High molecular weight hyaluronic acid (HA) is present in articular joints and synovial fluid at high concentrations; yet despite numerous studies, the role of HA in joint lubrication is still not clear. Free HA in solution does not appear to be a good lubricant, being negatively charged and therefore repelled from most biological, including cartilage, surfaces. Recent enzymatic experiments suggested that mechanically or physically (rather than chemically) trapped HA could function as an "adaptive" or "emergency" boundary lubricant to eliminate wear damage in shearing cartilage surfaces. In this work, HA was chemically grafted to a layer of self-assembled amino-propyl-triethoxy-silane (APTES) on mica and then cross-linked. The boundary lubrication behavior of APTES and of chemically grafted and cross-linked HA in both electrolyte and lipid 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) solutions was tested with a surface forces apparatus (SFA). Despite the high coefficient of friction (COF) of μ ≈ 0.50, the chemically grafted HA gel significantly improved the lubrication behavior of HA, particularly the wear resistance, in comparison to free HA. Adding more DOPC lipid to the solution did not improve the lubrication of the chemically grafted and cross-linked HA layer. Damage of the underlying mica surface became visible at higher loads (pressure >2 MPa) after prolonged sliding times. It has generally been assumed that damage caused by or during sliding, also known as "abrasive friction", which is the main biomedical/clinical/morphological manifestation of arthritis, is due to a high friction force and, therefore, a large COF, and that to prevent surface damage or wear (abrasion) one should therefore aim to reduce the COF, which has been the traditional focus of basic research in biolubrication, particularly in cartilage and joint lubrication. Here we combine our results with previous ones on grafted and cross-linked HA on lipid bilayers, and lubricin-mediated lubrication, and conclude that for cartilage surfaces, a high COF can be associated with good wear protection, while a low COF can have poor wear resistance. Both of these properties depend on how the lubricating molecules are attached to and organized at the surfaces, as well as the structure and mechanical, viscoelastic, elastic, and physical properties of the surfaces, but the two phenomena are not directly or simply related. We also conclude that to provide both the low COF and good wear protection of joints under physiological conditions, some or all of the four major components of joints-HA, lubricin, lipids, and the cartilage fibrils-must act synergistically in ways (physisorbed, chemisorbed, grafted and/or cross-linked) that are still to be determined.  相似文献

We have investigated the effects of humidity (water content or activity from 0 to approximately 0.98) on the frictional properties of surfactant-coated ZnS nanoparticles of various shapes, specifically, nanorods and nanowires, dispersed in an organic solvent (dodecane). The friction coefficients were found to be sensitive to even trace amounts of water, increasing logarithmically with time after the systems were exposed to humid air, doubling after 2-4 h of exposure time to air of relative humidity approximately 98%. We also show that increasing the humidity caused noticeable effects on the interactions of the nanoparticles, increasing their adhesion and aggregation through capillary forces. These effects should be considered in the design of organic solvents containing nanoparticles with physisorbed surfactants, for example, lube oils with nanoparticles additives, particularly those exposed to atmospheric conditions.  相似文献

Using a surface forces apparatus, we have investigated the adhesive and lubrication forces of mica surfaces separated by a molecularly thin, subnanometer film of a high-molecular-weight (2.3 MDa) anionic polysaccharide from the algae Porphyridium sp. adsorbed from aqueous solution. The adhesion and friction forces of the confined biopolymer were monitored as a function of time, shearing distance, and driving velocity under a large range of compressive loads (pressures). Although the thickness of the dilute polysaccharide was <1 nm, the friction was low (coefficient of friction = 0.015), and no wear was ever observed even at a pressure of 110 atm over 3 decades of velocity, so long as the shearing distances were less than twice the contact diameter. Atomic force microscopy in solution shows that the biopolymer is able to adsorb to the mica surface but remains mobile and easily dragged upon shearing. The adhesion (adsorption) of this polysaccharide even to negatively charged surfaces, its stable low friction, its robustness (high-load carrying capacity and good wear protection), and the weak (logarithmic) dependence of the friction force on the sliding velocity make this class of polyelectrolytes excellent candidates for use in water-based lubricant fluids and as potential additives to synovial fluid in joints and other biolubricating fluids. The physical reasons for the remarkable tribological properties of the ultrathin polysaccharide monolayer are discussed and appear to be quite different from those of other polyelectrolytes and proteins that act as thick "polymer brush" layers.  相似文献

