Cooperativity between weak hydrogen bonds can be revealed in molecular clusters isolated in the gas phase. Here we examine the structure, internal dynamics, and origin of the weak intermolecular forces between sevoflurane and a benzene molecule, using multi‐isotopic broadband rotational spectra. This heterodimer is held together by a primary C? H???π hydrogen bond, assisted by multiple weak C? H???F interactions. The multiple nonbonding forces hinder the internal rotation of benzene around the isopropyl C? H bond in sevoflurane, producing detectable quantum tunneling effects in the rotational spectrum.  相似文献

Three new psychollatine-derived monoterpene indole alkaloids were obtained from Psychotria umbellata Thonn.: 3,4-Dehydro-18,19-β-epoxy-psychollatine (2), N4-[1-((R)-2-hydroxypropyl)]-psychollatine (3), and N4-[1-((S)-2-hydroxypropyl)]-psychollatine (4). Their structures were determined by 1H and 13C NMR spectra, 2D correlations (COSY, HMQC, and HMBC), and mass and UV spectra. Compounds 3 and 4 were synthesized for structural confirmation and for the determination of the stereochemistry of the hydroxyl group.  相似文献

The recent study focusing on paper coated with microfibrillated cellulose (MFC) revealed the ability of such a structure to achieve a controlled release of molecules introduced into its nanoporous network. The present study examines this concept using a chlorhexidine digluconate-based (CHX) antibacterial solution. Various analyses were performed, optical microscopy, FE-SEM and AFM to underline the structure of the nanoporous MFC network. Release studies were conducted in an aqueous medium following two different protocols and antibacterial tests were done to evaluate the efficiency of the final materials obtained. MFC coating provided a slower and more progressive release of CHX. Indeed, papers impregnated with CHX were active for 18 days, whereas papers coated with CHX/MFC retained their antibacterial activity for 45 days. In parallel, similar tests were carried out using a model coating slurry, and although the rate of release of CHX was also slowed down, the quantities released were insufficient to confer any antibacterial activity. In conclusion, this study suggests that the use of MFC as a coating could be very promising since it allows a controlled and progressive release of molecules preserving long-term antibacterial activity.  相似文献

Two di-alkoxysilanes, with (AMDES, aminopropylmethyldiethoxysilane) or without (DMDES, dimethyldiethoxysilane) an amine function, and a tri-alkoxy aminosilane (APTES, aminopropyltriethoxysilane) as well as their mixtures were introduced in paper as fiber strengthening agents. The polymerization and copolymerization of these polysiloxanes in the paper were investigated. In all the cases where APTES was present, the formation of networks was established by measuring the soluble fraction amount extracted from the treated papers. A slight decrease of the opacity of the paper sheets when AMDES was part of the treatment was noted. The presence of APTES reduced this opacity loss. The study of the physicochemical properties of the treated paper (mechanical strength and alkalinity) demonstrated that, besides the required deacidification feature, the different treatments allowed an efficient strengthening of the cellulose fibers to various extents. Contact angle measurements indicated a decrease of the hydrophilic character of papers treated with the mixture APTES/AMDES and the occurrence of a hydrophobic character of the papers treated with APTES alone. These results were consistently obtained for both spray and immersion treatment processes.  相似文献

The extensive use of mesenchymal stem cells (MCS) in tissue engineering and cell therapy increases the necessity to improve their expansion. Among these, porcine MCS are valuable models for tissue engineering and are classically expanded in static T-flasks. In this work, different processes of stirred cultures were evaluated and compared. First, the effect of glucose, glutamine, antioxidant, and growth factors concentrations on porcine MSC expansion were analyzed in a suitable medium by performing kinetic studies. Results showed that a lower glucose concentration (5.5 mM) enabled to increase maximal cell concentration by 40 % compared with a higher one (25 mM), while addition of 2 to 6 mM of glutamine increased maximal cell concentration by more than 25 % compared with no glutamine supplementation. Moreover, supplementation with 1 μM thioctic acid increased maximal cell concentration by 40 % compared with no supplementation. Using this adapted medium, microcarriers cultures were performed and compared with T-flasks expansion. Porcine MSC were shown to be able to proliferate on the five types of microcarriers tested. Moreover, cultures on Cytodex 1, Cytopore 2, and Cultispher G exhibited a MSC growth rate more than 40 % higher compared with expansion in T-flasks, while MSC metabolism was similar.  相似文献

The discovery of novel protein–protein interaction (PPI) modulators represents one of the great molecular challenges of the modern era. PPIs can be modulated by either inhibitor or stabilizer compounds, which target different though proximal regions of the protein interface. In principle, protein–stabilizer complexes can guide the design of PPI inhibitors (and vice versa). In the present work, we combine X‐ray crystallographic data from both stabilizer and inhibitor co‐crystal complexes of the adapter protein 14‐3‐3 to characterize, down to the atomic scale, inhibitors of the 14‐3‐3/Tau PPI, a potential drug target to treat Alzheimer’s disease. The most potent compound notably inhibited the binding of phosphorylated full‐length Tau to 14‐3‐3 according to NMR spectroscopy studies. Our work sets a precedent for the rational design of PPI inhibitors guided by PPI stabilizer–protein complexes while potentially enabling access to new synthetically tractable stabilizers of 14‐3‐3 and other PPIs.  相似文献
