The first total synthesis of (+/-)-ingenol has been achieved. The key features of the synthesis include the use of a highly diastereoselective Michael reaction to fix the C-11 methyl stereochemistry and the incorporation of the dimethylcyclopropane via diastereoselective carbene addition to the Delta13,14 olefin. The intramolecular dioxenone photoaddition-fragmentation sequence leads to the establishment of the critical C-8/C-10 trans intrabridgehead stereochemistry, a central challenge in the synthesis of ingenanes. The completion of the synthesis proceeds using the C-6alpha hydroxymethyl group as the sole handle for oxidation of seven contiguous carbon centers.  相似文献

Octupolar oligomers containing 2-12 molecules of 1,3,5-tricyano-2,4,6-tris(styryl)benzene derivatives have been synthesized and their nonlinear optical and two-photon absorption (TPA) properties were determined. The beta(0) values are in the range of (85-1219) x 10(-30) esu and increase monotonically with the increasing number of the octupolar units within the molecule. The two-photon-induced fluorescence excitation spectra are quite similar to the single-photon absorption spectra except that the wavelength is doubled, indicating that the one- and two-photon allowed excited states are the same. The peak TPA cross-section values (delta(max)) measured with nanosecond pulses by the two-photon-induced fluorescence method are in the range (3010-62, 930) x 10(-50) cm(4)s photon(-1). The delta(max) increases as the number of the octupolar units in the molecules increases. A linear relationship is observed between delta(max) and beta, and this delta-beta relationship serves as a useful design strategy for the synthesis of novel octupolar oligomers and polymers with large TPA and beta.  相似文献

Rat pheochromocytoma (PC12) cells have been used to investigate neurite outgrowth. Nerve growth factor (NGF) has been well known to induce neurite outgrowth from PC12 cells. RhoA belongs to Ras-related small GTP-binding proteins, which regulate a variety of cellular processes, including cell morphology alteration, actin dynamics, and cell migration. NGF suppressed GTP-RhoA levels after 12 h in PC12 cells and was consistently required for a long time to induce neurite outgrowth. Constitutively active (CA)-RhoA suppressed neurite outgrowth from PC12 cells in response to NGF, whereas dominant-negative (DN)-RhoA stimulated it, suggesting that RhoA inactivation is essential for neurite outgrowth. Here, we investigated the mechanism of RhoA inactivation. DN-p190RhoGAP abrogated neurite outgrowth, whereas wild-type (WT)-p190RhoGAP and WT-Src synergistically stimulated it along with accelerating RhoA inactivation, suggesting that p190RhoGAP, which can be activated by Src, is a major component in inhibiting RhoA in response to NGF in PC12 cells. Contrary to RhoA, Rap1 was activated by NGF, and DN-Rap1 suppressed neurite outgrowth, suggesting that Rap1 is also essential for neurite outgrowth. RhoA was co-immunoprecipitated with Rap1, suggesting that Rap1 interacts with RhoA. Furthermore, a DN-Rap-dependent RhoGAP (ARAP3) prevented RhoA inactivation, abolishing neurite formation from PC12 cells in response to NGF. These results suggest that NGF activates Rap1, which, in turn, up-regulates ARAP3 leading to RhoA inactivation and neurite outgrowth from PC12 cells. Taken together, p190RhoGAP and ARAP3 seem to be two main factors inhibiting RhoA activity during neurite outgrowth in PC12 cells in response to NGF.  相似文献

Dual emitting cores for thermally activated delayed fluorescent (TADF) emitters were developed. Relative to the corresponding TADF emitter with a single emitting core the TADF emitter with a dual emitting core, 3,3′,5,5′‐tetra(carbazol‐9‐yl)‐[1,1′‐biphenyl]‐2,2′,6,6′‐tetracarbonitrile, showed enhanced light absorption accompanied by a high photoluminescence quantum yield. The quantum and power efficiencies of the TADF devices were enhanced by the dual emitting cores.  相似文献

Hyperglycemia-induced oxidative stress accelerates endothelial cell dysfunctions, which cause various complications of diabetes. The protective effects of 6,6′-bieckol (BEK), one of phlorotannin compound purified from Ecklonia cava against high-glucose-induced oxidative stress was investigated using human umbilical vein endothelial cells (HUVECs), which is susceptible to oxidative stress. High glucose (30 mM) treatment induced HUVECs’ cell death, but BEK, at concentration 10 or 50 μg/ml, significantly inhibited the high-glucose-induced cytotoxicity. Furthermore, treatment with BEK dose-dependently decreased thiobarbituric acid reactive substances (TBARS), intracellular reactive oxygen species (ROS) generation, and nitric oxide level increased by high glucose. In addition, high glucose levels induced the overexpressions of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and nuclear factor-kappa B (NF-κB) proteins in HUVECs, but BEK treatment reduced the overexpressions of these proteins. These findings indicate that BEK is a potential therapeutic agent that will prevent diabetic endothelial dysfunction and related complications.  相似文献

