Three tetrahydroisoquinolinedione derivatives

N‐(2‐Chloro­benzyl)‐1,2,3,4‐tetra­hydro­iso­quinoline‐1,3‐dione, C₁₆H₁₂ClNO₂, crystallizes in P2₁/n with three crystallographically independent mol­ecules in the asymmetric unit, which differ slightly in conformation, N‐(2‐bromo‐4‐methyl­phenyl)‐1,2,3,4‐tetra­hydro­iso­quinoline‐1,3‐dione, C₁₆H₁₂BrNO₂, crystallizes in P2₁/n with one mol­ecule in the asymmetric unit andN‐(2,3‐di­chloro­phenyl)‐1,2,3,4‐tetra­hydro­iso­quinoline‐1,3‐dione, C₁₅H₉Cl₂NO₂, crystallizes in P2₁/c with one mol­ecule in the asymmetric unit. In all three structures, the heterocyclic rings adopt approximately planar conformations. The pyridine rings are orthogonal to the substituted phenyl rings. In all three structures, the crystal packing is stabilized by intermolecular C—H?O hydrogen bonds.     

Continuous flow and flow injection stripping voltammetric determination of silver(I), mercury(II), and bismuth(III) at a bulk modified graphite tube electrode

The epoxy-impregnated graphite tube electrode bulk modified with 2-mercaptobenzoxazole, employed in a wall-jet configuration, was found to be useful for the continuous flow and flow injection stripping voltammetric determinations of Ag^I, Hg^II and Bi^III. For continuous flow, detection limits for Ag^I, Hg^II and Bi^III were 1.8 × 10⁻¹⁰ M, 1.9 × 10⁻⁹ M and 9.5 × 10⁻⁹ M, respectively (10 min accumulation, S/N = 3). Precisions for 5.00 × 10⁻⁹ M Ag^I, 1.00 × 10⁻⁸ M Hg^II and 1.00 × 10⁻⁷ M Bi^III were 10.5%, 5.77 % and 7.90% (relative standard deviations, n = 6), respectively. In the case of flow injection stripping, with a 500 μL injection loop, detection limits of 0.59 ng, 2.0 ng and 120 ng were obtained for Ag^I, Hg^II and Bi^III, respectively (S/N = 3). Selected metal ions, inorganic and organic substances were investigated for interferences. The electrode was tested with a certified sample and then applied to the determinations of the metal ions in a urine and a sea-water sample.     

Stereoselective synthesis of (+)-SCH 351448: a unique ligand system for sodium, calcium, and other cations

(+)-SCH 351448 (Na+ salt A) was synthesized employing ring-closing olefin metathesis reaction of an open diene diester intermediate for construction of the 28-membered macrodiolide structure. The open diene diester was prepared from the monomeric hydroxy carboxylic acid and two different olefin fragments. The monomeric hydroxy acid was synthesized via Julia-Julia coupling reaction of intermediates derived from the same olefinic fragments. Oxane units in these fragments were prepared by radical cyclization reactions of beta-alkoxyacrylates. Analogous SCH 351448 salts incorporating other mono- and divalent cations may be prepared. Under acidic conditions, SCH 351448 (Na+ salt A) was the most stable complex, but SCH 351448 (Ca2+ salt) and (Na+ salt B) appear to be physiologically important species.     

Field and flow programming in frit-inlet asymmetrical flow field-flow fractionation

