Two Burkholderia gladioli strains isolated from the lungs of cystic fibrosis patients were found to produce unusual lipodepsipeptides containing a unique citrate-derived fatty acid and a rare dehydro-β-alanine residue. The gene cluster responsible for their biosynthesis was identified by bioinformatics and insertional mutagenesis. In-frame deletions and enzyme activity assays were used to investigate the functions of several proteins encoded by the biosynthetic gene cluster, which was found in the genomes of about 45 % of B. gladioli isolates, suggesting that its metabolic products play an important role in the growth and/or survival of the species. The Chrome Azurol S assay indicated that these metabolites bind ferric iron, which suppresses their production when added to the growth medium. Moreover, a gene encoding a TonB-dependent ferric-siderophore receptor is adjacent to the biosynthetic genes, suggesting that these metabolites may function as siderophores in B. gladioli. 相似文献

Α simple, relatively rapid, sensitive and cost-effective method based on ultrasound-assisted emulsification microextraction (USAEME) followed by gas chromatography coupled with mass spectrometry has been developed for the determination of seven endocrine disruptor pesticides (chlorpyrifos, deltamethrin, dimethoate, fenitrothion, malathion, pendimethalin and procymidone) in apple juice. This approach is based on the emulsification of organic extraction solvent in a diluted apple juice sample by ultrasound radiation and further separation of both liquids phases by centrifugation. The influence of the different parameters affecting the procedure (extraction solvent, extraction solvent volume, ultrasound time, centrifugation time, ionic strength and pH) was evaluated in order to optimise the efficiency of the extraction process. Target analytes were extracted from a 0.5 g apple juice sample that was diluted by 10 times with aqueous buffer solution (pH 7). The optimised USAEME procedure used 100 μL of chloroform as extraction solvent, 8 min of ultrasound extraction, ionic strength (2.5% w/v) and 7.5 min of centrifugation at 3800 rpm. The optimised method presented recoveries between 70 and 113% for the target analytes. Acceptable linearity for all target analytes was recorded with correlation coefficients (r) higher than 0.992. The limits of quantification were found between 1.1 and 4.6 μg kg?1 ensuring compliance with the maximum residue limits established by the European Commission. The proposed method was applied for the determination of the endocrine disruptor pesticides in apple samples proving its suitability to the Commission Implementing Regulation (EU) no. 400/2014. 相似文献

Colistimethate sodium (CMS) is widely administrated for the treatment of life-threatening infections caused by multidrug-resistant Gram-negative bacteria. Until now, the quality control of CMS formulations has been based on microbiological assays. Herein, an ultra-high-performance liquid chromatography coupled to ultraviolet detector methodology was developed for the quantitation of CMS in injectable formulations. The design of experiments was performed for the optimization of the chromatographic parameters. The chromatographic separation was achieved using a Waters Acquity BEH C8 column employing gradient elution with a mobile phase consisting of (A) 0.001 M aq. ammonium formate and (B) methanol/acetonitrile 79/21 (v/v). CMS compounds were detected at 214 nm. In all, 23 univariate linear-regression models were constructed to measure CMS compounds separately, and one partial least-square regression (PLSr) model constructed to assess the total CMS amount in formulations. The method was validated over the range 100–220 μg mL−1. The developed methodology was employed to analyze several batches of CMS injectable formulations that were also compared against a reference batch employing a Principal Component Analysis, similarity and distance measures, heatmaps and the structural similarity index. A selective assay for the determination of one of the most important class of phenolic compounds, namely trihydroxybenzoates (monomeric and polymeric compounds having at least one gallate moiety) based on their enhancing effect on the chemiluminogenic reaction between gold ions and luminol is described for the first time. In the presence of trihydroxybenzoate derivatives, the light emission generated when alkaline luminol is oxidized by gold ions is amplified several orders of magnitude compared to other common phenolic compounds which exhibit minor reactivity or no reactivity at all (e.g. hydroxycinnamates, flavonols, benzenediols). Based on this property, the experimental conditions were optimized in order to enable the determination of total trihydroxybenzoates in complex mixtures without resorting to separation techniques. The method was applied to samples of different composition (teas, herbal infusions and wines) with satisfactory analytical features yielding detection limits at the 10−7 mol L−1 level, intra-day precision of 3.1%, inter-day precision less than 10% and recoveries between 88.7 and 97.6%. The strengths and weaknesses of the method were identified and discussed in relation to its application in real samples. 相似文献

