Ion-association complexes of antazoline HCl [I], hydralazine HCl [II], amiloride HCl [III] and quinine sulphate [IV] with [Co(SCN)4]2– and [Co(NO2)6]3– were precipitated and the excess unreacted cobalt complex was determined. A new method using atomic emission and atomic absorption spectrometry for the determination of the above drugs in pure solutions and pharmaceutical preparations is given. The drugs can be determined in the ranges 0.3–3.0, 0.19–1.96, 0.3–3.0, and 0.78–7.82 mg/25 ml solutions of I, II, III, and IV, respectively, with mean relative standard deviations of 0.65–2.03 % and recovery values of 95.76–101.2% indicating high precision and accuracy.  相似文献

The reactive adenosine derivative, adenosine 5′-O-[S-(4-hydroxy-2,3-dioxobutyl)]-thiophosphate (AMPS-HDB), contains a dicarbonyl group linked to the purine nucleotide at a position equivalent to the pyrophosphate region of NAD+. AMPS-HDB was used as a chemical label towards Candida boidinii formate dehydrogenase (CbFDH). AMPS-HDB reacts covalently with CbFDH, leading to complete inactivation of the enzyme activity. The inactivation kinetics of CbFDH fit the Kitz and Wilson model for time-dependent, irreversible inhibition (KD = 0.66 ± 0.15 mM, first order maximum rate constant k3 = 0.198 ± 0.06 min−1). NAD+ and NADH protects CbFDH from inactivation by AMPS-HDB, showing the specificity of the reaction. Molecular modelling studies revealed Arg174 as a candidate residue able to be modified by the dicarbonyl group of AMPS-HDB. Arg174 is a strictly conserved residue among FDHs and is located at the Rossmann fold, the common mononucleotide-binding motif of dehydrogenases. Arg174 was replaced by Asn, using site-directed mutagenesis. The mutant enzyme CbFDHArg174Asn was showed to be resistant to inactivation by AMPS-HDB, confirming that the guanidinium group of Arg174 is the target for AMPS-HDB. The CbFDHArg174Asn mutant enzyme exhibited substantial reduced affinity for NAD+ and lower thermostability. The results of the study underline the pivotal and multifunctional role of Arg174 in catalysis, coenzyme binding and structural stability of CbFDH.  相似文献

Various chitosan (CS)-based nanoparticles (CS-NPs) of ciprofloxacin hydrochloride (CHCl) have been investigated for therapeutic delivery and to enhance antimicrobial efficacy. However, the Box–Behnken design (BBD)-supported statistical optimization of NPs of CHCl has not been performed in the literature. As a result, the goal of this study was to look into the key interactions and quadratic impacts of formulation variables on the performance of CHCl-CS-NPs in a systematic way. To optimize CHCl-loaded CS-NPs generated by the ionic gelation process, the response surface methodology (RSM) was used. The BBD was used with three factors on three levels and three replicas at the central point. Tripolyphosphate, CS concentrations, and ultrasonication energy were chosen as independent variables after preliminary screening. Particle size (PS), polydispersity index (PDI), zeta potential (ZP), encapsulation efficiency (EE), and in vitro release were the dependent factors (responses). Prepared NPs were found in the PS range of 198–304 nm with a ZP of 27–42 mV. EE and drug release were in the range of 23–45% and 36–61%, respectively. All of the responses were optimized at the same time using a desirability function based on Design Expert® modeling and a desirability factor of 95%. The minimum inhibitory concentration (MIC) of the improved formula against two bacterial strains, Pseudomonas aeruginosa and Staphylococcus aureus, was determined. The MIC of the optimized NPs was found to be decreased 4-fold compared with pure CHCl. The predicted and observed values for the optimized formulation were nearly identical. The BBD aided in a better understanding of the intrinsic relationship between formulation variables and responses, as well as the optimization of CHCl-loaded CS-NPs in a time- and labor-efficient manner.  相似文献

