Adherence of uropathogenic Escherichia coli to host tissue is mediated by pili, which are hair-like protein structures extending from the outer cell membrane of the bacterium. The chaperones FimC and PapD are key components in pilus assembly since they catalyse folding of subunits that are incorporated in type 1 and P pili, respectively, and also transport the subunits across the periplasmic space. Recently, compounds that inhibit pilus biogenesis and interfere with chaperone-subunit interactions have been discovered and termed pilicides. In this paper NMR spectroscopy was used to study the interaction of different pilicides with PapD and FimC in order to gain structural knowledge that would explain the effect that some pilicides have on pilus assembly. First relaxation-edited NMR experiments revealed that the pilicides bound to the PapD chaperone with mM affinity. Then the pilicide-chaperone interaction surface was investigated through chemical shift mapping using 15N-labelled FimC. Principal component analysis performed on the chemical shift perturbation data revealed the presence of three binding sites on the surface of FimC, which interacted with three different classes of pilicides. Analysis of structure-activity relationships suggested that pilicides reduce pilus assembly in E. coli either by binding in the cleft of the chaperone, or by influencing the orientation of the flexible F1-G1 loop, both of which are part of the surface by which the chaperone forms complexes with pilus subunits. It is suggested that binding to either of these sites interferes with folding of the pilus subunits, which occurs during formation of the chaperone-subunit complexes. In addition, pilicides that influence the F1-G1 loop also appear to reduce pilus formation by their ability to dissociate chaperone-subunit complexes.  相似文献

An approach is described for controlling the spatial organization of mammalian cells using ferromagnetic nanowires in conjunction with patterned micromagnet arrays. The nanowires are fabricated by electrodeposition in nanoporous templates, which allows for precise control of their size and magnetic properties. The high aspect ratio and large remanent magnetization of the nanowires enable suspensions of cells bound to Ni nanowires to be controlled with low magnetic fields. This was used to produce one- and two-dimensional field-tuned patterning of suspended 3T3 mouse fibroblasts. Self-assembled one-dimensional chains of cells were obtained through manipulation of the wires' dipolar interactions. Ordered patterns of individual cells in two dimensions were formed through trapping onto magnetic microarrays of ellipsoidal permalloy micromagnets. Cell chains were formed on the arrays by varying the spacing between the micromagnets or the strength of fluid flow over the arrays. The positioning of cells on the array was further controlled by varying the direction of an external magnetic field. These results demonstrate the possibility of using magnetic nanowires to organize cells.  相似文献

A mathematical model is proposed for the process of vacuum superplasticforming. The model exploits the fact that in most industrialapplications the sheet aspect ratio (thickness/sheet width)is small. After an initial consideration of some of the moregeneral properties and the literature of superplastic materials,the elastic/plastic deformation of an internally-inflated thin-walledcylinder is examined. Plates of arbitrary geometry are thenconsidered. A quasisteady model in which the sheet moves througha sequence of steady states is developed. Some simplified closed-formsolutions are examined, but for general cases a system of nonlinearpartial differential equations must be solved numerically. Anefficient and accurate semi-explicit numerical scheme is proposedand a simplified stability analysis is presented; the methodis then used to compute properties of superplastic vacuum mouldedsheets in a number of practically motivated cases.  相似文献

Voltammetric, photo-physical and photo-electrochemical properties of the Dawson polyoxometalate anions alpha-[S(2)M(18)O(62)](4-) (M = Mo, W) are presented, both in the presence and absence of a series of [Ru(II)L(n)](+/2+) cations [L(n) = (bpy)(3), (bpy)(2)(Im)(2), (bpy)(2)(dpq), (bpy)(2)(box) and (biq)(2)(box)]. Electrochemical processes for both the anion and Ru(II/III) couples were detected in solutions of the salts [Ru(II)L(n)](2)[S(2)M(18)O(62)] in dimethylformamide (0.1 M Bu(4)NPF(6)) by both cyclic and hydrodynamic voltammetries. Responses were also detected when the solid salts were adhered to the surface of a glassy carbon electrode in contact with an electrolyte in which they are insoluble (CH(3)CN; 0.1M Bu(4)NPF(6)). Photolysis experiments were performed on solutions of the salts [R(4)N](4)[S(2)M(18)O(62)] (R = n-butyl or n-hexyl) and [Ru(II)L(n)](2)[S(2)M(18)O(62)] at 355 and 420 nm in dimethylformamide and acetonitrile in the presence and absence of benzyl alcohol (10% v/v). When associated with [Ru(bpy)(3)](2+), the molybdate anion exhibited a large increase in the quantum yield for photo-reduction at 420 nm. The quantum yield for the tungstate analogue was lower but the experiments again provided clear evidence for sensitization of the photo-reduction reaction in the visible spectral region. The origin of this sensitization is ascribed to the new optical transition observed around 480 nm in static ion clusters {[Ru(bpy)(3)][S(2)M(18)O(62)]}(2-) and {[Ru(bpy)(3)](2)[S(2)M(18)O(62)]} present in solution. Measurable photocurrents resulted from irradiation of solutions of the anions with white light in the presence of the electron donor dimethylformamide. Evidence is also presented for possible quencher-fluorophore interactions in the presence of certain [Ru(II)L(n)](+) cations.  相似文献

We report on the first experimental observation of a new threshold behavior observed in the 5(2)G partial channel in photodetachment of K(-). It arises from the repulsive polarization interaction between the detached electron and the residual K(5(2)G) atom, which has a large negative dipole polarizability. In order to account for the observation in the K(5(2)G) channel, we have developed a semiclassical model that predicts an exponential energy dependence for the cross section. The measurements were made with collinear laser-ion beams and a resonance ionization detection scheme.  相似文献

The true potential energy curves forX 1 Σ +, a3 π r,A 1 π,e 3 Σ ,E 1 Σ +,c 3 π i states of astrophysically important molecule SiO, whose spectra were observed in the sunspots, have been constructed using the method of Lakshman and Rao. Dissociation energy and ionization potential have been estimated as 65,350 cm−1 and 92,854 cm−1 respectively and found to be in good agreement with the values cited in literature. The true potential energy curves forX 1 Σ +,D 1 π andE 1 Σ + states of SiS molecule have been constructed by the same method and its dissociation energy in the ground state has been estimated using Hulburt-Hirschfelder potential function as 54,765 cm−1 in good agreement with 53,250.9 cm−1 given by Herzberg.  相似文献

Solid bisphenol-A epoxy resin of medium molecular mass was cured using a Lewis acid initiator (ytterbium(III) trifluoromethanesulfonate) in three different proportions (0.5, 1 and 2 phr). A kinetic study was performed in a differential scanning calorimeter. The complete kinetic triplet was determined (activation energy, pre-exponential factor, and integral function of the degree of conversion) for each system. A kinetic analysis was performed with an integral isoconversional procedure (free model), and the kinetic model was determined both with the Coats-Redfern method (the obtained isoconversional value being accepted as the effective activation energy) and through the compensation effect. All the systems followed the same isothermal curing model simulated from non-isothermal ones. The growth-of-nuclei Avrami kinetic model A3/2 has been proposed as the polymerization kinetic model. The addition of initiator accelerated the reaction especially when 2 phr was added. 0.5 and 1 phr showed very few kinetic differences between them.  相似文献
