Polymeric micelles (Lactosome) in the size of 20–30 nm were labeled with radionuclides of 111In (111In-DOTA-Lactosome) for SPECT imaging and 90Y (90Y-DOTA-Lactosome) for β-ray irradiation for mammary tumor in mice. The tumor site at the femoral right leg grafted with 4T1 cells was clearly imaged at 24 h after the intravenous injection. Biodistribution revealed that the half-life time of 111In-DOTA-Lactosome was 11 h, which enabled the nanoparticle selectively accumulated in tumor site due to the enhanced permeability and retention (EPR) effect. The anti-tumor therapeutic effect of 90Y-DOTA-Lactosome was observed depending on the dose frequency and amount. Under the condition of the percutaneous ethanol injection treatment, the therapeutic effect of 90Y-DOTA-Lactosome was enhanced due to the super EPR effect. Owing to the super EPR effect, co-administration of 90Y-DOTA-Lactosome and DOXIL® inhibited the tumor growth during 15 days with their administrations.  相似文献

4-Oxo-4-H-1-benzopyran-3-carboxylic acids and their derivatives (esters and amides), 2, are known to possess interesting pharmacological activities.1 As part of our synthetic studies of biologically active compounds, we needed to prepare a variety of 2 (carboxylic acids, esters, amides, etc.). An elegant method for the preparation of 4-oxo-4H-1-benzopyran-3-carboxaldehydes, 1, was reported independently by Nohara et al. 2 and Harnish.3 However, despite a rather extensive effort by Nohara et al., the oxidation of 1 to 2 (Y=OH) has not been realized in good yield; thus the yields by Jones oxidation, the best reported method, ranged only from 9.5 to 39%.4  相似文献

We synthesized porphyrinylamide 4b and its N-methylated derivative 5b. Direct N-methylation of porphyrinylamides 4 proved unsuccessful, so 5b was obtained via N-methylation of 5-aminoporphyrin 10 with potassium hexamethyldisilazide. The secondary amide 4b exists in trans-amide form, while N-methylated amide 5b exists in the cis-amide form, both in solution and in the crystal. Thus, N-methylation of the amide bond of 4b resulted in trans to cis conformational alteration.  相似文献

Shimalactones A and B are neuritogenic polyketides possessing characteristic oxabicyclo[2.2.1]heptane and bicyclo[4.2.0]octadiene ring systems that are produced by the marine fungus Emericella variecolor GF10. We identified a candidate biosynthetic gene cluster and conducted heterologous expression analysis. Expression of ShmA polyketide synthase in Aspergillus oryzae resulted in the production of preshimalactone. Aspergillus oryzae and Saccharomyces cerevisiae transformants expressing ShmA and ShmB produced shimalactones A and B, thus suggesting that the double bicyclo‐ring formation reactions proceed non‐enzymatically from preshimalactone epoxide. DFT calculations strongly support the idea that oxabicyclo‐ring formation and 8π‐6π electrocyclization proceed spontaneously after opening of the preshimalactone epoxide ring through protonation. We confirmed the formation of preshimalactone epoxide in vitro, followed by its non‐enzymatic conversion to shimalactones in the dark.  相似文献

A convergent total synthesis of (?)-dactylolide is described. Constructing the 2,6-disubstituted exo-methylene THP moiety was achieved by the intramolecular allylation of α-acetoxy ether. The cyclization precursor was prepared from two segments, an alcohol and carboxylic acid derivatives, by esterification followed by reductive acetylation.  相似文献

The muon spin rotation ( +SR) and NMR measurements provides clear evidences of the antiferromagnetic order of Cu-moment below 35 K for La2–x Ba x CuO4 and below 15 K for La2–x Sr x CuO4 in the narrow range ofx where the high-T c superconductivity (SC) is suppressed remarkably. The results suggest that the freezing of spin fluctuations of Cu-moment is relevant to the local suppression of SC under an change of the electronic state coupled with the lattice instability.  相似文献

Film formation by ionized-cluster beam epitaxy is characterized by various processes including sputtering, ion implantation, substrate heating, and migration effects. These processes can be utilized to form good crystalline epitaxial films. The film formation mechanism has been studied from the view point of atomistic model and it is found to be different from that observed with the other methods. Some properties of Si and ZnO are shown as typical examples of the standard type and the reactive type of ionized cluster beam epitaxy.  相似文献

Temporal variations in N concentration and δ15N value of annual tree rings (1 year of time resolution) of two Japanese Black Pine (Pinus thunbergii) and three Japanese Red Pine (Pinus densiflora) trees under current breeding activity of the Great Cormorant (Pharacrocorax carbo) and the Black-tailed Gull (Larus crassirostris), respectively, in central and northeastern Japan were studied. Both species from control sites where no avian input occurs show negative values (δ15N = around?4 ‰ to?2 ‰) which are common among higher plants growing under high rainfall regimes. The δ15N values of P. densiflora show uniformly positive values several years before and after the breeding event, indicating N translocation that moved the absorbed N of a given growth year to tree rings of the previous year while a clear historical value of soil N dynamics was kept intact in the annual rings of P. thunbergii. Long-term N trends inferred from tree rings must take into account tree species with limited translocation rates that can retain actual N annual acquisition.  相似文献
