Nonsteroidal antiinflammatory drugs(NSAIDs) are known as clinically effective agents for treatment of inflammatory diseases. Inhibition of cyclooxygenase has been thought to be a major facet of the pharmacological mechanism of NSAIDs. However, it is difficult to ascribe the antiinflammatory effects of NSAIDs solely to the inhibition of prostaglandin synthesis. Human neutrophil elastase (HNElastase; HNE, EC 3.4.21.37) has been known as a causative factor in inflammatory diseases. To investigate the specific relationship between HNElastase inhibition and specificity of molecular structure of several NSAIDs, HNElastase was purified by Ultrogel AcA54 gel filtration, CM-Sephadex ion exchange, and HPLC (with TSK 250 column) chromatography. HNElastase was inhibited by aspirin and salicylate in a competitive manner and by naproxen, ketoprofen, phenylbutazone, and oxyphenbutazone in a partial competative manner, but not by ibuprofen and tolmetin. HNElastase-phenylbutazone-complex showed strong Raman shifts at 200, 440, 1124, 1194, 1384, 1506, and 1768 cm(-1). The Raman bands 1194, 1384, and 1768 cm(-1) may represent evidences of the conformational change at -N=N-phi radical, pyrazol ring, and -C=O radical of the elastase-drug complex, respectively. Phenylbutazone might be bound to HNElastase by ionic and hydrophobic interaction, and masked the active site. Inhibition of HNElastase could be another mechanism of action of NSAIDs besides cyclooxygenase inhibition in the treatment of inflammatory diseases. Different inhibition characteristics of HNE-lastase by NSAIDs such as aspirin, phenylbutazone-like drugs and ineffective drugs could be important points for drawing the criteria for appropriate drugs in clinical application.  相似文献

Surface sorption experiments of U(VI) onto the surfaces of a Korean granite rock are carried out in order to investigate the kinetics and reversibility of U(VI) sorption as a function of pH and surface types such as fresh intact surfaces and natural fracture surfaces. It was shown that the effect of pH is significant in the sorption of U(VI) onto both types of the granite surfaces. However the sorption rates do not greatly depend upon the pH regardless of the surface types. A two-step first order kinetic behavior dominates onto both the intact surfaces and natural fracture surfaces of granite and that the linearization approach of the kinetic model agrees well with experimental sorption data. The desorption results showed that the sorption process of U(VI) was a little irreversible for the two types of granite surfaces regardless of pH and surface types. This kinetic approach could give a better understanding of U(VI) sorption onto granite surfaces depending on pH and surface types. To obtain effective and safe topical depigmenting agents, we synthesized hydroxybenzoates, alkoxybenzoates, and 3,4,5-trimethoxycinnamate containing a thymol moiety and screened then for high-level inhibitory activity against melanin synthesis in cultured melanocytes. Eight compounds were tested for their depigmenting effect and cytotoxicity using a murine melanocyte cell line. We found that 3,4,5-trialkoxybenzoates and 3,4,5-trimethoxycinnamate, synthesized by conjugating 3,4,5-trialkoxybenzoic acids and 3,4,5-trimethoxycinnmic acid with thymol, showed a potent depigmenting effect and low cytotoxicity. Compound 4h, 5-methyl-2-(methylethyl)phenyl (2E)-3-(3,4,5-trimethoxyphenyl)prop-2-enoate, showed the most potent depigmenting effect (IC50=10 microM) with low cytotoxicity (IC50=200 microM).  相似文献

We consider quantum unbounded spin systems (lattice boson systems) in -dimensional lattice space Z. Under appropriate conditions on the interactions we prove that in a region of high temperatures the Gibbs state is unique, is translationally invariant, and has clustering properties. The main methods we use are the Wiener integral representation, the cluster expansions for zero boundary conditions and for general Gibbs state, and explicitly -dependent probability estimates. For one-dimensional systems we show the uniqueness of Gibbs states for any value of temperature by using the method of perturbed states. We also consider classical unbounded spin systems. We derive necessary estimates so that all of the results for the quantum systems hold for the classical systems by straightforward applications of the methods used in the quantum case.  相似文献

Muon spin relaxation (SR) studies have been performed in the normal spinel LiTi2O4 and the A-15 superconductor V3Si to measure the magnetic penetration depth . The relaxation rate(T) 1/2 in field-cooled measurements shows a sharp increase belowT c followed by saturation at low temperatures in both systems. This feature implies an isotropic energy gap without anomalous zeros, and most likelys-wave pairing. The low temperature penetration depth (T 0) is determined to be 2100Å for LiTi2O4 and 1300Å for V3Si respectively. Assuming a clean limit relation –2 n s /m *, we derive the Fermi temperatureT F n s/ 2/3 m * from the relaxation rate and the Sommerfeld constant asT F 3/4–1/4. Unlike conventional superconductors, both LiTi2O4 and V3Si have a large ratio ofT c /T F 0.01, only slightly smaller than those ratios in more exotic superconductors.We thank C. Ballard and K.
