A multicommuted flow system is proposed for spectrophotometric determination of hydrosoluble vitamins (ascorbic acid, thiamine, riboflavine and pyridoxine) in pharmaceutical preparations. The flow manifold was designed with computer-controlled three-way solenoid valves for independent handling of sample and reagent solutions and a multi-channel spectrophotometer was employed for signal measurements. Periodic re-calibration as well as the standard addition method was implemented by using a single reference solution. Linear responses (r=0.999) were obtained for 0.500-10.0 mg l−1 ascorbic acid, 2.00-50.0 mg l−1 thiamine, 5.00-50.0 mg l−1 riboflavine and 0.500-8.00 mg l−1 pyridoxine. Detection limits were estimated as 0.08 mg l−1 (0.5 μmol l−1) ascorbic acid, 0.8 mg l−1 (2 μmol l−1) thiamine, 0.2 mg l−1 (0.5 μmol l−1) riboflavine and 0.1 mg l−1 (0.9 μmol l−1) pyridoxine at 99.7% confidence level. A mean sampling rate of 60 determinations per hour was achieved and coefficients of variation of 1% (n=20) were estimated for all species. The mean reagent consumption was 25-fold lower in relation to flow-based procedures with continuous reagent addition. Average recoveries between 95.6 and 100% were obtained for commercial pharmaceutical preparations. Results agreed with those obtained by reference methods at 95% confidence level. The composition and the antinociceptive activity of the essential oil of Stevia serrata Cav. from a population located in the west highlands of Guatemala were evaluated. A yield of 0.2% (w/w) of essential oil was obtained by hydrodistillation of the dried aerial parts of the plant. The essential oil analysed by GC-FID and GC-MS showed a high content of sesquiterpenoids, with chamazulene (60.1%) as the major component and 91.5% of the essential oil composition was identified. To evaluate antinociceptive activity in mice, the essential oil of S. serrata Cav. was administered as gavage, using three different doses. In the formalin test, the animals were pre-treated with oral doses of the essential oil before the administration of formalin. Oral administration of S. serrata Cav. essential oil produced a marked antinociceptive activity. Therefore, the plant could be domesticated as a source of essential oil rich in chamazulene for developing medicinal products. 相似文献
A compact, reliable and low cost flow injection chemiluminescence system is described. The flow system consists of a set of solenoid micro-pumps that can dispense reproductive micro-volumes of solutions. The luminometer was based on a coiled cell constructed from polyethylene tubing that was sandwiched between two large area photodiodes. The whole equipment costs about US$ 750 and weights ca. 3 kg. Equipment performance was evaluated by measuring low concentrations of hydrogen peroxide by oxidation of luminol and for the determination of ammonium, based on its inhibition of the luminescence provided by the reaction of luminol and sodium hypochlorite. Linear responses were achieved within 1.0-80 μmol L−1 H2O2 and 0.6-60 μmol L−1 NH4+ with detection limits estimated as 400 nmol L−1 H2O2 and 60 nmol L−1 NH4+ at the 99.7% confidence level. Coefficients of variation were 1.0 and 1.8%, estimated for 20 μmol L−1 H2O2 and 15 μmol L−1 NH4+ (n = 20), respectively. Reagent consumption of 55 μg luminol, effluent volume of 950 μL per determination and sampling rate of 120 samples per hour were also achieved. 相似文献
A simple and reliable method for simultaneous electrochemical determination of ascorbic acid (AA) and dopamine (DA) is presented in this work. It was based on the use of the cationic surfactant cetylpyridinium chloride (CPC) that enables the separation of the oxidation peaks potential of AA and DA. Cyclic voltammetry (CV) as well as pulse differential voltammetry (PDV) were used in order to verify the voltammetric behaviour in micellar media. In the cationic surfactant CPC, a remarkable electrostatic interaction is established with negatively charged AA, as a consequence, the oxidation peak potential shifted toward less positive potential and the peak current increased. On the other hand, the positively charged DA is repelled from the electrode surface and the oxidation peak potential shifts toward more positive potential in comparison to the bare electrode. Therefore, the common overlapped oxidation peaks of AA and DA can be circumventing by using CPC. Parameter that affects the Epa and Ipa such as CPC concentration and pH were studied. Under optimised conditions, the method presented a linear response to AA and DA in the concentration range from 5 to 75 μmol L−1 and 10 to 100 μmol L−1, respectively. A multi-syringe flow injection system for the determination of total phenolics based on the 4-aminoantipyrine reaction is proposed. In-line preconcentration using solid-phase extraction was implemented, offering an environmentally friendly alternative to organic solvent extraction performed in the batch procedure