The extraordinary climbing ability of geckos is partially attributed to the fine structure of their toe pads, which contain arrays consisting of thousands of micrometer-sized stalks (setae) that are in turn terminated by millions of fingerlike pads (spatulae) having nanoscale dimensions. Using a surface forces apparatus (SFA), we have investigated the dynamic sliding characteristics of setal arrays subjected to various loading, unloading, and shearing conditions at different angles. Setal arrays were glued onto silica substrates and, once installed into the SFA, brought toward a polymeric substrate surface and then sheared. Lateral shearing of the arrays was initiated along both the "gripping" and "releasing" directions of the setae on the foot pads. We find that the anisotropic microstructure of the setal arrays gives rise to quite different adhesive and tribological properties when sliding along these two directions, depending also on the angle that the setae subtend with respect to the surface. Thus, dragging the setal arrays along the gripping direction leads to strong adhesion and friction forces (as required during contact and attachment), whereas when shearing along the releasing direction, both forces fall to almost zero (as desired during rapid detachment). The results and analysis provide new insights into the biomechanics of adhesion and friction forces in animals, the coupling between these two forces, and the specialized structures that allow them to optimize these forces along different directions during movement. Our results also have practical implications and criteria for designing reversible and responsive adhesives and articulated robotic mechanisms.  相似文献

A new device has been designed, and a prototype built and tested, that can simultaneously measure the displacements and/or the components of a force in three orthogonal directions. The "3D sensor" consists of four or eight strain gauges attached to the four arms of a single cross-shaped force-measuring cantilever spring. Finite element modeling (FEM) was performed to optimize the design configuration to give desired sensitivity of force, displacement, stiffness, and resonant frequency in each direction (x, y, and z) which were tested on a "mesoscale" device and found to agree with the predicted values to within 4-10%. The device can be fitted into a surface forces apparatus (SFA), and a future smaller "microscale" microfabricated version can be fitted into an atomic force microscope (AFM) for simultaneous measurements of the normal and lateral (friction) forces between a tip (or colloidal bead probe) and a surface, and the topography of the surface. Results of the FEM analysis are presented, and approximate equations derived using linear elasticity theory are given for the sensitivity in each direction. Initial calibrations and measurements of thin film rheology (lubrication forces) using the "mesoscale" prototype show the device to function as expected.  相似文献

We present a newly designed electrochemical surface forces apparatus (EC-SFA) that allows control and measurement of surface potentials and interfacial electrochemical reactions with simultaneous measurement of normal interaction forces (with nN resolution), friction forces (with μN resolution), and distances (with ? resolution) between apposing surfaces. We describe three applications of the developed EC-SFA and discuss the wide-range of potential other applications. In particular, we describe measurements of (1) force-distance profiles between smooth and rough gold surfaces and apposing self-assembled monolayer-covered smooth mica surfaces; (2) the effective changing thickness of anodically growing oxide layers with ?-accuracy on rough and smooth surfaces; and (3) friction forces evolving at a metal-ceramic contact, all as a function of the applied electrochemical potential. Interaction forces between atomically smooth surfaces are well-described using DLVO theory and the Hogg-Healy-Fuerstenau approximation for electric double layer interactions between dissimilar surfaces, which unintuitively predicts the possibility of attractive double layer forces between dissimilar surfaces whose surface potentials have similar sign, and repulsive forces between surfaces whose surface potentials have opposite sign. Surface roughness of the gold electrodes leads to an additional exponentially repulsive force in the force-distance profiles that is qualitatively well described by an extended DLVO model that includes repulsive hydration and steric forces. Comparing the measured thickness of the anodic gold oxide layer and the charge consumed for generating this layer allowed the identification of its chemical structure as a hydrated Au(OH)(3) phase formed at the gold surface at high positive potentials. The EC-SFA allows, for the first time, one to look at complex long-term transient effects of dynamic processes (e.g., relaxation times), which are also reflected in friction forces while tuning electrochemical surface potentials.  相似文献

