探索并建立了一种快速、 简便且高通量定量磷酸化蛋白质组的策略, 即采用连续互补的磷酸化富集方法SMOAC(Sequential enrichment of metal oxide affinity chromatography)结合TMT(Tandem mass tag)标记技术定量磷酸化蛋白质组学. 以3例经紫草素处理的及3例正常的人肝癌 HepG2 细胞为实验材料, 经Trypsin酶解后的肽段用TMT10-plex试剂进行等量标记, 标记肽段先经TiO2富集, 收集包含磷酸化肽段的洗脱液, 接着用次氮基三醋酸铁(Fe-NTA)对TiO2的流穿液和清洗液进行二次富集, 再次收集包含磷酸化肽段的洗脱液. 整个实验流程做2组, 对其中一组的2次洗脱液分别分析, 另一组的2次洗脱液合并分析. 在SMOAC的2次洗脱液合并分析中鉴定到4263个磷酸化蛋白上超过13000条磷酸化肽, 富集特异性>97%, 其中被定量的磷酸化蛋白为3848个, 占总鉴定量的90%以上. 研究结果表明, SMOAC 能够有效提高磷酸化肽段的鉴定效率, 且能与TMT等量标记试剂结合, 实现对少量蛋白样品的磷酸化蛋白定量分析.  相似文献

Lotus (Nelumbo nucifera) seeds are widely consumed as functional food or herbal medicine, of which cotyledon (CL) is the main edible part, and lotus plumule (LP) is commonly utilized in traditional Chinese medicine. However, few studies have been conducted to investigate the chemical components of CL and LP in dry lotus seeds, not to mention the comparison between wild and domesticated varieties. In this study, a widely targeted metabolomics approach based on Ultra Performance Liquid Chromatography-electrospray ionization-Tandem mass spectrometry (UPLC-ESI-MS/MS) was utilized to analyze the metabolites in CL and LP of China Antique (“CA”, a wild variety) and Jianxuan-17 (“JX”, a popular cultivar). A total of 402 metabolites were identified, which included flavonoids (23.08% to 27.84%), amino acids and derivatives (14.18–16.57%), phenolic acids (11.49–12.63%), and lipids (9.14–10.95%). These metabolites were classified into ten clusters based on their organ or cultivar-specific characters. Most of these metabolites were more abundant in LP than in CL for both varieties, except for metabolites belonging to organic acids and lipids. The analysis of differentially accumulated metabolites (DAMs) demonstrated that more than 25% of metabolites detected in our study were DAMs in CL and LP comparing “JX” with “CA”, most of which were less abundant in “JX”, including 35 flavonoids in LP, 23 amino acids and derivatives in CL, 7 alkaloids in CL, and 10 nucleotides and derivatives in LP, whereas all of 11 differentially accumulated lipids in LP were more abundant in “JX”. Together with the fact that the seed yield of “JX” is much higher than that of “CA”, these results indicated that abundant metabolites, especially the functional secondary metabolites (mainly flavonoids and alkaloids), were lost during the process of breeding selection.  相似文献

We present the first colloidal synthesis of highly uniform single-crystalline Bi19S27Br3 nanowires (NWs) with a mean diameter of ∼9 nm and tunable lengths in the range of 0.15–2 μm in the presence of foreign metal ions (Al3+). The Al3+ ions not only control the growth of NWs, but also achieve species transformation, i.e., from Bi2S3 to Bi19S27Br3, and are not present in the resulting NWs. This colloidal chemistry approach can be expanded to prepare a family of single-crystalline Bi19S27(Br3–x,Ix) alloyed NWs with controlled compositions (0 ≤ x ≤ 3). Interestingly, these alloyed NWs show an unusual composition-independent band gap of ∼0.82 eV, and theoretical calculations indicate that this phenomenon comes from the very minor contributions of the halogens to the valence band maximum and conduction band minimum. The photodetectors made of Bi19S27(Br3–x,Ix) alloyed NWs show a pronounced photoresponse with high stability and reproducibility, which makes the NWs potentially useful candidates in optoelectronic devices.  相似文献

采用溶液法制备聚乳酸(PLA)薄膜,并利用偏光显微镜在线研究了不同温度及不同牵引速率下,单根蚕丝纤维(SF)诱导PLA柱晶的形成,并利用显微红外光谱表征柱晶结构.结果表明,当牵引速率大于或等于临界速率时,纤维表面有连续的柱晶生成,反之柱晶不连续;当牵引速率大于临界饱和速率时,柱晶的结晶度和取向度分别趋于稳定值;在等温条件下,随着牵引速率的增大,柱晶的成核诱导期缩短,晶体的成核密度增大,取向度增加;当牵引速率一定时,与130℃相比,温度为120℃时形成柱晶的成核诱导期较短,成核密度更大,结晶度更高,柱晶层的生长更快.  相似文献

To better utilize the infrared(IR)region in sunlight for photovoltaic devices(PVs),upconversion nanoparticles(UCNPs)have been proposed to improve power conversion efficiency(PCE).However,researchers recently have found that the upconversion(UC)effect is negligible in PVs performance improvement for their ultra-low UC photoluminescence quantum yields of UCNPs solid film,while the real mechanism of UCNPs in PVs has not been clearly studied.Herein,based on the material inorganic perovskitesγ-CsPbI3,NaYF4:20%Yb3+,2%Er3+UCNPs were integrated into different transport layer to optimize device performance.Compared with reference device,the short-circuit current density and PCE of optimized device reached 20.87 mA/cm2(20.39 mA/cm2)and 18.34%(17.72%),respectively,without sacrificing open-circuit voltage and filling factor.Further experimental characterizations verified that the improved performance was attributable to enhanced visible light absorption instead of IR.To theoretically explain the statement,the light field distribution in device was simulated and the absorption in different layers was calculated.The results revealed that the introduction of UCNPs with different refractive index from other layers caused light field disturbance,and improved visible light captured by γ-CsPbI3.Importantly,through experiments and theoretical calculation,the research deeply explored the potential mechanism of UCNPs in optimizing PVs performance.  相似文献

Despite the fact that d-band center theory links the d electron structure of transition metals to their catalytic activity, it is yet unknown how the synergistic effect of multi-d electrons impacts catalytic performance. Herein, novel LaNi1-xCoxRu intermetallics containing 5d, 4d, and 3d electrons were prepared. In these compounds, the 5d orbital of La transfers electrons to the 4d orbital of Ru, which provides adsorption sites for H*. The 3d orbitals of Ni and Co interact with the 5d and 4d orbitals to generate an anisotropic electron distribution, which facilitates the adsorption and desorption of OH*. The synergistic effect of multi-d electrons ensures efficient catalytic activity. The optimized LaNi0.5Co0.5Ru has an overpotential of 43mV at 10 mA cm−2 for alkaline electrocatalytic hydrogen evolution reaction. Beyond offering a variety of new electrocatalysts, this work reveals the multi-d electron synergy in promoting catalytic reaction.  相似文献

Due to the significance of corresponding products,enantioselective borylative cyclization reactions have been studied intensively in recent years.Many groups have developed efficient methods to transform unsaturated system into asymmetric cyclic organoboron compounds with the ring-size range from three-membered to six-membered in general.Notably,in some cases,fused rings which contain more than two contiguous chiral centers could be obtained by this kind of strategies.This review summarized and reviewed the recent advances in this field and classified these work according to the species of metal catalysts.  相似文献