and in previously described flow methodologies. Therefore, phenolic compounds were retained in Amberlite XAD4 resin using acidic conditions. The elution was carried out using 0.10molL–1 NaOH solution, promoting the ionisation of the retained phenolic compounds, and thus their desorption.Since different elution profiles were obtained for several phenol derivatives, the calibration curves were established using the peak area as analytical signal. Moreover, a dynamic calibration range was applied by using a variable sample volume (0.500–6.00mL) and calibration curves based on the mass of phenol retained in the pre-concentration column. A methodology based on a fixed sample volume (24.0mL) was also proposed for determination at concentration levels below 50µgL–1. A limit of detection of 52ng of phenol (9 or 2µgL–1) was achieved with a sampling frequency of 10–16h–1 for the variable volume method and 4h–1 for the fixed volume method. The application of the proposed methodology to reference material provided results that were within the certified acceptance limits. 相似文献
Evolution of the commutation concept has lead to the proposal and development of different generations of flow analyzers. Since the inception of the air-segmented flow systems till the availability of modern flow injection, sequential-injection and other flow-based analytical systems, a noteworthy improvement of the commutating devices has been noted.
Multi-functional manifold is described as a polyvalent approach for methodology implementation in a flow analyzer. It permits the investigation of mixing conditions under different flow patterns (unsegmented, segmented, monosegmented) with optional exploitation of the stopped-flow approach. For this purpose, spectrophotometric or turbidimetric measurements eventually affected by Schlieren noise were considered. Potentialities and limitations of the manifold are discussed in relation with methods based on relatively fast or slow chemical reactions. As applications, phosphate and chloride determinations in plant digests and natural waters were selected.
The manifold is characterized by high versatility and may work in connection with different flow configurations. Development will certainly lead to simple, versatile and miniaturized analyzers, able to run samples in a personalized fashion. In addition, random reagent selection, full automation, expansion of the analytical application range and increasing potentialities of the already existing methodologies are devised. 相似文献
Background: The disease caused by hepatitis C virus (HCV) is asymptomatic, silent, and progressive liver disease. In HCV-infected patients the increase in serum HA is associated with the development of hepatic fibrosis and disease progression. Methods: HCV-RNA detection was performed in all serological samples of blood donors that tested positive using HCV Ultra ELISA. Determination of hyaluronan (HA) was performed in positive HCV samples using ELISA-like fluorometric method. The HA content was compared to HCV viral load, genotype of the virus, liver fibrosis as well as ALT and GGT liver biomarkers. Results: Persistently normal ALT (<40 U/L) and GGT (<50 U/L) serum levels were detected in 75% and 69% of the HCV-Infected blood donors, respectively. Based on ROC analysis, the HA value < 34.2 ng/mL is an optimal cut-off point to exclude HCV viremia (specificity = 91%, NPV = 99%). Applying HA value ≥34.2 ng/mL significant liver fibrosis (≥F2) can be estimated in 46% of the HCV-infected blood donors. HA serum level (≥34.2 ng/mL) associated with a high ALT level (>40 U/mL) can correctly identify HCV infection and probable liver fibrosis (sensitivity = 96% and specificity = 90%) in asymptomatic blood donors. Conclusions: A high level of HA (≥34.2 ng/mL) in association with ALT (≥40 U/L) in serum can provide a good clinical opportunity to detect HCV-infected asymptomatic persons that potentially require a liver biopsy confirmation and antiviral treatment to prevent the development of advanced liver fibrosis or cirrhosis. 相似文献
The issue of self-cleaning significantly gained popularity due to the work of Barthlott and coworkers on the so called “Lotos-Effect®”. They found out, that the cleanliness of the Lotos leaves originates from a combined effect of surface topography and hydrophobicity. The symbol of the beautiful Lotos flower as well as the fascination of surfaces being cleaned without any manual activity, simply by a rain shower, has since then stimulated the fantasy of many researchers. Our vision is to copy this mechanism from mother nature and to implement it into coating systems in such a way, that conventional application techniques, e.g. spray-coating, can be applied without the necessity of further process steps like e.g. soft lithography. Three different approaches will be presented in this paper. Roughness and contact angle measurements have been used to quantify the self-cleaning properties. 相似文献
