The potential of carbon nanotubes for the solid phase extraction of parabens in cosmetic products and the detection using a corona-charged aerosol detector (C-CAD) is presented in this work. The analytical procedure is based on a conventional solid phase extraction step for which 20 mg of multi-walled carbon nanotubes were packed in a 3-mL commercial SPE cartridge. Methylparaben, ethylparaben, propylparaben and butylparaben were thus isolated and preconcentrated from the pre-treated samples and subsequently separated on a RP-C18 column using acetonitrile:water, 50:50 (v/v) as mobile phase. The analytical signals for the individual parabens were obtained using C-CAD. The experimental variables affecting the extraction procedure and the instrumental detection have been deeply studied. Limits of detection were in the range of 0.5–2.1 mg L−1, while the linear range was extended up to 400 mg L−1. The average precision of the method varied between 3.3–3.8% (repeatability) and 4.3–7.6% (reproducibility). Finally, the optimized procedure was applied to the determination of the target preservatives in a variety of cosmetic products with satisfactory results.  相似文献

A liquid chromatography with diode array detection coupled to dual electrospray atmospheric pressure chemical ionization time-of-flight mass spectrometry (HPLC/ESI-APCI-TOF-MS) method is described for the rapid determination of five monophosphate nucleotides (cytidine 5′-monophosphate, uridine 5′-monophosphate, adenosine 5′-monophosphate, inosine 5′-monophosphate and guanosine 5′-monophosphate) in baby foods. The method is based on the deproteinisation of foods and direct analysis of nucleotides by ion-pair HPLC using isocratic elution with a mobile phase of 5% (v/v) methanol and 95% (v/v) 0.1 M formate buffer (pH 5.5) containing 0.01 M N,N-dimethylhexylamine (DMHA) at a flow-rate of 0.7 mL min−1. The HPLC was hyphenated with two different detection systems, photodiode-array (DAD) and ESI-APCI-TOF-MS in negative mode. The method was validated for linearity, detection and quantitation limits, selectivity, accuracy and precision. The recoveries obtained for spiked samples were satisfactory for all the analytes. The method was successfully applied to the analysis of nucleotides in different baby and/or functional food samples, as cereals, purees and dairy products. A study was also carried out on the stability of nucleotides in acidified dairy infant food with pasteurized yoghourt and follow-on formulae samples stored at room temperature and at 30 °C.  相似文献

An innovative approach is presented to interpret the refractive index of binary liquid mixtures. The concept of refractive index “before mixing” is introduced and shown to be given by the volume‐fraction mixing rule of the pure‐component refractive indices (Arago–Biot formula). The refractive index of thermodynamically ideal liquid mixtures is demonstrated to be given by the volume‐fraction mixing rule of the pure‐component squared refractive indices (Newton formula). This theoretical formulation entails a positive change of refractive index upon ideal mixing, which is interpreted in terms of dissimilar London dispersion forces centred in the dissimilar molecules making up the mixture. For real liquid mixtures, the refractive index of mixing and the excess refractive index are introduced in a thermodynamic manner. Examples of mixtures are cited for which excess refractive indices and excess molar volumes show all of the four possible sign combinations, a fact that jeopardises the finding of a general equation linking these two excess properties. Refractive indices of 69 mixtures of water with the amphiphile (R,S)‐1‐propoxypropan‐2‐ol are reported at five temperatures in the range 283–303 K. The ideal and real refractive properties of this binary system are discussed. Pear‐shaped plots of excess refractive indices against excess molar volumes show that extreme positive values of excess refractive index occur at a substantially lower mole fraction of the amphiphile than extreme negative values of excess molar volume. Analysis of these plots provides insights into the mixing schemes that occur in different composition segments. A nearly linear variation is found when Balankina’s ratios between excess and ideal values of refractive indices are plotted against ratios between excess and ideal values of molar volumes. It is concluded that, when coupled with volumetric properties, the new thermodynamic functions defined for the analysis of refractive indices of liquid mixtures give important complementary information on the mixing process over the whole composition range.  相似文献

