We report a novel electrochemical method for detecting sequence‐specific DNA based on competitive hybridization that occurs in a homogeneous solution phase instead of on a solution‐electrode interface as in previously reported competition‐based electrochemical DNA detection schemes. The method utilizes the competition between the target DNA (t‐DNA) and a ferrocene‐labeled peptide nucleic acid probe (Fc‐PNA) to hybridize with a probe DNA (p‐DNA) in solution. The neutral PNA backbone and the electrostatic repulsion between the negatively‐charged DNA backbone and the negatively‐charged electrode surface are then exploited to determine the result of the competition through measurement of the electrochemical signal of Fc. Upon the introduction of the t‐DNA, the stronger hybridization affinity between the t‐DNA and p‐DNA releases the Fc‐PNA from the Fc‐PNA/p‐DNA hybrid, allowing it to freely diffuse to the negatively charged electrode to produce a significantly enhanced electrochemical signal of Fc. Therefore, the presence of the t‐DNA is indicated by the appearance or enhancement of the electrochemical signal, rendering a signal‐on DNA detection, which is less susceptible to false positive and can produce more reliable results than signal‐off detection methods. All the competitive hybridizations occur in a homogeneous solution phase, resulting in very high hybridization efficiency and therefore extremely short assay time. This simple and fast signal‐on solution‐competition‐based electrochemical DNA detection strategy has promising potential to find application in fields such as nucleic acid‐based point‐of‐care testing. 相似文献

In this paper, an M/G/1 queue with exponentially working vacations is analyzed. This queueing system is modeled as a two-dimensional embedded Markov chain which has an M/G/1-type transition probability matrix. Using the matrix analytic method, we obtain the distribution for the stationary queue length at departure epochs. Then, based on the classical vacation decomposition in the M/G/1 queue, we derive a conditional stochastic decomposition result. The joint distribution for the stationary queue length and service status at the arbitrary epoch is also obtained by analyzing the semi-Markov process. Furthermore, we provide the stationary waiting time and busy period analysis. Finally, several special cases and numerical examples are presented. 相似文献

We consider a network of sensors that measure the intensities of a complex plume composed of multiple absorption–diffusion source components. We address the problem of estimating the plume parameters, including the spatial and temporal source origins and the parameters of the diffusion model for each source, based on a sequence of sensor measurements. The approach not only leads to multiple‐source detection, but also the characterization and prediction of the combined plume in space and time. The parameter estimation is formulated as a Bayesian inference problem, and the solution is obtained using a Markov chain Monte Carlo algorithm. Floating spherical Gaussian orbital (FSGO ) open-shell calculations have been made to determine the potential energy surface of planar square and rectangular arrangements of the four-electron system H4. This surface is discussed in relation to the bimolecular isotope exchange reaction H2+D2-→ 2HD. The changes in energy and geometry accompanying the coplanar approach of two hydrogen molecules interacting chemically have also been investigated. Calculations on the electronic energies of planar T-shaped and kite arrangements of H4 of various sizes show that it is unlikely that these configurations can serve as transition states for the exchange reaction. However, the energy curve for linear configurations of H4 (H? H? H … H), calculated as a function of the H3 … H distance with the symmetric linear H3 (H-H-H) unit fixed at the internuclear distance of 1.9080 a.u., is found to have a deep minimum (?1.9176 a.u.) at an r(H3 … H) distance of 1.5846 a.u. The overall results suggest that the following mechanism for the exchange reaction, H2+H2→H2+H+H→H3+H→H+H2+ H→H2+H2 could be advantageous as it requires a barrier height of 0.1604 a.u. which is significantly lower than that calculated from the saddle point energy (0.1950 a.u.). However, the problem of reconciling this with the experimental activation energy of 0.0685 a.u. still remains. 相似文献

In this paper we report the catalytic effects of various gold nanoparticles for silver electrodeposition on indium tin oxide (ITO)-based electrodes, and successfully apply this methodology for signal amplification of the hybridization assay. The most widely used gold nanoparticle-based hybridization indicators all promote silver electrodeposition on the bare ITO electrodes, with decreasing catalytic capability in order of 10 nm gold, DNA probe-10 nm gold conjugate, streptavidin-5 nm gold, and streptavidin-10 nm gold. Of greater importance, these electrocatalytic characteristics are affected by any surface modifications of the electrode surfaces. This is illustrated by coating the ITO with an electroconducting polymer, poly(2-aminobenzoic acid)(PABA), as well as avidin molecules, which are promising immobilization platforms for DNA biosensors. The catalytic silver electrodeposition of the gold nanoparticles on the PABA-coated ITO surfaces resembles that on the bare surfaces. With avidin covalently bound to the PABA, it is interesting to note that the changes in electrocatalytic performance vary for different types of gold nanoparticles. For the streptavidin-5 nm gold, the silver electrodeposition profile is unaffected by the presence of the avidin layer, whereas for both the 10 nm Au and DNA probe-10 nm gold conjugate, the deposition profiles are suppressed. The streptavidin-5 nm gold is employed as the hybridization indicator, with avidin-modified (via PABA) ITO electrode as the immobilization platform, to enable signal amplification by the silver electrodeposition process. Under the conditions, this detection strategy offers a signal-to-noise ratio of 20. We believe that this protocol has great potential for simple, reproducible, highly selective and sensitive DNA detection on fully integrated microdevices in clinical diagnostics and environmental monitoring applications. 相似文献
