The European Southern Observatory (ESO) together with external research institutes have built a Multi-Conjugate Adaptive Optics (MCAO) Demonstrator (MAD) to perform wide field-of-view adaptive optics correction (2′ in K band). The aim of MAD is to demonstrate the on-sky feasibility of the MCAO technique and to evaluate its critical aspects in the framework of both the 2nd generation instrumentation for the Very Large Telescope (VLT) and the Overwhelmingly Large Telescope (OWL). The MAD module will be installed on the VLT to perform on-sky observations. MAD comprises two deformable mirrors and two different multi-reference wavefront sensors with natural guide stars. In this article we present the MAD design, some aspects of the MAD calibration and the first closed-loop results in the laboratory in Single Conjugated Adaptive Optics (SCAO) and Ground Layer Adaptive Optics (GLAO) configurations.  We present a discrete iterative interpolation scheme (DIIS) to improve the convergence rate of electrostatic calculations in the polarizable continuum model (PCM) to describe solvent effects on molecular solutes. The electrostatic calculations may easily become the bottleneck of the calculation when the solute size is large. For large molecules iterative procedures turn out to be computationally more convenient than matrix inversion or closure methods. The DIIS scheme is compared here to another iterative procedure (DAMP) and to the biconjugate gradient (BCG) method. The comparisons show that DIIS leads to a sizeable saving of computational time for the C-PCM and IEF-PCM methods (average 40%) compared to DAMP, and more than 50% with respect to the BCG method. There is a vast genomic resource for enzymes active on carbohydrates. Lagging far behind, however, are functional chemical tools for the rapid characterization of carbohydrate-active enzymes. Activity-based probes (ABPs) offer one chemical solution to these issues with ABPs based upon cyclophellitol epoxide and aziridine covalent and irreversible inhibitors representing a potent and widespread approach. Such inhibitors for enzymes active on polysaccharides are potentially limited by the requirement for several glycosidic bonds, themselves substrates for the enzyme targets. Here, it is shown that non-hydrolysable trisaccharide can be synthesized and applied even to enzymes with challenging subsite requirements. It was found that incorporation of carbasugar moieties, which was accomplished by cuprate-assisted regioselective trans-diaxial epoxide opening of carba-mannal synthesised for this purpose, yields inactivators that act as powerful activity-based inhibitors for α-1,6 endo-mannanases. 3-D structures at 1.35–1.47 Å resolutions confirm the design rationale and binding to the enzymatic nucleophile. Carbasugar oligosaccharide cyclophellitols offer a powerful new approach for the design of robust endoglycosidase inhibitors, while the synthesis procedures presented here should allow adaptation towards activity-based endoglycosidase probes as well as configurational isosteres targeting other endoglycosidase families.  相似文献

Maleimide chemistry is widely used in the site-selective modification of proteins. However, hydrolysis of the resultant thiosuccinimides is required to provide robust stability to the bioconjugates. Herein, we present an alternative approach that affords simultaneous stabilisation and dual functionalisation in a one pot fashion. By consecutive conjugation of a thiol and an amine to dibromomaleimides, we show that aminothiomaleimides can be generated extremely efficiently. Furthermore, the amine serves to deactivate the electrophilicity of the maleimide, precluding further reactivity and hence generating stable conjugates. We have applied this conjugation strategy to peptides and proteins to generate stabilised trifunctional conjugates. We propose that this stabilisation-dual modification strategy could have widespread use in the generation of diverse conjugates.

An alternative approach to maleimide conjugate stabilisation is presented, by the consecutive addition of a thiol and an amine to dibromomaleimides. The amine serves to simultaneously deactivate the maleimide and enable dual functionalisation.  相似文献

Rats fed a diet deficient in vitamin D were found to exhibit a refractory cyclic AMP response of kidney slices to parathyroid hormone and a marked decrease in membrane parathyroid hormone-dependent adenylate cyclase activity. Both the characteristic calcium deficiency (hypocalcemia) and secondary elevation of circulating parathyroid hormone appeared before the first noticeable decrease in hormone-dependent enzyme activity. After repletion of D-deficient rats with vitamin D2, we found that serum calcium and parathyroid hormone were both restored to normal levels before the depressed enzyme response to the hormone was reversed. Moreover, infusion of parthyroid hormone into vitamin D-replete rats led to a marked reduction in parathyroid hormone-dependent adenylate cyclase activity, which was partly restored to control level 3 hours after discontinuing the hormone infusion. Taken as a whole, this study suggests that the elevated endogenous parathyroid hormone in the vitamin D-deficient rat is involved in the "down-regulation" of renal cyclic AMP responsiveness to the hormone. However, these experiments do not rule out the possibility that calcium deficiency and/or vitamin D per se participate in the regulation of the renal cyclic AMP response to parathyroid hormone.  相似文献

We present a model to evaluate the radiative and nonradiative lifetimes of electronic excited states of a molecule close to a metal particle of complex shape and, possibly, in the presence of a solvent. The molecule is treated quantum mechanically at Hartree-Fock (HF) or density-functional theory (DFT) level. The metal/solvent is considered as a continuous body, characterized by its frequency dependent local dielectric constant. For simple metal shapes (planar infinite surface and spherical particle) a version of the polarizable continuum model based on the integral equation formalism has been used, while an alternative methodology has been implemented to treat metal particles of arbitrary shape. In both cases, equations have been numerically solved using a boundary element method. Excitation energies and nonradiative decay rates due to the energy transfer from the molecule to the metal are evaluated exploiting the linear response theory (TDHF or TDDFT where TD--time dependent). The radiative decay rate of the whole system (molecule + metal/solvent) is calculated, still using a continuum model, in terms of the response of the surrounding to the molecular transition. The model presented has been applied to the study of the radiative and nonradiative lifetimes of a lissamine molecule in solution (water) and close to gold spherical nanoparticles of different radius. In addition, the influence of the metal shape has been analyzed by performing calculations on a system composed by a coumarin-type molecule close to silver aggregates of complex shape.  相似文献

The reaction of ring opening of epoxides under the action of a halogenic acid has been investigated using as model the system C2H4O + HF → CH2OHCH2F. The physical conditions chosen for the model more directly correspond to the gas phase reaction and under these conditions the preferred mechanism leads to the formation of a halohydrine having the same configuration at the C atom as the reagent. Parallel investigations have been performed on other mechanisms which postulate the preliminary formation of the conjugate acid of the oxirane (C2H4OH+) and proceed via the well known mechanismsA 1 orA 2. In this case the best mechanism corresponds to the so-called “borderlineA 2” mechanism. This last type of mechanism probably is the dominant one in protic solutions, but by coupling the present calculations with experimental conductometric measurements in anhydrous aprotic media one could consider the first concerted mechanism as a possible candidate also for the reaction in “inert” media. A qualitative analysis of the transition state indicates, in addition, that the propension for the retention path, is probably emphasized by the use of HF as reactant, and that with other acids, like HC1, or even by assuming the presence of dimers like HF·HF, the inversion path could be preferred. The investigations have been done by determining the geometry of the transition state and the reaction coordinate withab initio SCF STO-3G calculations on the whole nuclear configuration space (21 dimensions). These calculations have been supplemented by a few CI calculations on the same basis set and by a few SCF calculations with a larger basis set.  相似文献