Abstract Proteus bacteria are important human opportunistic pathogens which frequently cause urinary tract infections. According to Bergey's Manual of Systematic Bacteriology,1 this genus includes three species: P. mirabilis, P. vulgaris, and P. myxofaciens. A novel species of P. penneri has been recently proposed2,3 for strains formerly called P. vulgaris biogroup I. Proteus is an antigenically heterogeneous group of bacteria, and this is mainly associated with diverse composition and structures of O-specific polysaccharide chains of outer-membrane lipopolysaccharides (O-antigens). The Kauffman-Perch serological classification4 of P. mirabilis and P. vulgaris includes 49 O-serogroups. However, a number of S-strains remain unclassified,5 including strains of P. penneri. 相似文献

Tears have a significant role in antioxidant defense in ocular tissues and since their collection is quick and noninvasive, their analysis would facilitate monitoring of pathophysiological changes. However, their low volume and low content of antioxidants makes analysis difficult; methods of high sensitivity are needed. In this paper, we present a method for tear analysis of two antioxidant molecules (ascorbic and uric acid) and of a lipid peroxidation indicator (malondialdehyde) with capillary electrophoresis. Tears were collected with Schirmer strips, extracted with a low‐pH phosphate buffer, centrifuged through membrane filters and an antioxidant was added. They were stable at ?70°C for 15 days. After pilot experiments, optimum electrophoretic separation was achieved in a 25 mm borate buffer, pH 10.0, containing 100 mm sodium dodecyl sulfate at 25°C and 20 kV. The developed method has good repeatability (<5% RSD), precision (<15% relative error values) and high sensitivity (LLOQ values of 20, 2.3 and 2.5 μM for ascorbate, urate and malondialdehyde, respectively). It was applied to the analysis of tears from healthy individuals and the antioxidant levels are in agreement with those obtained with other techniques. Biobased pigments are environmentally friendly alternatives to synthetic variants with an increased market demand. Production of pigments via fermentation is a promising process, yet optimization of the production yield and rate is crucial. Herein, we evaluated the potential of Penicillium purpurogenum to produce biobased pigments. Optimum sugar concentration was 30 g/L and optimum C:N ratio was 36:1 resulting in the production of 4.1–4.5 AU (namely Pigment Complex A). Supplementation with ammonium nitrate resulted in the production of 4.1–4.9 AU (namely Pigment Complex B). Pigments showed excellent pH stability. The major biopigments in Pigment Complex A were N-threonyl-rubropunctamin or the acid form of PP-R (red pigment), N-GABA-PP-V (violet pigment), PP-O (orange pigment) and monascorubrin. In Pigment Complex B, a novel biopigment annotated as N-GLA-PP-V was identified. Its basic structure contains a polyketide azaphilone with the same carboxyl-monascorubramine base structure as PP-V (violet pigment) and γ-carboxyglutamic acid (GLA). The pigments were not cytotoxic up to 250 μg/mL. 相似文献

Bulk free‐radical polymerization of 2‐vinylpyridine (2VP) in the presence of 2,2,6,6‐tetramethylpiperidine‐N‐oxyl (TEMPO) was studied under different conditions (temperature and presence of additives). Linear poly‐(2‐vinylpyridine) with a narrow molecular weight distribution and controllable molecular weight was prepared in the presence of acetic anhydride at 95 °C up to a conversion of 66%. At higher conversions side reactions became very important (pseudoliving polymerization). By applying this procedure, well‐defined random copolymers of 2VP with styrene or tert‐butylmethacrylate as well as block copolymers of 2VP with styrene were synthesized. © 2001 John Wiley & Sons, Inc. E-commerce will strongly penetrate the market if coupled with appropriate technologies and mechanisms. Mobile agents may enhance the intelligence and improve the efficiency of systems in the e-marketplace. We propose a dynamic multi-lateral negotiation model and construct an efficient negotiation strategy based on a ranking mechanism that does not require a complicated rationale on behalf of the buyer agents. This strategy can be used to extend the functionality of autonomous intelligent agents, so that they quickly reach to an agreement aiming to maximise their owner’s utility. The framework proposed considers both contract and decision issues, is based on real market conditions, and has been empirically evaluated. Moreover, it is shown that in a linear framework like the one we employ, more elaborate ranking mechanisms do not necessarily improve efficiency. 相似文献
