Alternative methods of reducing oxygen during anaerobic enrichment in the Bacteriological Analytical Manual (BAM) Shigella culture method were evaluated and compared to the current and less practical GasPak method. The alternative anaerobic methods included the use of reducing agents in Shigella broth and reducing culture container headspace volume to minimize atmospheric effects on oxygen concentration in Shigella broth during enrichment. The reducing agents evaluated were sodium thioglycollate, L-cystine, L-cysteine, titanium(III) citrate, and dithiothreitol, each at concentrations of 0.1, 0.05, and 0.01%. The use of Oxyrase for Broth with the enrichment medium (Shigella broth) was evaluated at concentrations of 10, 20 and 30 microL/mL. Recoveries of chill- and freeze-stressed S. sonnei strains 357 and 20143 were determined with each anaerobic method, including the GasPak method, using inoculation levels ranging from 10(0)to 10(3) cells. For each anaerobic method, strain, inoculation level, and stress type, 5 replicate enrichments were evaluated by streaking to MacConkey agar for isolation. The numbers of cultures with each method from which S. sonnei was isolated were used to compare the alternative anaerobic methods to the GasPak method. The alternative anaerobic method with which chill- and freeze-stressed S. sonnei strains 357 and 20143 were isolated most consistently was the use of Oxyrase for Broth in Shigella broth at a concentration of 20 microL/mL. This method was compared to the GasPak anaerobic method in evaluations on the recovery of S. sonnei strains 357 and 20143 from artificially contaminated test portions of parsley, cilantro, green onions, strawberries, carrots, and celery. A third anaerobic method included the use of 0.5 cm mineral oil overlay on cultures containing Oxyrase for Broth at concentrations of 20 microL/mL. Recovery rates of strain 357 were significantly greater (p < 0.05) with the GasPak method than with Oxyrase for Broth, with and without the 0.5 cm mineral oil overlay, for test portions of parsley, cilantro, and celery. When Oxyrase for Broth was used with Shigella broth, strain 357 was isolated at higher rates from all produce types, except cilantro, when 0.5 cm mineral oil overlay was applied to enrichment cultures. The use of mineral oil overlay with Oxyrase for Broth also improved recovery of strain 20143 from test portions of all produce types except green onion and strawberries. These differences were significant (p < 0.05) with parsley, carrots, and cilantro (1 of 2 evaluations). No statistically significant differences (p > 0.05) between the GasPak and Oxyrase for Broth anaerobic methods occurred when mineral oil overlay was used with Oxyrase for Broth. The use of Oxyrase for Broth with a 0.5 cm mineral oil overlay is a practical alternative for anaerobic enrichment with the BAM method in the analysis of some produce types. Differences in recovery among the different produce types and methods occurred between S. sonnei strains 357 and 20143, emphasizing the need for additional S. sonnei strains in future evaluations. 相似文献

In an attempt to obtain the para-f isomer, rac-(1R,4aR,9aR)-2-methyl-1,3,4,9a-tetrahydro-2H-1,4a-propanobenzofuro[2,3-c]pyridin-6-ol, via mesylation of an intermediate 9[small alpha]-hydroxyphenylmorphan, we obtained, instead, a rearranged chloro compound with a 5-membered nitrogen ring, 7-chloro-3a-(2,5-dimethoxyphenyl)-1-methyl-octahydroindole. This indole underwent a second rearrangement to give us the desired para-f isomer. The structures of the intermediate indole and the final product were unequivocally established by X-ray crystallography. A resynthesis of the known rac-(1R,4aR,9aR)-2-methyl-1,3,4,9a-tetrahydro-2H-1,4a-propanobenzofuro[2,3-c]pyridin-8-ol, the ortho-f isomer, was achieved using the reaction conditions for the para-f isomer, as well as under Mitsunobu reaction conditions where, unusually, the oxide-bridge ring in the 5-phenylmorphan was closed to obtain the desired product. The synthesis of the para-f isomer adds an additional compound to those oxide-bridged phenylmorphans that were initially visualized and synthesized; the establishment of the structure and configuration of 8 of the theoretically possible 12 racemates has now been achieved. The X-ray crystallographic structure analysis of the para-f isomer provides essential data that will be needed to establish the configuration of a ligand necessary to interact with an opioid receptor. 相似文献

