Ondansetron, a widely used antiemetic agent, is a P‐glycoprotein (P‐gp) substrate and therefore expression of P‐gp at the blood–brain barrier limits its distribution to the central nervous system (CNS), which was observed to be reversed by coadministration with P‐gp inhibitors. Tariquidar is a potent and selective third‐generation P‐gp inhibitor, and coadministration with ondansetron has shown improved ondansetron distribution to the CNS. There is currently no reported bioanalytical method for simultaneously quantifying ondansetron with a third‐generation P‐gp inhibitor. Therefore, we aimed to develop and validate a method for ondansetron and tariquidar in rat and human plasma samples. A full validation was performed for both ondansetron and tariquidar, and sample stability was tested under various storage conditions. To demonstrate its utility, the method was applied to a preclinical pharmacokinetic study following coadministration of ondansetron and tariquidar in rats. The presented method will be valuable in pharmacokinetic studies of ondansetron and tariquidar in which simultaneous determination may be required. In addition, this is the first report of a bioanalytical method validated for quantification of tariquidar in plasma samples.  相似文献

Template‐assisted formation of multicomponent Pd6 coordination prisms and formation of their self‐templated triply interlocked Pd12 analogues in the absence of an external template have been established in a single step through Pd? N/Pd? O coordination. Treatment of cis‐[Pd(en)(NO3)2] with K3tma and linear pillar 4,4′‐bpy (en=ethylenediamine, H3tma=benzene‐1,3,5‐tricarboxylic acid, 4,4′‐bpy=4,4′‐bipyridine) gave intercalated coordination cage [{Pd(en)}6(bpy)3(tma)2]2[NO3]12 ( 1 ) exclusively, whereas the same reaction in the presence of H3tma as an aromatic guest gave a H3tma‐encapsulating non‐interlocked discrete Pd6 molecular prism [{Pd(en)}6(bpy)3(tma)2(H3tma)2][NO3]6 ( 2 ). Though the same reaction using cis‐[Pd(NO3)2(pn)] (pn=propane‐1,2‐diamine) instead of cis‐[Pd(en)(NO3)2] gave triply interlocked coordination cage [{Pd(pn)}6(bpy)3(tma)2]2[NO3]12 ( 3 ) along with non‐interlocked Pd6 analogue [{Pd(pn)}6(bpy)3(tma)2](NO3)6 ( 3′ ), and the presence of H3tma as a guest gave H3tma‐encapsulating molecular prism [{Pd(pn)}6(bpy)3(tma)2(H3tma)2][NO3]6 ( 4 ) exclusively. In solution, the amount of 3′ decreases as the temperature is decreased, and in the solid state 3 is the sole product. Notably, an analogous reaction using the relatively short pillar pz (pz=pyrazine) instead of 4,4′‐bpy gave triply interlocked coordination cage [{Pd(pn)}6(pz)3(tma)2]2[NO3]12 ( 5 ) as the single product. Interestingly, the same reaction using slightly more bulky cis‐[Pd(NO3)2(tmen)] (tmen=N,N,N′,N′‐tetramethylethylene diamine) instead of cis‐[Pd(NO3)2(pn)] gave non‐interlocked [{Pd(tmen)}6(pz)3(tma)2][NO3]6 ( 6 ) exclusively. Complexes 1 , 3 , and 5 represent the first examples of template‐free triply interlocked molecular prisms obtained through multicomponent self‐assembly. Formation of the complexes was supported by IR and multinuclear NMR (1H and 13C) spectroscopy. Formation of guest‐encapsulating complexes ( 2 and 4 ) was confirmed by 2D DOSY and ROESY NMR spectroscopic analyses, whereas for complexes 1 , 3 , 5 , and 6 single‐crystal X‐ray diffraction techniques unambiguously confirmed their formation. The gross geometries of H3tma‐encapsulating complexes 2 and 4 were obtained by universal force field (UFF) simulations.  相似文献

