Measurement of X‐ray absorption spectroscopy (XAS) in transmission is the method of choice for strong or concentrated samples. In a typical XAS experiment above 5 keV the sample is placed between the first (I0) and second (I1) ion chambers and a standard foil is placed between the second (I1) and third (I2) ion chambers for simultaneous calibration of energy during sample analysis. However, some fluorescence from the foil may be registered in I1, causing anomalies in the transmission signal of the sample, especially when the sample edge jump is relatively small. To remedy this, Soller slits were constructed and placed between the foil and I1 to minimize back‐fluorescence from the foil. A comparison of blank and standard samples, measured with or without Soller slits or under a worst‐case scenario, demonstrates the advantages of Soller slits when analyzing weak signal samples via transmission XAS.  相似文献

Robust and selective quantification methods are required to better analyze feed supplementation effectiveness with specific amino acids. In this work, a reversed-phase high-performance liquid chromatography method with fluorescence detection is proposed and validated for lysine quantification, one of the most limiting amino acids in ruminant nutrition and essential towards milk production. To assess and widen method applicability, different matrices were considered: namely Li2CO3 buffer (the chosen standard reaction buffer), phosphate buffer solution (to mimic media in cellular studies), and rumen inoculum. The method was validated for all three matrices and found to be selective, accurate (92% ± 2%), and precise at both the inter- and intra-day levels in concentrations up to 225 µM, with detection and quantification limits lower than 1.24 and 4.14 µM, respectively. Sample stability was evaluated when stored at room temperature, 4 °C, and −20 °C, showing consistency for up to 48 h regardless of the matrix. Finally, the developed method was applied in the quantification of lysine on real samples. The results presented indicate that the proposed method can be applied towards free lysine quantification in ruminant feeding studies and potentially be of great benefit to dairy cow nutrition supplementation and optimization.  相似文献

Three corrosion inhibitors for copper-3-amino-1,2,4-triazole (ATA), benzotriazole (BTAH), and 1-hydroxybenzotriazole (BTAOH)-were investigated by corrosion experiments and atomistic computer simulations. The trend of corrosion inhibition effectiveness of the three inhibitors on copper in near-neutral chloride solution is determined experimentally as BTAH ? ATA ? BTAOH. A careful analysis of the results of computer simulations based on density functional theory allowed to pinpoint the superior inhibiting action of BTAH and ATA as a result of their ability to form strong N-Cu chemical bonds in deprotonated form. While these bonds are not as strong as the Cl-Cu bonds, the presence of solvent favors the adsorption of inhibitor molecules onto the surface due to stronger solvation of the Cl(-) anions. Moreover, benzotriazole displays the largest affinity among the three inhibitors to form intermolecular aggregates, such as [BTA-Cu](n) polymeric complex. This is another factor contributing to the stability of the protective inhibitor film on the surface, thus making benzotriazole an outstanding corrosion inhibitor for copper. These findings cannot be anticipated on the basis of inhibitors' molecular electronic properties alone, thus emphasizing the importance of a rigorous modeling of the interactions between the components of the corrosion system in corrosion inhibition studies.  相似文献

MALDI imaging mass spectrometry (MALDI-IMS) has become a powerful tool for the detection and localization of drugs, proteins, and lipids on-tissue. Nevertheless, this approach can only perform identification of low mass molecules as lipids, pharmaceuticals, and peptides. In this article, a combination of approaches for the detection and imaging of proteins and their identification directly on-tissue is described after tryptic digestion. Enzymatic digestion protocols for different kinds of tissues—formalin fixed paraffin embedded (FFPE) and frozen tissues—are combined with MALDI-ion mobility mass spectrometry (IM-MS). This combination enables localization and identification of proteins via their related digested peptides. In a number of cases, ion mobility separates isobaric ions that cannot be identified by conventional MALDI time-of-flight (TOF) mass spectrometry. The amount of detected peaks per measurement increases (versus conventional MALDI-TOF), which enables mass and time selected ion images and the identification of separated ions. These experiments demonstrate the feasibility of direct proteins identification by ion-mobility-TOF IMS from tissue. The tissue digestion combined with MALDI-IM-TOF-IMS approach allows a proteomics “bottom-up” strategy with different kinds of tissue samples, especially FFPE tissues conserved for a long time in hospital sample banks. The combination of IM with IMS marks the development of IMS approaches as real proteomic tools, which brings new perspectives to biological studies.  相似文献

By building on our earlier work, we establish uncertainty principles in terms of Heisenberg inequalities and of the ambiguity functions associated with magnetic structures on certain coadjoint orbits of infinite-dimensional Lie groups. These infinite-dimensional Lie groups are semidirect products of nilpotent Lie groups and invariant function spaces thereon. The recently developed magnetic Weyl calculus is recovered in the special case of function spaces on abelian Lie groups.  相似文献

Detection of the interactions between low molecular weight organic compounds and metals in the form of sols on a nanoscale is analytically challenging. This study aims to provide experimental evidence using a combination of UV-Vis absorption spectrometry, surface-enhanced Raman spectrometry (SERS), and static secondary ion mass spectrometry (S-SIMS). The field of application is thermography where silver images are formed via heat-catalyzed reactions. Several organic compounds called tone modifiers and stabilizers are used in thermographic materials for the optimization of the image quality. With exploitation of the strengths of each of the above-mentioned methods, an affinity ranking of several tone modifiers and a stabilizer was established on the basis of competitive adsorption experiments using different model systems. Specifically, silver sols, SERS probes, and sputter-coated silver substrates were exposed to systems with one or two additives. The UV-Vis results provided insight on the aggregation of silver nanoparticles in a hydrosol, which was necessary for the interpretation of the SERS data. Both SERS and S-SIMS measurements led to a similar ranking of the relative affinity of the additives in two components, which was largely consistent with empirical knowledge derived from macroscopic behavior.  相似文献

The response is subject to left truncation and right censoring. Under this model, which reduces for special choices of f{\phi} to e.g. Cox proportional hazards model or the additive hazards model with time dependent coefficients, we study the estimation of the vector \pmbb(z){\pmb{\beta}(z)} . A least squares approach is proposed and the asymptotic properties of the proposed estimator are established. The estimator is also compared with a competing maximum likelihood based estimator by means of simulations. Finally, the method is applied to a larynx cancer data set.  相似文献
