In searching for tyrosinase inhibitors from plants using L-3,4-dihydroxyphenylalanine (L-DOPA) as a substrate, quercetin was found to be partially oxidized to the corresponding o-quinone under catalysis by mushroom tyrosinase (EC 1.14.18.1). Simultaneously, L-DOPA was also oxidized to dopaquinone and both o-quinones were further oxidized, respectively. The remaining quercetin partially formed adducts with dopaquinone through a Michael type addition. In general, flavonols form adducts with dopaquinone as long as their 3-hydroxyl group is free. Quercetin enhanced melanin production per cell in cultured murine B16-F10 melanoma cells, but this effect may be due in part to melanocytotoxicity. The concentration leading to 50% viable cells lost was established as 20 microM and almost complete lethality was observed at 80 microM.  相似文献

Voice instabilities were studied using excised human larynx experiments and biomechanical modeling. With a controlled elongation of the vocal folds, the experiments showed registers with chest-like and falsetto-like vibrations. Observed instabilities included abrupt jumps between the two registers exhibiting hysteresis, aphonic episodes, subharmonics, and chaos near the register transitions. In order to model these phenomena, a three-mass model was constructed by adding a third mass on top of the simplified two-mass model. Simulation studies showed that the three-mass model can vibrate in both chest-like and falsetto-like patterns. Variation of tension parameters which mimic activities of laryngeal muscles could induce transitions between both registers. For reduced prephonatory areas and damping constants, extended coexistence of chest and falsetto registers was found, in agreement with experimental data. Subharmonics and deterministic chaos were observed close to transitions between the registers. It is concluded that the abrupt changes between chest and falsetto registers can be understood as shifts in dominance of eigenmodes of the vocal folds.  相似文献

Poly(butylene succinate) (PBS) and PBS-based ionomers (PBSi) with 1.0 and 3.0 mol% sodium sulfonate ionic group were synthesized and blended with poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) (PHB-HHx) via direct melt compounding. FT-IR analysis demonstrated that the sodium metal ion–carbonyl interaction between PHB-HHx and PBS ionomer increased in strength with the ionic group concentration. Both non-isothermal and isothermal analyses showed the crystallization rates of PHB-HHx to decrease as the strength of the sodium metal ion–carbonyl interaction increased. However, the constant value obtained for the Avrami exponent indicated that the presence of PBS ionomer did not interfere in any way with the nucleation mechanism or the geometry of the crystal growth of PHB-HHx. DMTA analysis confirmed that PBS ionomer reduced the crystallinity of PHB-HHx, and this phenomenon increased in proportion to the ionic group content. As the ionic group concentration increased, the sodium metal–carbonyl interaction between PHB-HHx and PBS ionomer became much stronger, resulting in the improvement of the miscibility for the blend. The interaction parameter obtained by analyzing the equilibrium melting temperature was negative for all bend systems, with the ionomer having ionic group content, displaying a more negative value. Based on the Lauritzen–Hoffman secondary nucleation theory, the regime of the PHB-HHx/PBS ionomer blend remained unchanged throughout the crystallization process. In addition, both the nucleation constant and surface free energy were found to decrease as both ionomer content and ionic group concentration increased.  相似文献

Poly(methyl methacrylate) (PMMA)/zinc oxide (ZnO) or carbazole polymer (PCEM)/ZnO nanocomposites, which are composed of high molecular weight PMMA or PCEM with narrow molecular weight distributions and ZnO nanoparticles, were successfully prepared by atom transfer radical polymerization (ATRP) initiated by 2-bromo-2-methylpropionyl (BMP) group (ZnBM) introduced onto the ZnO nanoparticle surfaces. Introduction of the BMP group onto the ZnO surfaces was achieved by esterification of OH group of the ZnO surfaces. The chemically attached OH group-having ZnO nanoparticles (ZnHM) were fabricated by sol-gel reaction from zinc acetate dihydrate, followed by treatment of the ZnO nanoparticles with 2-hydroxypropionic acid (HPA). The ZnHM nanoparticles showed one UV absorption and two emission bands: UV emission peak and broad visible emission band, while the ZnBM exhibited broad UV absorption and no emission spectra. The PMMA/ZnO nanocomposites displayed UV absorption and photoluminescent (PL) band with blue emission on the basis of the ZnHM nanoparticles, where the ZnO nanoparticles dispersed homogeneously in the PMMA matrix. The PCEM/ZnO nanocomposites depicted UV emission peak due to the carbazole unit in the UV range, but no visible emission. Thermal properties of the PMMA/ZnO nanocomposites were improved by dispersion of the ZnO nanoparticles into the PMMA, but the PCEM/ZnO nanocomposites showed no improvement of the thermal properties.  相似文献