The copper(I)‐catalyzed alkyne‐azide cycloaddition (CuAAC), so‐called “click” reaction, is one of most useful synthetic strategies to connect two polymer chains. 1,2,3‐Triazole ring (TA) produced by the click reaction has good thermal and chemical stability. However, we observed that block copolymers synthesized by the click reaction showed thermal degradation to give homopolymers when they are thermally annealed at high temperature, which is required for obtaining equilibrium microdomain structure. To investigate the origin of thermal instability of block copolymers, we synthesized model polystyrenes (PSs) using systematically designed bi‐functional atom transfer radical polymerization (ATRP) initiators containing TA. PS including both ester and TA groups showed thermal decomposition at relatively low temperature (e.g., 140 °C). MALDI‐TOF analysis clearly demonstrated that the cleavage site is the ester group adjacent to TA. We also found that the bromine group located at the polymer chain end plays an important role in pyrolysis of ester groups at low temperature. The pyrolysis occurs by syn‐elimination of the ester group. This result implies that the phase behavior of block copolymer synthesized by click reaction should be carefully investigated when high temperature thermal annealing is required. © 2016 Wiley Periodicals, Inc. DNA methylation is the most promising biomarker for estimating human age. There are various methods used for analyzing DNA methylation. Among those, the SNaPshot assay-based method provides a semi-quantitative measurement of DNA methylation using capillary electrophoresis on genetic analyzers. However, DNA methylation measures produced using different types of genetic analyzers have never been compared, although differences in methylation values can directly affect age estimates. To evaluate the differences between the results generated by different genetic analyzers, we analyzed the same blood, saliva, and control methylated DNA using three genetic analyzers—the Applied Biosystems 3130, 3500, and SeqStudio—and compared the methylation values at five CpG sites: ELOVL2, FHL2, KLF14, MIR29B2C, and TRIM59. The methylation value at each of the five CpG sites decreased in the order 3130, 3500, and SeqStudio. The differences in the results produced by the different genetic analyzers resulted in significant errors when applying the 3500 and SeqStudio data to a previous age estimation model constructed using the 3130 Genetic Analyzer data. Therefore, DNA methylation measurements from 3500 and SeqStudio were corrected using the regression functions obtained by plotting the DNA methylation data of one instrument versus the other to facilitate the application of DNA methylation data from one instrument to the age prediction model based on other instruments. The age prediction accuracy obtained by applying corrected 3500 and SeqStudio data to the existing age estimation model was as high as observed in the 3130 data.  相似文献

The neutron irradiated reactor pressure vessel (RPV) steels at various dose of 0–1018 n/cm2 have been studied with Mössbauer, x-ray diffraction, and VSM. The Mössbauer data shows that the value of magnetic hyperfine field of Fe atom that exist at martensite is 330 kOe at site 1 and 305 kOe at site 2. At room temperature, the total absorption area of Mössbauer spectra with respect to irradiation of neutron is constant for the dose of 0–1016 n/cm2, while over the dose of 0–1017 n/cm2 the absorption area decreases rapidly. But the doublet area for the dose of 0–1016 n/cm2 is constant, while over the dose of 1017 n/cm2 it increases with increasing the fluence level of neutron. The coercivity and remanence of the neutron irradiated samples do not change significantly. But the maximum induction decreases by 5% at 1018 n/cm2, compared with that of the as-received sample.  相似文献

Ni2+‐complexed poly(2‐acetamidoacrylic acid) (PAAA) hydrogel beads were developed for the site‐specific reversible immobilization and purification of the histidine‐tagged green fluorescent protein (His‐tagged GFP). PAAA hydrogel beads were prepared by photopolymerization, and significantly improved mechanical properties of PAAA hydrogel beads were observed in comparison with PAAA hydrogel from our previous study. Confocal laser scanning microscopy was used to determine the binding of His‐tagged GFP to the hydrogel beads in three‐dimensional space. Photoluminescence spectroscopy revealed 89% of binding efficiency of His‐tagged GFP to the Ni2+‐PAAA hydrogel beads, 51% of yielding recovery. The maximum binding capacity of His‐tagged GFP was estimated to be 0.45 µg/mg of Ni2+‐PAAA hydrogel beads. The recombinant His‐tagged GFP from the soluble fraction of E. coli BL21(DE3) cell lysates was purified with Ni2+‐PAAA hydrogel beads. The major advantage of the Ni2+‐PAAA hydrogel beads system was simple preparation procedures of producing the matrix, because PAAA hydrogel beads had relatively enhanced mechanical strength than soft hydrogels.