The separation of wide molecular mass (Mr) ranges of macromolecules using frit inlet asymmetrical flow field-flow fractionation (FI-AFlFFF) has been improved by implementing a combination of field and flow programming. In this first implementation, field strength (governed by the cross flow-rate through the membrane-covered accumulation wall) is decreased with time to obtain faster elution and improved detection of the more strongly retained (high Mr) materials. The channel outlet flow-rate is optionally held constant, increased, or decreased with time. With circulation of the flow exiting the accumulation wall to the inlet frit, the dual programming of cross flow and channel outlet flow could be implemented using just two pumps. With this flow configuration, the channel outlet flow-rate is always equal to the channel inlet flow-rate, and these may be programmed independently of the cross flow-rate through the membrane. FI-AFlFFF retains its operational advantage over conventional asymmetrical flow FFF (AFlFFF). Unlike conventional AFlFFF, FI-AFlFFF does not require time consuming, and experimentally inconvenient, sample focusing and relaxation steps involving valve switching and interruption of sample migration. The advantages of employing dual programming with FI-AFlFFF are demonstrated for sets of polystyrene sulfonate standards in the molecular mass range of 4 to 1000 kDa. It is shown that programmed FI-AFlFFF successfully expands the dynamic separation range of molecular mass.     

Growth promotion of HepG2 hepatoma cells by antisense-mediated knockdown of glypican-3 is independent of insulin-like growth factor 2 signaling

Glypican-3 (GPC3) encodes a cell-surface heparan- sulfate proteoglycan and its expression is frequently silenced in ovarian cancer, mesotheliomas, and breast cancer cell lines and ectopic expression of GPC3 inhibited the growth of these cells, suggesting that GPC3 plays a negative role in cell proliferation. In contrast, up-regulation of GPC3 is often observed in hepatoma, neuroblastoma, and Wilms' tumor. Whether GPC3 plays the same growth inhibitory role in these tumors remains to be studied. Here we report that antisense-mediated knockdown of GPC3 in the HepG2 hepatoma cells significantly promotes the growth of hepatoma cells. In addition, we show that this growth promotion is independent of insulin-like growth factor 2 (IGF2) signaling. Our data suggest that GPC3 plays a growth-suppressing role in hepatoma and provide cell biological evidence inconsistent with the hypothesis that GPC3 acts as a growth suppressor by downregulating IGF2.     

Dye-sensitized solar cells based on composite solid polymer electrolytes

The ionic conductivity of polymer electrolytes and their interfacial contact with dye-attached TiO2 particles were enhanced markedly by the addition of amorphous oligomer into polymer electrolytes, resulting in very high overall energy conversion efficiency.     

Synthesis and manganese complexes of pentagonal bipyramidal ligands: N,N′-disubstituted pentaaza macrocycles

Two novel heptadentate ligands, pentaaza macrocycles with two pendant xpyridyl and phenol groups, were prepared and the crystal structure of the manganese(II) complex of N,N′-bis(2-pyridylmethyl)-pentaaza macrocycle revealed a pentagonal bipyramidal geometry.     

Elemental analysis of Korean women’s blood serums using instrumental neutron activation analysis

Instrumental neutron activation analysis was used to assess the concentration of the inorganic trace elements in Korean women’s blood serums. It was found that a high concentration of Na and Cl incurs an analytical interference, but the 12 elements such as Br, Ca, Cl, Co, Cr, Cs, Fe, K, Na, Rb, Se and Zn could be determined under the condition of an interference minimization. Serum samples collected from 63 women were analyzed and the concentration level and range of the elements were evaluated. NIST SRMs were analyzed simultaneously for quality control. The average values of the Na and Cl determined in the serum samples were 3,365 and 3,533 mg/l, Ca was 96.4 mg/l and K was 191 mg/l. Besides, Br, Se and Zn have a concentration level of 6.46, 0.13 and 0.98 mg/l, respectively. It was found that there is no significant difference between the present values and the reported values.     

Syntheses of 1beta-methylcarbapenems bearing 5-methyl-4-hydroxypyrrolidinone

A new series of 1beta-methylcarbapenems 1a-d bearing 5-methyl-4-mercaptopyrrolidinone rings has been prepared and evaluated for in vitro antibacterial activity and pharmacokinetic parameters. Most compounds showed excellent antibacterial activity and high stability to dehydropeptidase-1. We have synthesized optically active 5-methyl-4-hydroxypyrrolidinones from enantiomerically pure aziridine esters.
