Here we present a combined experimental and theoretical study on the secondary structure of isolated proteins as a function of charge state. In infrared spectra of the proteins ubiquitin and cytochrome c, amide I (C=O stretch) and amide II (N–H bend) bands can be found at positions that are typical for condensed‐phase proteins. For high charge states a new band appears, substantially red‐shifted from the amide II band observed at lower charge states. The observations are interpreted in terms of Coulomb‐driven transitions in secondary structures from mostly helical to extended C5‐type hydrogen‐bonded structures. Support for this interpretation comes from simple energy considerations as well as from quantum chemical calculations on model peptides. This transition in secondary structure is most likely universal for isolated proteins that occur in mass spectrometric experiments.  相似文献

The numerical simulation of the mechanical behavior of industrial materials is widely used for viability verification, improvement and optimization of designs. Elastoplastic models have been used to forecast the mechanical behavior of different materials. The numerical solution of most elastoplastic models comes across problems of ill-condition matrices. A complete representation of the nonlinear behavior of such structures involves the nonlinear equilibrium path of the body and handling of singular (limit) points and/or bifurcation points. Several techniques to solve numerical problems associated to these points have been disposed in the specialized literature. Two examples are the load-controlled Newton–Raphson method and displacement controlled techniques. However, most of these methods fail due to convergence problems (ill-conditioning) in the neighborhood of limit points, specially when the structure presents snap-through or snap-back equilibrium paths. This study presents the main ideas and formalities of the Tikhonov regularization method and shows how this method can be used in the analysis of dynamic elastoplasticity problems. The study presents a rigorous mathematical demonstration of existence and uniqueness of the solution of well-posed dynamic elastoplasticity problems. The numerical solution of dynamic elastoplasticity problems using Tikhonov regularization is presented in this paper. The Galerkin method is used in this formulation. Effectiveness of Tikhonov’s approach in the regularization of the solution of elastoplasticity problems is demonstrated by means of some simple numerical examples.  相似文献

Magnetic Resonance (MR) white matter hyperintensities have been shown to predict an increased risk of developing cognitive decline. However, their actual role in the conversion to dementia is still not fully understood. Automatic segmentation methods can help in the screening and monitoring of Mild Cognitive Impairment patients who take part in large population-based studies. Most existing segmentation approaches use multimodal MR images. However, multiple acquisitions represent a limitation in terms of both patient comfort and computational complexity of the algorithms. In this work, we propose an automatic lesion segmentation method that uses only three-dimensional fluid-attenuation inversion recovery (FLAIR) images. We use a modified context-sensitive Gaussian mixture model to determine voxel class probabilities, followed by correction of FLAIR artifacts. We evaluate the method against the manual segmentation performed by an experienced neuroradiologist and compare the results with other unimodal segmentation approaches. Finally, we apply our method to the segmentation of multiple sclerosis lesions by using a publicly available benchmark dataset. Results show a similar performance to other state-of-the-art multimodal methods, as well as to the human rater.  相似文献

In this paper an eigenvalue complementarity problem (EiCP) is studied, which finds its origins in the solution of a contact problem in mechanics. The EiCP is shown to be equivalent to a Nonlinear Complementarity Problem, a Mathematical Programming Problem with Complementarity Constraints and a Global Optimization Problem. A finite Reformulation–Linearization Technique (Rlt)-based tree search algorithm is introduced for processing the EiCP via the lattermost of these formulations. Computational experience is included to highlight the efficacy of the above formulations and corresponding techniques for the solution of the EiCP.  相似文献

In this paper we simultaneously consider three extensions to the standard Orienteering Problem (OP) to model characteristics that are of practical relevance in planning reconnaissance missions of Unmanned Aerial Vehicles (UAVs). First, travel and recording times are uncertain. Secondly, the information about each target can only be obtained within a predefined time window. Due to the travel and recording time uncertainty, it is also uncertain whether a target can be reached before the end of its time window. Finally, we consider the appearance of new targets during the flight, so-called time-sensitive targets, which need to be visited immediately if possible. We tackle this online stochastic UAV mission planning problem with time windows and time-sensitive targets using a re-planning approach. To this end, we introduce the Maximum Coverage Stochastic Orienteering Problem with Time Windows (MCS-OPTW). It aims at constructing a tour with maximum expected profit of targets that were already known before the flight. Secondly, it directs the planned tour to predefined areas where time-sensitive targets are expected to appear. We have developed a fast heuristic that can be used to re-plan the tour, each time before leaving a target. In our computational experiments we illustrate the benefits of the MCS-OPTW planning approach with respect to balancing the two objectives: the expected profits of foreseen targets, and expected percentage of time-sensitive targets reached on time. We compare it to a deterministic planning approach and show how it deals with uncertainty in travel and recording times and the appearance of time-sensitive targets.  相似文献

Understanding of the dynamic particulate flow structures within a dense gas-fly ash pneumatic conveyor must be improved in order to better aid its design guidance.The complex pulsatile movement of the gas-fly ash mixture dominates the flow performance within the pipeline,and historically,non-invasive measurement devices such as the electrical capacitance tomography(ECT) were often used to sufficiently capture the flow dynamics.However,inadequate studies have been conducted on the pulsatile flow phenomenon,which directly relate to the gas-fly ash two-phase flow performance.This paper aims to investigate the pulsatile flows using an ECT device.Initially,pulsatile flow patterns under various experimental conditions were obtained through ECT.Pulses within a flow were then characterised into pulse growth and decay segments,which represent the superficial fluidisation and deaeration processes during conveying.Subsequently,structural and statistical analyses were performed on the pulse growth and decay segments.Results suggested that the increasing air mass flow rate led to the decrease of the superficial fluidisation/deaeration magnitude,however,the increase of the superficial fluidisation/deaeration durations.Also,the air mass flow rate was indicated as the dominant factor in determining the pulsing statistical parameters.This research provides fundamental insights for further modelling the dense fly ash pneumatic flows.  相似文献

The synthesis of ethyl (2′-hydroxy-4′,5′-methylendioxophenyl)acetate, a fragment of the antihyperglycemic natural coumarin subcoriacin, is reported. We found an expeditious route to the title compound in five steps. Final metal catalyzed acid ethanolysis of the vinylic 1,1-methylthio methylsulfoxide derivative afforded the required aryl acetic ester, but in the absence of metal catalyst, an unexpected Pummerer rearrangement produced the 2,3-dimethylthiofuran derivative as the major product. This last result provides an alternative entry to 2,3-dimethlythiobenzofurans.  相似文献

An efficient biological route to production of gold nanoparticles which allows the nanoparticles to be easily recovered remains elusive. Live cells of the green microalga Chlorella vulgaris were incubated with a solution of gold chloride and harvested by centrifugation. Nanoparticles inside intact cells were identified by transmission electron microscopy and confirmed to be metallic gold by synchrotron based X-ray powder diffraction and X-ray absorption spectroscopy. These intracellular gold nanoparticles were 40–60 nm in diameter. At a concentration of 1.4% Au in the alga, a better than 97% recovery of the gold from solution was achieved. A maximum of 4.2% Au in the alga was obtained. Exposure of C. vulgaris to solutions containing dissolved salts of palladium, ruthenium, and rhodium also resulted in the production of the corresponding nanoparticles within the cells. These were surmised to be also metallic, but were produced at a much lower intracellular concentration than achieved with gold. Iridium was apparently toxic to the alga. No nanoparticles were observed using platinum solutions. C. vulgaris provides a possible route to large scale production of gold nanoparticles.  相似文献
