In this study, we developed a method for the determination of PF‐04620110 (2‐{(1r,4r)‐4‐[4‐(4‐amino‐5‐oxo‐7,8‐dihydropyrimido[5,4‐f][1,4]oxazepin‐6(5H)‐yl)phenyl]cyclohexyl}acetic acid), a novel diacylglycerol acyltransferase 1 (DGAT‐1) inhibitor, in rat plasma and validated it using liquid chromatography–tandem mass spectrometry (LC‐MS/MS). Rat plasma samples were processed following a protein precipitation method by using acetonitrile and were then injected into an LC‐MS/MS system for quantification. PF‐04620110 and imipramine (internal standard) were separated using a Hypersil Gold C18 column, with a mixture of acetonitrile and 10 mm ammonium formate (90:10, v/v) as the mobile phase. The ion transitions monitored in positive‐ion mode [M + H]+ of multiple‐reaction monitoring were m/z 397.0 → 260.2 for PF‐04620110 and m/z 280.8 → 86.0 for imipramine. The detector response was specific and linear for PF‐04620110 at concentrations within the range 0.05–50 µg/mL and the signal‐to‐noise ratios for the samples were ≥10. The intra‐ and inter‐day precision and accuracy of the method matched the acceptance criteria for assay validation. PF‐04620110 was stable under various processing and/or handling conditions. PF‐04620110 concentrations in the rat plasma samples could be measured up to 24 h after intravenous or oral administration of PF‐04620110, suggesting that the assay is useful for pharmacokinetic studies in rats. An efficient and facile two-step strategy for the synthesis of (Z)-aurone from arylacetylenes and salicyladehydes, via silver(I) nitrate mediated cyclization/oxidation in the presence of potassium carbonate has been developed. A series of N,N‐disubstituted‐4‐[(4‐aminophenyl)diazenyl]benzylidene‐4′‐alkylanilines (azo dyes) were synthesized from the reaction of the corresponding benzaldehyde with alkylanilines. These azo dyes exhibit nematic and SmC phases on heating. Their order parameter, photo‐stability and miscibility were studied by investigation of a representative sample.  相似文献

5-(7-Hydroxyheptyl)-1,2-dithiolan-3-one 1-oxide was designed and synthesized in our laboratories that contain the heterocycle of 1,2-dithiolan-3-one 1-oxide, a reactive core of antibiotic leinamycin. On the basis of a realization of thermodynamic requirements as given in part I, simple kinetic considerations are applied to the processes of weak bond fission and block recombination from two distinct bases: (a) independence of rate constants and viscosity (or average molecular weight) of the polymer system; and (b) a diffusion-controlled situation where the rate of block recombination is governed by the viscosity of the medium. On the independent rate constant model, an activation energy for the recombination process of less than 40 kJ [mole (of weak bonds)]?1 gives an acceptable rate of equilibration at ambient temperatures; this criterion ought to be readily satisfied in practice. However, considerations of diffusion control suggest that equilibration may be too slow to be practicable and this is probably the major limitation on preparing rigid polymers with temperature-varying molecular weights  相似文献

The porosity of 1‐hexanethiol‐functionalised gold nanoparticle films was assessed and utilised as chemiresistor sensors. Electrochemical capacitance measurements showed that the accessibility of electrolytes of different ionic strengths into the pores depended on the thickness of the electric double layer formed. A large variation in capacitance was measured in 0.01–1000 mM NaClO4, implying a wide pore size distribution. The change in morphology of the nanoparticle films upon storage in air, water and ethanol for two weeks was investigated. There was a significant decrease in the electrochemical capacitance at high electrolyte concentrations for the ethanol‐stored films compared to the freshly‐prepared films suggesting a decrease in the number of small pores of radii in the range of 0.3–3 nm. This was further supported by optical topographical measurements where a decrease in the thickness of ethanol‐stored films was observed relative to the freshly‐prepared films. The porous nature of the nanoparticle films was found to have an effect on the chemical sensing behaviour. When used as chemiresistor sensors, for the detection of heptane in water, the ethanol‐stored films provided larger resistance changes and longer response times. This suggests that the more densely packed ethanol‐stored films provided more sites that enabled film swelling, and that diffusion of the analyte occurred through the narrower water‐filled pores. This demonstrates the effect of different storage conditions on film morphology and subsequently sensor response.  相似文献

Abstract

Since polyimides were invented more than 30 years ago, several books dealing with the subject have been published [33, 34, 40u, 48b, 55, 64]. While some of these books [33–35] have very thorough and systematic coverage of the chemistry and general physical, mechanical, and electrical properties of polyimides, most of the literature [40, 48b, 64] is made up of collections of results derived from application-related research on various polyimides. None of the earlier treatises is totally devoted to an understanding of the underlying principles involved in just a single, important property such as the glass transition temperatures of polyimides.  相似文献
