The use of combinatorial chemistry has become commonplace within the pharmaceutical industry. Less widespread but gaining in popularity is the derivation of activity models from the high-throughput assays of these libraries. Such models are then used as filters during the design of refined daughter libraries. The design of these second generation libraries, which efficiently test and conform to the derived activity model from the large space of virtual possibilities, remains an area of considerable research. We present here a computationally efficient method for the design of optimally dense (in model matching compounds) synthetic matrices from in silico virtual libraries.  相似文献

Abstract— The photochemistry of 2,7-diiodo-9-diazofiuorene was studied to examine its suitability as a photolabeling agent of hydrophobic sites in biological assemblies. Irradiation of the diazo compound generates 2,7-diiodofluorenylidene. The carbene was detected by laser transient absorption spectroscopy and characterized by its chemical and physical properties. Like fluorenylidene, the triplet is the ground state of 2,7-diiodofluorenylidene. However, the substituted triplet carbene does not reform its higher energy, electrophilic singlet state fast enough for reactions of the upper state to compete with the irreversible consumption of the triplet. Thus, the products from the reactions of diiodofluorenylidene contain a higher proportion of those characteristic of the triplet carbene than occurs in the reactions of fluorenylidene. This will limit the utility of this diazocompound as a photolabeling agent.  相似文献

A set of compounds consisting of a new and diverse collection of biarylamides was examined using quantitative structure-activity relationship techniques for the purpose of developing a model to describe inhibition of gram-positive bacterial growth (minimum inhibition concentration). The model was sought in order to obtain insight for designing new molecules. A detailed analysis of the underlying structure-activity relationship helped provide insight concerning which structural features of the molecules modulated the activity of the compounds against gram-positive organisms.  相似文献

The 351.1 nm photoelectron spectrum of the 1-pyrazolide-d(3) anion has been measured. The photoelectron angular distributions indicate the presence of nearly degenerate electronic states of the 1-pyrazolyl-d(3) radical. Equation-of-motion ionization potential coupled-cluster singles and doubles (EOMIP-CCSD) calculations have been performed to study the low-lying electronic states. The calculations strongly suggest that three electronic states, energetically close to each other, are accessed in the photodetachment process. Strong interactions of the pseudo-Jahn-Teller type in each pair of the three states are evident in the calculations for the radical at the anion geometry. Model diabatic potentials of the three states have been constructed around the anion geometry in terms of the anion reduced normal coordinates up to the second order. An analytic method to parametrize the quadratic vibronic coupling (QVC) model potentials has been introduced. Parameters of the QVC model potentials have been determined from the EOMIP-CCSD and CCSD(T) calculations. Simulations of the 1-pyrazolide-d(3) spectrum have been performed with the model Hamiltonian, treating all vibronic interactions amongst the three states simultaneously. The simulation reproduces the fine structure of the observed spectrum very well, revealing complicated nonadiabatic effects in the low-lying states of the radical. The ground state of the 1-pyrazolyl-d(3) radical is (2)A(2) and the electron affinity is 2.935+/-0.006 eV. The first excited state is (2)B(1) with a term energy of 32+/-1 meV. While the high-symmetry (C(2v)) stationary points of the X (2)A(2) and A (2)B(1) states are minima, that of the state is a saddle point as a result of the pseudo-Jahn-Teller interactions with the other two states. The topology of the adiabatic potential energy surfaces is discussed.  相似文献

The objective of this study is to synthesize ZnO and Mg doped ZnO (Zn1−xMgxO) nanoparticles via the sol-gel method, and characterize their structures and to investigate their biological properties such as antibacterial activity and hemolytic potential.Nanoparticles (NPs) were synthesized by the sol-gel method using zinc acetate dihydrate (Zn(CH3COO)2.2H2O) and magnesium acetate tetrahydrate (Mg(CH3COO)2.4H2O) as precursors. Methanol and monoethanolamine were used as solvent and sol stabilizer, respectively. Structural and morphological characterizations of Zn1−xMgxO nanoparticles were studied by using XRD and SEM-EDX, respectively. Photocatalytic activities of ZnO and selected Mg-doped ZnO (Zn1−xMgxO) nanoparticles were investigated by degradation of methylene blue (MeB). Results indicated that Mg doping (both 10% and 30%) to the ZnO nanoparticles enhanced the photocatalytic activity and a little amount of Zn0.90 Mg0.10 O photocatalyst (1.0 mg/mL) degraded MeB with 99% efficiency after 24 h of irradiation under ambient visible light. Antibacterial activity of nanoparticles versus Escherichia coli ( E. coli ) was determined by the standard plate count method. Hemolytic activities of the NPs were studied by hemolysis tests using human erythrocytes. XRD data proved that the average particle size of nanoparticles was around 30 nm. Moreover, the XRD results indicatedthat the patterns of Mg doped ZnO nanoparticles related to ZnO hexagonal wurtzite structure had no secondary phase for x ≤ 0.2 concentration. For 0 ≤ x ≤ 0.02, NPs showed a concentration dependent antibacterial activity against E. coli . While Zn0.90Mg0.10 O totally inhibited the growth of E. coli , upper and lower dopant concentrations did not show antibacterial activity.  相似文献

Infrared (IR) absorption spectra of matrix-isolated HCCCH(2) have been measured. Propargyl radicals were generated in a supersonic pyrolysis nozzle, using a method similar to that described in a previous study (Jochnowitz, E. B.; Zhang, X.; Nimlos, M. R.; Varner, M. E.; Stanton, J. F.; Ellison, G. B. J. Phys. Chem. A 2005, 109, 3812-3821). Besides the nine vibrational modes observed in the previous study, this investigation detected the HCCCH(2) X? (2)B(1) out-of-plane bending mode (ν(8)) at 378.0 (±1.9) cm(-1) in a cryogenic argon matrix. This is the first experimental observation of ν(8) for the propargyl radical. In addition, seven overtone and combination bands have also been detected and assigned. Ab initio coupled-cluster anharmonic force field calculations were used to guide the analysis. Furthermore, ν(12), the HCCCH(2) in-plane bending mode, has been assigned to 333 (±10) cm(-1) based on the detection of its overtone (2ν(12), 667.7 ± 1.0 cm(-1)) and a possible combination band (ν(10) + ν(12), 1339.0 ± 0.8 cm(-1)). This is the first experimental estimation of ν(12) for the propargyl radical.  相似文献

A computationally inexpensive energy correction is suggested for radicals described by the equation-of-motion coupled cluster method for ionized states in the singles and doubles approximation (EOMIP-CCSD). The approach is primarily intended for doublet states that are qualitatively described by Koopmans' approximation. Following a strategy similar to those used in multireference coupled cluster theory, the proposed correction accounts for all correlation effects through third order in perturbation theory and also includes selected contributions to higher-order energies. As an initial test of the numerical performance of the method, total energies and energy splittings are calculated for some small prototype radicals.  相似文献