The lamellar mesophase formed by surfactant 1,4-bis(2-ethylhexyl) sodium sulfosuccinate (AOT) in deuterated water is mixed with poly(dimethylacrylamide) (PDMAA) polymers of low molecular weight (Mn= (2-20) x 10(3)). The mixtures separate into microphases (lamellar plus isotropic polymer solution). Their microstructures are studied by microscopy, small-angle X-ray scattering (SAXS), and deuterium NMR (2H NMR). According to SAXS, the lamellar phase fractionates the molecular weight distribution of the polymer, by dissolving only chains with coil sizes smaller than the thickness of the water layers between lamellae, and keeping larger chains segregated from the lamellar phase. The fraction of polymer that is segregated from the lamellar phase grows with Mn of the polymer. In 2H NMR, there are two signals, a quadrupolar doublet (water molecules hydrating the anisotropic lamellar phase contribute to this doublet) and a singlet (water molecules in the isotropic polymer solution contribute to this singlet). These two signals are deconvoluted to analyze the phases. Mixing with the polymer produces the partial dispersion of the lamellar phase into small fragments (microcrystallites). The structure of these microcrystallites is such that they conserve the regular long period spacing of the macrophase, and are thus identified in SAXS, but they are smaller than the minimum size required to produce quadrupolar splitting (about 4 microm), and therefore, in 2H NMR, they contribute to the singlet. 2H NMR can thus not distinguish between small microcrystallites and an isotropic polymer solution segregated from the lamellar phase; instead small microcrystallites are detected as an apparent increase of the isotropic solution. The degree of dispersion produced by the polymer in the lamellar phase is correlated with the degree of segregation that the polymer suffers. Thus, much greater dispersion into microcrystallites is produced by the higher Mn polymers than by the lower Mn polymers (in the range covered by the present samples, although with a much higher molecular weight sample (3 x 10(6)) that is totally segregated no such microcrystallites were detected).  相似文献

A liquid chromatography electrospray mass spectrometry (LC/ESI/MS) method has been developed to identify and quantify microcystins in fish liver and intestine. Microcystins (MCs) were extracted from 500 mg sample with methanol-water (85:25, v/v) and the extracts concentrated to 250 microl. The parameters were optimized by a full factorial 2(3) design. Neither laborious pre-treatment nor clean up were necessary. MCs were separated using conventional C18 column and an acetonitrile-acidified water (pH 3) gradient. Negative samples (without MCs) were discriminated by liquid chromatography diode array detection (LC/DAD). The limits of detection (LOD) and the limits of quantification (LOQ) resulted equal for MC-RR, MC-YR, and MC-LR and were 0.1 and 0.5 microg g(-1), respectively. MCs recoveries at three levels in spiked samples (0.5-3.0 microg g(-1)) were > 92%, with relative standards deviations (RSDs) < 16% for liver samples and > 68% with RSDs < 18% for intestine samples. The proposed method was applied to determine MC-LR in exposed fish to evaluate the bioaccumulation risk. The results showed the transference of MC-LR from cyanobacterial cells to fish tissues.  相似文献

The reaction of ScCl(3)(THF)(3) or YCl(3) in a 1:1 molar ratio under reflux for 8 h with [{Li(bdmpza)(H(2)O)}(4)] [bdmpza = bis(3,5-dimethylpyrazol-1-yl)acetate], [{Li(bdmpzdta)(H(2)O)}(4)] [bdmpzdta = bis(3,5-dimethylpyrazol-1-yl)dithioacetate], and (Hbdmpze) [bdmpze = 2,2-bis(3,5-dimethylpyrazol-1-yl)ethoxide] affords the corresponding complexes [MCl(2)(kappa(3)-bdmpzx)(THF)] (x = a, M = Sc (1), Y (2); x = dta, M = Sc (3), Y (4); x = e, M = Sc (5), Y (6)). However, when the reaction was carried out for 1 h under reflux between ScCl(3)(THF)(3) and [{Li(bdmpzdta)(H(2)O)}(4)], a new anionic complex [Li(THF)(4)][ScCl(3)(kappa(3)-bdmpzdta)] (7) was obtained. Reaction of [{Li(bdmpza)(H(2)O)}(4)] with YCl(3) in a 2:1 molar ratio under reflux for 8 h gave the complex [YCl(kappa(3)-bdmpza)(2)] (8). The same reaction, but with the lithium compound [{Li(bdmpzdta)(H(2)O)}(4)], led to the formation of an anionic complex [Li(THF)(4)][YCl(3)(kappa(3)-bdmpzdta)] (9). The X-ray crystal structures of 7 and 9 were established. Finally, the addition of 1 equiv of [{Li(bdmpza)(H(2)O)}(4)] or [{Li(bdmpzdta)(H(2)O)}(4)] to a solution of YCl(3) in THF under reflux, followed by the addition of 1 equiv of 1,10-phenanthroline, resulted in the formation of the corresponding complexes [YCl(2)(kappa(3)-bdmpzx)(phen)] (x = a (10), x = dta (11)). These complexes are the first examples of group 3 metals stabilized by heteroscorpionate ligands. In addition, we have explored the reactivity of some of these complexes with alcohols and amides. For example, the direct reaction of [YCl(2)(kappa(3)-bdmpza)(THF)] (2) with several alcohols gave the alkoxide complexes [YCl(kappa(3)-bdmpza)(OR)] (R = Et (12), iPr (13)). Finally, the reaction between [ScCl(2)(kappa(3)-bdmpzdta)(THF)] (3) or [Li(THF)(4)][ScCl(3)(kappa(3)-bdmpzdta)] (7) and LiN(SiMe(3))(2).Et(2)O in 1:1 and 1:2 molar ratios gave rise to the complexes [ScCl(kappa(3)-bdmpzdta){N(SiMe(3))(2)}] (14) and [Sc(kappa(3)-bdmpzdta){N(SiMe(3))(2)}(2)] (15), respectively.  相似文献

