Recently, linear programming problems with special structures have assumed growing importance in mathematical programming.
It is well known that exploiting network structures within linear programs can lead to considerable improvement of the computational
solution of large-scale linear programming problems. A linear program is said to contain an embedded network structure provided
that some subset of its constraints can be interpreted as specifying conservation of flow. If a column of the constraint matrix
has at most two non-zeros, then it leads to embedded generalized network structure and if these non-zeros are unit elements
and of opposite signs, then it leads to embedded pure network structure.
In this paper, we are concerned with algorithms for detecting embedded pure network structures within linear programs. The
network extraction methods are presented in two groups. The first group covers deletion and addition based algorithms and
the second group covers GUB based algorithms. We have extended the GUB based algorithm appearing in the second group by introducing
Markowitz merit count approach for exploiting matrix non zeros. A set of well known test problems has been used to carry out
computational experiments which show that our extensions to the GUB based algorithms give better results than the algorithms

Mets motifs, which refer to methionine-rich sequences found in the high-affinity copper transporter Ctr1, also appear in other proteins involved in copper trafficking and homeostasis, including other Ctrs as well as Pco and Cop proteins isolated from copper-resistant bacteria. To understand the coordination chemistry utilized by these proteins, we studied the copper binding properties of a peptide labeled Mets7-PcoC with the sequence Met-Thr-Gly-Met-Lys-Gly-Met-Ser. By comparing this sequence to a series of mutants containing noncoordinating norleucine in place of methionine, we confirm that all three methionine residues are involved in a thioether-only binding site that is selective for Cu(I). Two independent methods, one based on mass spectrometry and one based on rate differences for the copper-catalyzed oxidation of ascorbic acid, provide an effective K(D) of approximately 2.5 microM at pH 4.5 for the 1:1 complex of Mets7-PcoC with Cu(I). These results establish that a relatively simple peptide containing an MX(2)MX(2)M motif is sufficient to bind Cu(I) with an affinity that corresponds well with its proposed biological function of extracellular copper acquisition. 相似文献

This article describes the development and full characterization of a microfluidic chip for electrofusion of human peripheral blood B-cells and mouse myeloma (NS-1) cells to generate hybridomas. The chip consists of an array of 783 traps, with dimensions that were optimized to obtain a final cell pairing efficiency of 33±6%. B cells were stained with a cytoplasmic stain CFDA to assess the different stages of cell fusion, i.e. dye transfer to NS-1 cells (initiating fusion) and membrane reorganization (advanced fusion). Six DC pulses of 100 μs (2.5 kV/cm) combined with an AC field (30 s, 2 MHz, 500 V/cm) and pronase treatment resulted in the highest electrofusion efficiency of paired cells (51±11%). Hybridoma formation, with a yield of 0.33 and 1.2%, was observed after culturing the fused cells for 14 days in conditioned medium. This work provides valuable leads to improve the current electrofusion protocols for the production of human antibodies for diagnostic and therapeutic applications. 相似文献

The possibilities of increasing the livingness in cationic isobutylene polymerizations has been investigated. It has been assumed that the living chain-end is polarized but non-dissociated and relatively stable, therefore, i) shifting of the dissociation equilibrium by adding salts with common or non-common anion in excess, and ii) increasing of the stability of the chain-end by complexing it with electrondonors, may increase the livingness of the system. Indeed, the addition of n-Bu4NCl or n-Bu4NI to systems with BCl3 or TiCl4 coinitiators, as well as the addition of dimethyl sulphoxide as electrondonor in a broad concentration range, improved the living character.