Circulating biomarkers have emerged as promising non-invasive, real-time surrogates for cancer diagnosis, prognostication and monitoring of therapeutic response. Emerging data, however, suggest that single markers are inadequate in describing complex pathologic transformations. Architecting assays capable of parallel measurements of multiple biomarkers can help achieve the desired clinical sensitivity and specificity while conserving patient specimen and reducing turn-around time. Here we describe a plasmon-enhanced Raman spectroscopic assay featuring nanostructured biomolecular probes and spectroscopic imaging for multiplexed detection of disseminated breast cancer markers cancer antigen (CA) 15-3, CA 27-29 and cancer embryonic antigen (CEA). In the developed SERS assay, both the assay chip and surface-enhanced Raman spectroscopy (SERS) tags are functionalized with monoclonal antibodies against CA15-3, CA27-29 and CEA, respectively. Sequential addition of biomarkers and functionalized SERS tags onto the functionalized assay chip enable the specific recognition of these biomarkers through the antibody-antigen interactions, leading to a sandwich spectro-immunoassay. In addition to offering extensive multiplexing capability, our method provides higher sensitivity than conventional immunoassays and demonstrates exquisite specificity owing to selective formation of conjugated complexes and fingerprint spectra of the Raman reporter. We envision that clinical translation of this assay may further enable asymptomatic surveillance of cancer survivors and speedy assessment of treatment benefit through a simple blood test.  相似文献

High‐performance countercurrent chromatography (HPCCC) with electrospray light‐scattering detection was applied for the first time to isolate a spirostanol and a novel furostanol saponin from Liriope platyphylla. Due to the large differences in KD values between the two compounds, a two‐step HPCCC method was applied in this study. The primary HPCCC employed methylene chloride/methanol/isopropanol/water (9:6:1:4 v/v, 4 mL/min, normal‐phase mode) conditions to yield a spirostanol saponin ( 1 ). After the primary HPCCC run, the solute retained in the stationary phase (SP extract) in HPCCC column was recovered and subjected to the second HPCCC on the n‐hexane/n‐butanol/water system (1:9:10 v/v, 5 mL/min, reversed‐phase mode) to yield a novel furostanol saponin ( 2 ). The isolated spirostanol saponin was determined to be 25(S)‐ruscogenin 1‐O‐β‐d ‐glucopyranosyl (1→2)‐[β‐d ‐xylopyranosyl (1→3)]‐β‐d ‐fucopyranoside (spicatoside A), and the novel furostanol saponin was elucidated to be 26‐O‐β‐d ‐glucopyranosyl‐25(S)‐furost‐5(6)‐ene‐1β‐3β‐22α‐26‐tetraol‐1‐O‐β‐d ‐glucopyranosyl (1→2)‐[β‐d ‐xylopyranosyl‐(1→3)]‐β‐d ‐fucopyranoside (spicatoside D).  相似文献

The authors describe an SPR sensor chip coated with gold nanoparticles (AuNPs) that enables highly sensitive determination of genetically modified (GM) crops. Detection is based on localized surface plasmon resonance (LSPR) with its known sensitivity to even minute changes in refractive index. The device consists of a halogen light source, a light detector, and a cuvette cell that contains a sensor chip coated with AuNPs. It is operated in the transmission mode of the optical path to enhance the plasmonic signal. The sample solution containing target DNA (e.g. from the GM crop) is introduced into the cuvette with the sensor chip whose surface was functionalized with a capture DNA. Following a 30-min hybridization, the changes of the signal are recorded at 540 nm. The chip responds to target DNA in the 1 to 100 nM concentration range and has a 1 nM detection limit. Features of this sensor chip include a short reaction time, ease of handling, and portability, and this enables on-site detection and in-situ testing.

An ongoing pandemic of coronavirus disease 2019 (COVID-19) is now the greatest threat to global public health. Herbal medicines and their derived natural products have drawn much attention in the treatment of COVID-19, but the detailed mechanisms by which natural products inhibit SARS-CoV-2 have not been elucidated. Here, we show that platycodin D (PD), a triterpenoid saponin abundant in Platycodon grandiflorum (PG), a dietary and medicinal herb commonly used in East Asia, effectively blocks the two main SARS-CoV-2 infection routes via lysosome- and transmembrane protease serine 2 (TMPRSS2)-driven entry. Mechanistically, PD prevents host entry of SARS-CoV-2 by redistributing membrane cholesterol to prevent membrane fusion, which can be reinstated by treatment with a PD-encapsulating agent. Furthermore, the inhibitory effects of PD are recapitulated by the pharmacological inhibition or gene silencing of NPC1, which is mutated in patients with Niemann–Pick type C (NPC) displaying disrupted membrane cholesterol distribution. Finally, readily available local foods or herbal medicines containing PG root show similar inhibitory effects against SARS-CoV-2 infection. Our study proposes that PD is a potent natural product for preventing or treating COVID-19 and that briefly disrupting the distribution of membrane cholesterol is a potential novel therapeutic strategy for SARS-CoV-2 infection.Subject terms: Viral infection, Lipid signalling  相似文献