Using a surface forces apparatus (SFA) and an atomic force microscope (AFM) we have studied the effects of surface roughness (root-mean-square (RMS) roughness between 0.3 and 220 nm) on the "contact mechanics", which describes the deformations and loading and unloading adhesion forces, of various polymeric surfaces. For randomly rough, moderately stiff, elastomeric surfaces, the force-distance curves on approach and separation are nearly reversible and almost perfectly exponentially repulsive, with an adhesion on separation that decreases only slightly with increasing RMS. Additionally, the magnitude of the preload force is seen to play a large role in determining the measured adhesion. The exponential repulsion likely arises from the local compressions (fine-grained nano- or submicron-scale deformations) of the surface asperities. The resulting characteristic decay lengths of the repulsion scale with the RMS roughness and correlate very well with a simple finite element method (FEM) analysis based on actual AFM topographical images of the surfaces. For "patterned" surfaces, with a nonrandom terraced structure, no similar exponential repulsion is observed, suggesting that asperity height variability or random roughness is required for the exponential behavior. However, the adhesion force or energy between two "patterned" surfaces fell off dramatically and roughly exponentially as the RMS increased, likely owing to a significant decrease in the contact area which in turn determines their adhesion. For both types of rough surfaces, random and patterned, the coarse-grained (global, meso- or macroscopic) deformations of the initially curved surfaces appear to be Hertzian.  相似文献

Geckos have developed a unique hierarchical structure to maintain climbing ability on surfaces with different roughness, one of the extremely important parameters that affect the friction and adhesion forces between two surfaces. Although much attention has been paid on fabricating various structures that mimic the hierarchical structure of a gecko foot, yet no systematic effort, in experiment or theory, has been made to quantify the effect of surface roughness on the performance of the fabricated structures that mimic the hierarchical structure of geckos. Using a modified surface forces apparatus (SFA), we measured the adhesion and friction forces between microfabricated tilted PDMS flaps and optically smooth SiO(2) and rough SiO(2) surfaces created by plasma etching. Anisotropic adhesion and friction forces were measured when sliding the top glass surface along (+y) and against (-y) the tilted direction of the flaps. Increasing the surface roughness first increased the adhesion and friction forces measured between the flaps and the rough surface due to topological matching of the two surfaces but then led to a rapid decrease in both of these forces. Our results demonstrate that the surface roughness significantly affects the performance of gecko mimetic adhesives and that different surface textures can either increase or decrease the adhesion and friction forces of the fabricated adhesives.  相似文献

Abstract

A novel series pyrrolo[3,2-b]pyridine-3-carboxamide linked 2-methoxypyridine derivatives have been designed, synthesized and confirmed by FT-IR, 1H NMR, 13C NMR, 19F NMR, MS, and elemental analysis. The synthesized compounds were screened for their antitubercular activity using microplate alamar blue assay method and antibacterial activity. Among the tested compounds, 4- fluorophenyl (8m), 4- chlorophenyl (8n) and 4-methoxyphenyl (8i) showed potent anti-TB activity (3.12?µg/mL) in comparison with reference drug, Pyrazinamide ((3.12?µg/mL). In addition, all compounds were docked into DprE1 (PDB code: 4KW5) to explore their binding interactions at the active site. The compounds exhibited essential key interactions as that of reported DprE1 inhibitors and hence, the synthesized compounds may be considered as molecular scaffolds for antitubercular activity. Compounds, 4-chlorophenyl (8n) and 4-flurophenyl (8m) showed significant antibacterial activity against Escherichia coli and Staphylococcus aureus strains. In silico prediction of toxicities, druglikeness and drug score profiles of the tested compounds are promising.  相似文献