Concentration-resolved 2D correlation gel permeation chromatography (GPC) has been used to examine the intricate details of the HCl-catalyzed polymerization of a polymeric silane-coupling agent (SCA), triethoxysilyl-terminated polystyrene (TESiPS). The concentration-resolved 2D correlation GPC maps directly reflect the marked difference in the aggregate-aggregate interactions of dilute and concentrated monomeric units, which govern the differences in the polymerized products. There is an optimum concentration of SCA for the enhancement of interfacial strength and subsequent polymerization. Thus, the concentration-resolved 2D correlation GPC technique can be used as a powerful tool for elucidation of aggregate-aggregate interactions and reaction mechanisms in a surface- or interface-enhanced reaction system. It has been shown that the yield value of polymerization products can be improved to a marked extent by choosing a high initial monomer concentration, due to the increase in the production of oligomers. Multiple reaction processes are promoted by the self-assembly of the monomeric and oligomeric components.  相似文献

Dealuminated zeolite Y was used as a crystalline support for a mononuclear ruthenium complex synthesized from cis-Ru(acac)2(C2H4)2. Infrared (IR) and extended X-ray absorption fine structure spectra indicated that the surface species were mononuclear ruthenium complexes, Ru(acac)(C2H4)2(2+), tightly bonded to the surface by two Ru-O bonds at Al(3+) sites of the zeolite. The maximum loading of the anchored ruthenium complexes was one complex per two Al(3+) sites; at higher loadings, some of the cis-Ru(acac)2(C2H4)2 was physisorbed. In the presence of ethylene and H2, the surface-bound species entered into a catalytic cycle for ethylene dimerization and operated stably. IR data showed that at the start of the catalytic reaction, the acac ligand of the Ru(acac)(C2H4)2(2+) species was dissociated and captured by an Al(3+) site. Ethylene dimerization proceeded approximately 600 times faster with a cofeed of ethylene and H2 than without H2. These results provide evidence of the importance of the cooperation of the Al(3+) sites in the zeolite and the H2 in the feed for the genesis of the catalytically active species. The results presented here demonstrate the usefulness of dealuminated zeolite Y as a nearly uniform support that allows precise synthesis of supported catalysts and detailed elucidation of their structures.  相似文献

Solid acid supports such as H-ZSM5, H-Mordenite, γ-Al2O3, USY and Beta catalysts were modified with Pt. These Pt/oxide catalysts were found to be active for propane formation through aqueous reforming of glycerol in the presence of hydrogen. The reforming reactions, possibly, proceeded through reaction cycles of dehydration on acid sites and hydrogenation on Pt sites over the catalysts.  相似文献

Gastric cancer is the most common cancer in Japan and infection with Epstein-Barr virus (EBV) is responsible for about 10% of gastric cancers worldwide. Although EBV infection may be involved at an early stage of gastric carcinogenesis, the mechanisms underlying its involvement remain unknown. To investigate the role of EBV in gastric carcinogenesis, we performed proteomic analyses of an EBV-infected gastric carcinoma cell line NU-GC-3 (EBV(+)) and its uninfected control (EBV(-)). 2-DE was combined with MS to identify differentially expressed proteins. We found that EBV infection upregulated one of the phosphorylated heat shock protein 27 kDa (HSP27). The phosphorylated HSP27 isoform which increased in EBV(+) cells can be induced by both heat shock and arsenite. The increase of phosphorylated HSP27 in EBV(+) cells was reduced by treatment with the phosphoinositide 3-kinase (PI3K) inhibitors (LY294002 and wortmannin). In addition, we found increased levels of phosphorylated Akt in EBV(+) cells. These findings suggest that EBV infection upregulates the phosphorylation of HSP27 via the PI3K/Akt pathway. Thus, activation of the PI3K/Akt pathway may contribute to the establishment of a malignant phenotype in EBV-infected gastric carcinomas.  相似文献

We have found that alcohols, carboxylic acids, and amides self-assemble into a unique molecular architecture, a hydrogen-bonded molecular macrocluster, when they are selectively adsorbed onto silica (glass and oxidized silicon) surfaces in nonpolar solvents such as cyclohexane. In our previous study, this phenomenon could be successfully applied to fabricate molecularly flat and defect-free nanofilms of several tens of nanometers thickness. In this study, we prepared a poly(N-isopropylacrylamide) [poly(NIPAAm)] film on the basis of in situ polymerization of a monomer macrocluster layer formed on silica surfaces and investigated how the molecular arrangement of the adsorbed NIPAAm monomers affects the efficiency of the polymerization of them. Poly(NIPAAm) films were prepared by the following two methods: (1) the one-solution method, the in situ photopolymerization of an NIPAAm monomer adsorption layer on silica in one solution (chloroform, cyclohexane, and toluene), and (2) the solution exchange method, adsorption of NIPAAm monomers onto a silica surface from NIPAAm (0.1 mol %) in chloroform, exhange of the solution to 0.005 mol % NIPAAm in cyclohexane, and then polymerization by UV irradiation. By the solution exchange method, molecularly flat, defect-free, and thermoresponsive films were obtained and the thickness could be controlled by the irradiation time, while only several nanometers thickness could be attained by the one-solution method. The structure of NIPAAm adsorption layers formed in each solution condition was characterized by attenuated total reflection Fourier transform infrared spectroscopy. It was revealed that only the solution exchange procedure induced the beta-sheet-like adsorbed structure of NIPAAm in which the double bonds of neighboring NIPAAm monomers were closely located, which should have resulted in effective polymerization.  相似文献
