Preparation of Secondary Carbinamines via Alkylation of N-Boryl Imines Generated from Partial Reduction of Nitriles with NABH4-Carboxylic Acid

Secondary carbinamines have been prepared from alkylation of N-boryl imines which were generated in situ from partial reduction of nitriles with sodium brohydride modified by carboxylic acid.     

Engineering Plant Alternative Oxidase Function in Mammalian Cells: Substitution of the Motif-like Sequence ENV for QDT Diminishes Catalytic Activity of Arum concinnatum AOX1a Expressed in HeLa Cells

Alternative oxidase (AOX) is a nonproton motive quinol–oxygen oxidoreductase which is a component of the mitochondrial respiratory chain in higher plants. In this study, we have characterized the catalytic activity and regulatory behaviors of Arum concinnatum AOX isoforms, namely AcoAOX1a and AcoAOX1b, and their artificial mutants in HeLa cells. We demonstrated that substitution of the motif-like sequence ENV on the C-terminal half of AcoAOX1a for QDT diminishes its activity and proposed that the innate inactivity of AcoAOX1b in HeLa cells is, at least in part, attributable to its QDT motif. Furthermore, we show that introduction of F130L in the hydrophilic N-terminal extension of AcoAOX1a resulted in greater activity in the presence of pyruvate. This result indicates that functional significance of the N-terminal extension is not particular to the conventional regulatory cysteine. On the basis of these findings, we discuss new insights into the structural integrity of AOX in HeLa cells and the applicability of mammalian cells for functional analysis of this enzyme.     

SYNTHESIS AND CHARACTERIZATION OF KINETICALLY STABILIZED 1,3-DIPHOSPHACYCLOBUTENE DERIVATIVES

1-(2,4,6-Tri-tert-butylphenyl)-2-phosphaethyne (1) was allowed to react with 0.5 equiv of an alkyllithium and subsequently with an alcohol to afford a bulky 1,3-diphosphacyclobutene, and its structure and coordination properties on transition metals were investigated. On the other hand, 1 was allowed to react with an alkyllithium and iodomethane to form a stable biradical, 1,3-diphosphacyclobutane-2,4-diyl.     

Synthesis of All Stereoisomers of Monomeric Spectomycin A1/A2 and Evaluation of Their Protein SUMOylation-Inhibitory Activity

A synthetic methodology to access all possible stereoisomers of spectomycin A1 (SMA1) and A2 (SMA2) has been established through late-stage diversification. The key reaction for the construction of all four diastereomers is an intramolecular cyclization based on the umpolung of π-allyl palladium species with bis(pinacolato)diborane (B₂(pin)₂). Silyl group assisted direct benzylic oxidation of each isomer enabled construction of the fragile β-hydroxytetralone skeleton to provide the SMAs. The relative and absolute stereochemistry of SMA2 was also determined, and the absolute stereochemistry of SMA1 was extrapolated based on the optical rotation of SMA2. The axial chirality of SMAs is discussed based on circular dichroism spectra and DFT calculations, and it is concluded that the M isomer is predominant in solution. Biochemical assessment of all isomers in vitro revealed that the C9 hydroxyl group and dimeric structure were both important for protein SUMOylation-inhibitory activity.     

New heterocyclic metallomesogens: synthesis,mesomorphic and thermal behaviours of Cu(II) complexes with 1,2,3-triazole-based Schiff bases ligands

Two series of new Cu(II) complexes derived from the reaction of copper acetate with the non-linear 1,2,3-triazole-based Schiff bases have successfully been synthesised. The structures of the ligands and its complexes were elucidated by elemental analysis, FT-IR, ¹H-NMR and UV–visible spectroscopic techniques. The differential scanning calorimetry and polarizing optical microscopy supported the anisotropic properties of uncoordinated ligands in which the focal conic fan-shaped texture and/or broken fan-shaped texture characteristics of respective SmA and SmC phases were recorded. However, not all of their corresponding Cu(II) complexes are mesogenic. Although the iodo-substituted ligands with even parity C₁₀H₂₉ to C₁₄H₃₃ are non-stable and exhibit SmA phase which is not reproducible, the ultimate Cu(II) complexes show exclusively stable SmA phase. This observation can be ascribed to the enhanced colinearity and molecular anisotropic by the presence of Cu-N and Cu-O coordination modes. On the other hand, the comparison studies show that different positions of ortho-hydroxyl group affect the mesomorphic and thermal behaviour of ligands and Cu(II) complexes.     

Total synthesis of pleurolactone

The stereoselective total synthesis of pleurolactone has been accomplished in 7 steps. The key synthetic features were the construction of four contiguous stereocenters using an endo-selective Diels-Alder reaction and high diastereoselective dihydroxylation.     

Palladium complexes bearing an N‐heterocyclic carbene–sulfonamide ligand for cooligomerization of ethylene and polar monomers

Herein, we report the synthesis of palladium complexes bearing an N‐heterocyclic carbene (NHC)‐sulfonamide bidentate ligand and their application in ethylene oligomerization and ethylene/polar monomer cooligomerization. These catalysts could smoothly catalyze ethylene oligomerization and ethylene/methyl acrylate cooligomerization albeit the performance was lower compared to that of a NHC–phenoxide catalyst. © 2018 Wiley Periodicals, Inc.