A novel gold nanoparticle-based protocol for detection of DNA hybridization based on a magnetically trigged direct electrochemical detection of gold quantum dot tracers is described. It relies on binding target DNA (here called DNA1) with Au(67) quantum dot in a ratio 1:1, followed by a genomagnetic hybridization assay between Au(67)-DNA1 and complementary probe DNA (here called DNA2) marked paramagnetic beads. Differential pulse voltammetry is used for a direct voltammetric detection of resulting Au(67) quantum dot-DNA1/DNA2-paramagnetic bead conjugate on magnetic graphite-epoxy composite electrode. The characterization, optimization, and advantages of the direct electrochemical detection assay for target DNA are demonstrated. The two main highlights of presented assay are (1) the direct voltammetric detection of metal quantum dots obviates their chemical dissolution and (2) the Au(67) quantum dot-DNA1/DNA2-paramagnetic bead conjugate does not create the interconnected three-dimensional network of Au-DNA duplex-paramagnetic beads as previously developed nanoparticle DNA assays, pushing down the achievable detection limits.  相似文献

The specific interaction of a variety of modified hevein domains to chitooligosaccharides has been studied by NMR spectroscopy in order to assess the importance of aromatic-carbohydrate interactions for the molecular recognition of neutral sugars. These mutant AcAMP2-like peptides, which have 4-fluoro-phenylalanine, tryptophan, or 2-naphthylalanine at the key interacting positions, have been prepared by solid-phase synthesis. Their three-dimensional structures, when bound to the chitin-derived trisaccharide, have been deduced by NMR spectroscopy. By using DYANA and restrained molecular dynamics simulations with the AMBER 5.0 force field, the three-dimensional structures of the protein-sugar complexes have been obtained. The thermodynamic analysis of the interactions that occur upon complex formation have also been carried out. Regarding binding affinity, the obtained data have permitted the deduction that the larger the aromatic group, the higher the association constant and the binding enthalpy. In all cases, entropy opposes binding. In contrast, deactivation of the aromatic rings by attaching fluorine atoms decreases the binding affinity, with a concomitant decrease in enthalpy. The role of the chemical nature of the aromatic ring for establishing sugar contacts has been thus evaluated.  相似文献

An optimized capacitively coupled contactless conductivity detector for microchip electophoresis is presented. The detector consists of a pair of top–bottom excitation electrodes and a pair of pickup electrodes disposed onto a very thin plastic microfluidic chip. The detection cell formed by the electrodes is completely encased and shielded in a metal housing. These approaches allow for the enhancement of signal coupling and extraction from the detection cell that result in an improved signal‐to‐noise‐ratio and detection sensitivity. The improved detector performance is illustrated by the electrophoretic separation of six cations (NH, K+, Ca2+, Na+, Mg2+, Li+) with a detection limit of approximately 0.3 μM and the analysis of the anions (Br?, Cl?, NO, NO, SO, F?) with a detection limit of about 0.15 μM. These LODs are significantly improved compared with previous reports using the conventional top–top electrode geometry. The developed system was applied to the analysis of ions in bottled drinking water samples.  相似文献

This work expands the phytochemical composition knowledge of Acanthus mollis and evaluates antioxidant and anti-inflammatory activities which could be related with its traditional uses. Extracts from leaves, obtained by sequential extraction, were screened using TLC and HPLC-PDA. The ethanol extract was the most active on DPPH assay (IC50 = 20.50 μg/mL) and inhibited nitric oxide (NO) production in RAW 264.7 macrophages (IC50 = 48.31 μg/mL). Significant amounts of cyclic hydroxamic and phenolic acids derivatives were detected. A lower antioxidant effect was verified for a fraction enriched with DIBOA derivatives (IC50 = 163.02 μg/mL), suggesting a higher contribution of phenolic compounds for this activity in ethanol extract. However, this fraction exhibited a higher inhibition of NO production (IC50 = 32.32 μg/mL), with absence of cytotoxicity. These results support the ethnomedical uses of this plant for diseases based on inflammatory processes. To our knowledge, it is the first report to the anti-inflammatory activity for DIBOA derivatives.  相似文献