Type 2 diabetes mellitus is considered to be a substantial socioeconomic burden worldwide on both patients and governments. Coumarins are biomolecules with a diversity of biological activities. The current investigation aimed to explore the ameliorative effects of cichoriin, which is a type of coumarin, on high-fat diet/streptozotocin (HFD/STZ)-induced diabetic rats. Methods: Rats were allocated into five groups. Group I was considered as the control group, while the other groups were HFD/STZ-induced diabetic rats. Group II was assigned as the diabetic control. Groups III and IV were treated with cichoriin (50 or 100 mg/kg, respectively). Group V received glibenclamide (5 mg/kg) (as a positive control). The blood glucose (BG), serum insulin, triglycerides (TG), total cholesterol (TC), total antioxidant capacity (TAC), catalase, hepatic superoxide dismutase (SOD) and content of malondialdehyde (MDA) were assessed. Histopathological and immunohistochemistry analysis of pancreatic tissue were performed. mRNA and protein expressions of GLUT4, AMPK, and PI3K were estimated. Results: Cichoriin treatment ameliorated HFD/STZ-induced diabetic conditions and mitigated the histopathological characteristics of the pancreas, as well as increasing pancreatic insulin expression. This decreased the levels of BG, TG, TC, and MDA and improved the TAC, catalase and SOD contents. Cichoriin demonstrated upregulation of mRNA and protein expressions of GLUT4, AMPK, and PI3K. The in silico binding of cichoriin with GLUT4, AMPK, and PI3K supported the possible current activities. Conclusion: Collectively, this work highlighted the potential role of cichoriin in mitigating HFD/STZ-induced diabetic conditions and showed it to be a valuable product.  相似文献

The reactions between strontium and iron nitrates have been studied in an open atmosphere system using three different molar ratios, 1:1 (I), 1:2 (II) and 2:1 (III) at different temperatures as pointed out from the DTA data. The reaction mechanism was discussed based on the chemical composition characterized by means of thermal analysis, X‐ray diffraction patterns, infrared spectra and magnetic susceptibility. It was found that the reaction products depend on both temperature of reaction and the ratio between reactants. The reaction products were found to be composed of a variety of iron compounds that possess different valences: SrFeO2.86, SrFeO2.97, SrFe2O4, SrFe12O19, Sr2Fe2O5 and Sr7Fe10O22 in addition to some accessory reaction products namely α‐Fe2O3 and FeO(OH).  相似文献

In the title compound, [Fe(C17H14P)2]2[Sb4Cl16]·C2H6O, the Fe atoms lie on inversion centres and the pairs of cyclopentadienyl rings are consequently in a fully staggered conformation. The centrosymmetric anionic clusters formed by [Sb4Cl16]4? are surrounded by the cations and are held together by weak C—H?Cl interactions. Membrane electrodes based on 2,2-dithiobis(benzothiazole), DTBBT, as a neutral carrier for silver ions are described. Silver-selective membrane electrodes formulated with 2wt% DTBBT ionophore and 50mol% TFPB in an FPNPE plasticized poly (vinyl chloride) exhibited near-Nernstian responses towards silver ions (60.3±0.5mVdecade–1) over a wide silver ion activity range of 0.83µM to 94mM. Increasing the amount of anionic sites, TFPB, to 100 or 150mol% (relative to the DTBBT weight) resulted in super-Nernstian responses toward silver ions. Membrane electrodes prepared using a low dielectric constant plasticizer, however, exhibited sub-Nernstian responses. Polymer membrane electrodes with optimal composition (i.e., 2wt% DTBBT, 50mol% TFPB in FPNPE plasticized poly(vinyl chloride)) exhibited high potentiometric selectivity towards silver ions over alkali, alkaline earth, transition metal ions, as well as heavy metal ions such as Hg2+ and Pb2+. A good correlation was found between the potentiometric selectivity coefficients and the change in the UV-visible spectra of the ionophore upon exposure to different metal ions. The overall performance of the silver-selective membrane electrodes based on DTBBT ionophore, which is available at low cost, was found to be comparable to the performance of silver electrodes prepared with Fluka silver ionophore-IV. A DTBBT-based silver electrode was used as an indicator electrode for titrations of silver ions using standard sodium chloride solutions. Sharp inflections occur at the end point, and the data obtained showed 99.4% recovery with a standard deviation of 0.7% (n=3). In addition, the applicability of the DTBBT-based silver-selective electrode is illustrated by measuring the silver concentrations in natural water spiked with silver nitrate and by analyzing the silver in electroplating wastewater samples. The results obtained utilizing a DTBBT-based silver electrode showed very good agreement with the standard methods of analysis.  相似文献