Constraints on possible Lorentz symmetry violation (LV) of order E/M(Planck) for electrons and photons in the framework of effective field theory (EFT) are discussed. Using (i) the report of polarized MeV emission from GRB021206 and (ii) the absence of vacuum Cerenkov radiation from synchrotron electrons in the Crab Nebula, we improve previous bounds by 10(-10) and 10(-2), respectively. An array of surface science measurements has revealed novel water adsorption behavior at the Fe(3)O(4)(001) surface. Following room temperature exposure to water, a low coverage of hydrogen atoms is observed, with no associated water hydroxyl group. Mild annealing of the hydrogenated surface leads to desorption of water via abstraction of surface oxygen atoms, leading to a reduction of the surface. These results point to an irreversible splitting of the water molecule. The observed phenomena are discussed in the context of recent DFT calculations (Mulakaluri, N.; Pentcheva, R.; Scheffler, M. J. Phys. Chem. C 2010, 114, 11148), which show that the Jahn-Teller distorted surface isolates adsorbed H in a geometry that could kinetically hinder recombinative desorption. In contrast, the adsorption geometry facilitates interaction between water hydroxyl species, which are concluded to leave the surface following a reactive desorption process, possibly via the creation of O(2). 相似文献

Recent studies reveal that amorphous intermediates are involved in the formation of clathrate hydrates under conditions of high driving force, raising two questions: first, how could amorphous nuclei grow into crystalline clathrates and, second, whether amorphous nuclei are intermediates in the formation of clathrate crystals for temperatures close to equilibrium. In this work, we address these two questions through large-scale molecular simulations. We investigate the stability and growth of amorphous and crystalline clathrate nuclei and assess the thermodynamics and kinetic factors that affect the crystallization pathway of clathrates. Our calculations show that the dissociation temperature of amorphous clathrates is just 10% lower than for the crystals, facilitating the formation of metastable amorphous intermediates. We find that, at any temperatures, the critical crystalline nuclei are smaller than critical amorphous nuclei. The temperature dependence of the critical nucleus size is well described by the Gibbs-Thomson relation, from which we extract a liquid-crystal surface tension in excellent agreement with experiments. Our analysis suggests that at high driving force the amorphous nuclei may be kinetically favored over crystalline nuclei because of lower free energy barriers of formation. We investigated the role of the initial structure and size of the nucleus on the subsequent growth of the clathrates, and found that both amorphous and sI crystalline nuclei yield crystalline clathrates. Interestingly, growth of the metastable sII crystal polymorph is always favored over the most stable sI crystal, revealing kinetic control of the growth and indicating that a further step of ripening from sII to sI is needed to reach the stable crystal phase. The latter results are in agreement with the observed metastable formation of sII CO(2) and CH(4) clathrate hydrates and their slow conversion to sI under experimental conditions. 相似文献

The A2A adenosine receptor is a protein belonging to a family of four GPCR adenosine receptors. It is involved in the regulation of several pathophysiological conditions in both the central nervous system and periphery. In the brain, its localization at pre- and postsynaptic level in striatum, cortex, hippocampus and its effects on glutamate release, microglia and astrocyte activation account for a crucial role in neurodegenerative diseases, including Alzheimer’s disease (AD). This ailment is considered the main form of dementia and is expected to exponentially increase in coming years. The pathological tracts of AD include amyloid peptide-β extracellular accumulation and tau hyperphosphorylation, causing neuronal cell death, cognitive deficit, and memory loss. Interestingly, in vitro and in vivo studies have demonstrated that A2A adenosine receptor antagonists may counteract each of these clinical signs, representing an important new strategy to fight a disease for which unfortunately only symptomatic drugs are available. This review offers a brief overview of the biological effects mediated by A2A adenosine receptors in AD animal and human studies and reports the state of the art of A2A adenosine receptor antagonists currently in clinical trials. As an original approach, it focuses on the crucial role of pharmacokinetics and ability to pass the blood–brain barrier in the discovery of new agents for treating CNS disorders. Considering that A2A receptor antagonist istradefylline is already commercially available for Parkinson’s disease treatment, if the proof of concept of these ligands in AD is confirmed and reinforced, it will be easier to offer a new hope for AD patients. 相似文献
