-exclusion chromatography (LC x SEC) was investigated as a tool for the characterization of functional poly(methyl methacrylate) (PMMA) polymers. Ultraviolet-absorbance and evaporative light-scattering detection (ELSD) were used. A simple method to quantify ELSD data is presented. Each data point from the ELSD chromatogram can be converted into a mass concentration using experimental calibration curves. The qualitative and quantitative information obtained on two representative samples is used to demonstrate the applicability of LC x SEC for determining the mutually dependent molar-mass distributions (MMD) and functionality-type distributions (FTD) of functional polymers. The influence of the molar mass on the retention behavior in LC was investigated using LC x SEC for hydroxyl-functional PMMA polymers. The critical conditions, at which retention is--by definition--independent of molar mass, were not exactly the same for PMMA series with different end-groups. Our observations are in close agreement with theoretical curves reported in the literature. However, for practical applications of LC x SEC it is not strictly necessary to work at the exact critical solvent composition. Near-critical conditions are often sufficient to determine the mutually dependent distributions (MMD and FTD) of functional polymers.  相似文献

2′-Deoxy-5-(isothiazol-5-yl)uridine ( 12 ) was synthesized starting from 2′-deoxy-5-iodouridine using a Pd-catalysed cross-coupling reaction with propiolaldehyde diethyl acetal followed by deprotection and ring closure using thiosulfate. 2′-Deoxyuridine 12 has a particular place among the 5-heteroaryl-substituted 2′-deoxyuridines in that it has a high affinity for herpes simplex virus type 1 (HSV-1)-encoded thymidine kinase (TK) without antiviral activity. Biochemical studies revealed that 12 is a substrate for viral TK. We further investigated the interaction of 12 with the HSV-1 thymidine kinase. The conformation of 12 in solution was established by NMR spectroscopy. The most stable conformer 12A has the S-atom of the isothiazole ring placed in the neighbourhood of the C(4)?O group of the pyrimidine moiety. The compound was docked in its most stable conformation in the active site of HSV-1 TK and subjected to energy minimization. This demonstrated that the isothiazole moiety binds in a cavity lined by the side chains of Tyr-132, Arg-163, Ala-167, and Ala-168 and that the C(3) atom of the isothiazole moiety is located in close proximity of the phenolic O-atom of Tyr-132 and the aliphatic part of the Arg-163 side chain.  相似文献

 The relationship between hydrogen bonding and NMR chemical shifts in the catalytic triad of low-pH α-chymotrypsin is investigated by combined use of the effective fragment potential [(2001) J Phys Chem A 105:293] and ONIOM–NMR [(2000) Chem Phys Lett 317:589] methods. Our study shows that while the His57 Nδ1−H bond is stretched by a relatively modest amount (to about 1.060 ?) this lengthening, combined with the polarization due to the molecular environment, is sufficient to explain the experimentally observed chemical shifts of 18.2 ppm. Furthermore, the unusual down-field shift of Hɛ1 (9.2 ppm) observed experimentally is reproduced and shown to be induced by interactions with the C=O group of Ser214 as previously postulated. The free-energy cost of moving Hδ1 from His57 to Asp102 is predicted to be 5.5 kcal/mol. A rapid method was developed for the simultaneous determination of hippuric and orotic acid in rennet whey by capillary zone electrophoresis using an uncoated capillary utilizing a 0.04 M amino-2-methyl-1,3-propanediol (AMPD)-N,N-bis(2-hydroxyethyl) glycine (BICINE) buffer (pH 8.8) with UV detection at 254 and 280 nm. Whey proteins were removed by ultrafiltration. The method was evaluated for external, internal and standard addition procedures for both peak areas and peak heights. The use of an internal standard (sorbic acid) eliminated injection errors and gave, when applied to peak areas, the same levels for hippuric and orotic acid in those obtained with high-performance liquid chromatography. Relative standard deviations were 1-2%. Peak heights gave erratic results owing to sample matrix effects on peak widths.  相似文献

The lateral alignment of [012] habit-modified calcite crystals with respect to a carboxylic acid terminated self-assembled monolayer (SAM) of thiols has been determined. The crystals were grown from a Kitano solution (pH 5.6-6.0), and the samples were investigated with scanning electron microscopy, X-ray diffraction, and polarization microscopy. For the first time, a lattice match in one direction, which is the nearest neighbor direction of the SAM and the calcite <100> direction, has been experimentally shown. The experimental results are in good agreement with the theoretical models proposed in previous work, and it is expected that this method can be applied to similar systems where inorganic crystals nucleate with a preferred orientation to a SAM.  相似文献

The asymmetric unit of the title compound, C11H5D16N2O2·0.33H2O, is formed by three crystallographically independent piperidin‐1‐yloxyl mol­ecules and a mol­ecule of water. The mol­ecules are crosslinked by nine hydrogen bonds into layers parallel with the ac plane. The reactions of a series of 5-alkyl-2-thiophenedithiocarboxylates with nickel(II) chloride afforded two types of complexes, blue nickel(II) complexes with two terminal dithiocarboxylate ligands, [Ni(S2CTR)2] and violet nickel(II) complexes with perthio- and dithiocarboxylate ligands, [Ni(S2CTR)(S3CTR)] (where T = 2,5-disubstituted thiophene, R = CnH2n+1, n = 4, 6, 8, 12, 16). The blue monomers are preferred for the shorter chains (C4 and C6) and the violet compounds form exclusively for the longer chains (C8, C12, and C16) in the alkylthiophene complexes. In addition to the above series, [Ni(S2CTCH3)2], was prepared in a one-pot reaction in THF and both the blue and violet products were isolated. It was possible to convert the blue complexes [Ni(S2CTR)2] (R = butyl, hexyl) into the corresponding violet complexes [Ni(S2CTR)(S3CTR)] after stirring in THF solutions for prolonged periods of time. Liquid-crystalline properties of these complexes were examined by DSC and POM. The violet complexes with C8 and C12 alkyl chains showed liquid-crystalline properties.  相似文献

Depending on the solvent used and the ratio of the reactants, N-acylpyridinium salts condense with indole to give 3-(N-acyl-1,4-dihydro-4-pyridyl)indole ( 1 ) or 4-(N-acyl-3-indolyl)pyridinium chloride ( 3 ). Compound 1 is an intermediate in the formation of compound 3 . The reaction mechanism has been studied, and a hydrogen transfer reaction is suggested as a key step. Alkaline hydrolysis, e.g., of 4-(N-acetyl-3-indolyl)pyridinium chloride ( 3a ), gave 3-(4-pyridyl)indole ( 2a ). The reaction of α-chlorosubstituted acyl halides with indole, in the presence of pyridine constitutes a convenient synthesis of 3-chloroacylindoles.  相似文献
