As a part of studies on MDR reversing agents, the structure of a benzylamido-9,10-dihydro-9,10-ethenoanthracene is reported: C24 H19 N O; orthorhombic; space group: Pca21; a = 9.214(2), b = 18.624(4), c = 10.170(2) Å; 1745.2(6) Å3; Z = 4. The three nonplanar rings from the 9,10-dihydro-9,10-ethenoanthracene skeleton of the molecule adopt a boat conformation. The benzamide side-chain is extended (conformation, trans-trans-gauche). Human ovarian carcinoma cells (MDAH 2774) were treated with sodium ascorbate (VC), menadione (VK3), or with a VC:VK3 combination for 1 h and then studied using light microscopy (LM) and scanning (SEM) and transmission electron (TEM) microscopy. Plasma membrane damage (blisters and blebs, hairy aspect) results from vitamin C (VC) treatment, while cytoskeletal damage and self-morsellation are caused by vitamin K3 (VK3) treatment. VC:VK3-treated cells exhibit exacerbated injuries characteristic of both VC and VK3 treatment as well as a significant decrease in cell diameters from 20-35 microm for control cells to 7-12 microm for VC:VK3 treatment. Moreover, after a 1-h exposure to the vitamin combination, autoschizis (43%), apoptosis (3%), and oncosis (1.9%) are observed at the percentages indicated. All cellular changes associated with autoschizis observed with SEM were confirmed by LM and TEM observations and are consistent with cell death by autoschizis: decrease in cell size, cytoplasmic self-excisions, degradation of the nucleus and nucleolus without formation of apoptotic bodies and, ultimately, karyorrhexis and karyolysis. These results also suggest that the vitamin combination may find clinical use in the treatment of ovarian cancer. 相似文献

The direct silylation by Me3SiCl/Li/THF of saturated nitriles having a hydrogen atom in the α position with respect to the function leads, according to a simple and rapid process, to the corresponding α-silylated nitriles in yields that are moderate but much higher than those given by the comparable previously known routes. α, β-Unsaturated nitriles give the derivatives resulting from the disilylation of the double bond. Allyl or benzyl cyanide and diphenyl acetonitrile first undergo the elimination of the nitrile group (which behaves as a halogen atom) whereas cyanamid affords bis(trimethylsilyl)carbodiimide giving after further silylation the unexpected tris(trimethylsilyl)amine. 相似文献

[reaction: see text] A new access to benzhydryl-phenylureas is described. These new interesting urea derivatives were obtained by reaction of substituted benzils with substituted phenylureas under microwave irradiation. Phenylthiourea, when reacted with benzil, gave 3-phenyl-thiohydantoin. Moreover, benzylurea, as phenethylurea, gave the corresponding 3-substituted hydantoin derivatives, demonstrating that only phenylurea derivatives can result in benzhydryl-phenylureas under the applied conditions. A small, highly sensitive, inexpensive fluorimeter for the detection of very low fluorescence intensities for analytical and clinical purposes is described. High sensitivity is achieved by use of an inexpensive continuous wave laser with a power of a few milliwatts. The optical arrangement of the instrument has been thoroughly designed so that the effects of Rayleigh and Raman scattering are minimized. Fluorescence is dected through optical filters with a side-on photomultiplier. Concentrations of rhodamine B in ethanol as low as 10?11 M can be measured. To use this instrument for a fluoroimmunoassay of urinary estrogens (10?9?10?7 M), a purified monoclonal IgG, raised against estradiol, was labelled with RITC (rhodamine isothiocyanate) in a 1:1 molar ratio. In this paper, the behaviour of ethers, sulfides, selenoethers, amines, germanes and stannanes, substituted on the β-C by a methoxycarbonyl, methyl ketone, or cyano group is compared and discussed. The compounds containing group VB or VIB element are quite stable, while those with a group IVB element fragment very easily. Fragmentation α to the heteroatom gives peaks for which the intensity increases with the size of the element:[PhO]+<[PhS]+<[PhSe]+<[Ph3Ge]+<[Ph3Sn]+. In connection with a total synthesis of cephalotaxine (1a), we have examined the addition of various nucleophilic reagents to [ABC] subunits 2 and 7 possessing a pyrrolobenzazepine core. In fact, this reaction implicates invariably the carbonyl group of 2. Regarding the reaction of 7 with nucleophiles, the most striking aspect is the complete lack of reactivity of the enaminonitrile moiety. For instance, the condensation of 7 with methylmagnesium bromide involves exclusively the cleavage of the dioxole ring, yielding regioisomers 9 and 10. With the aim of understanding the unexpected reactivity of 2 and 7 toward nucleophiles, crystallographic studies of 2 and 7 and an experimental electron density determination of 7 were carried out. The marked reactivity of the carbonyl group of 2 was interpreted by invoking the weakness of the amide resonance, due to a pronounced delocalization of the N(9) lone pair over the enaminonitrile moiety. The electron density study of 7 reveals this electron delocalization along the enaminonitrile fragment, highlighted and quantified through the bond geometries, topological indicators, and atomic charges, a phenomenon that is responsible for the failure of the addition of nucleophilic species. 相似文献
