以廉价的异香草醛和2,4,6-三羟基苯乙酮为起始原料,经过C-异戊烯基化、选择性甲基化、甲氧甲基化、羟醛缩合、去保护基、催化环化以及脱除甲基等步骤,分别以13.3%和23%的总收率首次完成了天然异戊烯基黄烷酮Glovanon(1)和5-O-methylglovanon(2)的全合成.合成的关键步骤是2,4,6-三羟基苯乙酮的单C-异戊烯基化.所有新化合物的结构都经过1H NMR,IR,MS确认.抑菌活性研究表明,两种新合成的化合物对藤黄微球菌(M.luteus)和大肠杆菌(E.coli)均有良好的抑制作用.  相似文献

Traditional real-time polymerase chain reaction (PCR) requires a purified DNA sample for PCR amplification and detection. This requires PCR tests be conducted in clean laboratories, and limits its applications for field tests. This work developed a method that can carry out DNA purification, amplification and detection in a single PCR tube. The polypropylene PCR tube was first treated with chromic acid and peptide nucleic acids (PNA) as DNA-capturer were immobilized on the internal surface of the tube. Cauliflower mosaic virus 35S (CaMV-35S) promoter in the crude extract was hybridized with the PNA on the tube surface, and the inhibitors, interfering agents and irrelevant DNA in the crude extract were effectively removed by rinsing with buffer solutions. The tube that has captured the target DNA can be used for the following real-time PCR (RT-PCR). By using this approach, the detection of less than 2500 copies of 35S plasmids in a complex sample could be completed within 3 hours. Chocolate samples were tested for real sample analysis, and 35S plasmids in genetically modified chocolate samples have been successfully identified with this method in situ. The novel One-PCR-tube method is competitive for commercial kits with the same time and simpler operation procedure. This method may be widely used for identifying food that contains modified DNA and specific pathogens in the field.  相似文献

A facile approach for the first total synthesis of two naturally occurring geranylated flavonoids, (±)‐puyanin ( 1 ) and (±)‐4′‐O‐methylbonannione ( 2 ) has been obtained with total yields of 27% and 17.8%, respectively. The key steps were regioselective cyclization of geranylated trihydroxychalcone and regioselective geranylation of 2,4,6‐trihydroxyacetophenone.  相似文献

The aim of this paper is to discuss the global existence and uniqueness of strong solution for a class of the isentropic compressible Navier-Stokes equations with non-Newtonian in one-dimensional bounded intervals. We prove two global existence results on strong solutions of the isentropic compressible Navier-Stokes equations. The first result shows only the existence, and the second one shows the existence and uniqueness result based on the first result, but the uniqueness requires some compatibility condition.  相似文献

Manipulating recognition and transport at the nanoscale holds great promise for technological breakthroughs in energy conversion, catalysis, and information processing. Living systems evolve specialized membrane proteins (MPs) embedded in lipid bilayers to exquisitely control communications across the insulating membrane boundaries. Harnessing MP functions directly in synthetic systems opens up enormous opportunities for nanotechnology, but there exist fundamental challenges of how to address the labile nature of lipid bilayers that renders them of inadequate value under a broad range of harsh non-biological conditions, and how to reconstitute MPs coherently in two or three dimensions into non-lipid-based artificial membranes. Here we show that amphiphilic block copolymers can be designed to direct proteorhodopsin reconstitution and formation of hierarchically ordered proteopolymer membrane arrays spontaneously, even when the membrane-forming polymer blocks are in entangled states. These findings unfold a viable approach for the development of robust and chemically versatile nanomembranes with MP-regulated recognition and transport performance.  相似文献

A rapid, sensitive and accurate high performance liquid chromatography method using tandem mass spectrometry detection for hydralazine in BALB/C mouse plasma and brain was developed and validated. The method involved a derivatization with 2,4-pentanedione at 50 °C for 1 h, and a step of solid phase extraction to purify and concentrate hydralazine derivative. Chromatographic separation was carried out on an Agilent ZORBAX SB-C18 column by elution with methanol?C0.01 mol L?1 ammonium acetate (60:40, v/v). The multiple reaction monitoring transition used for quantification was m/z 225.2 ?? 129.5 in the electrospray positive ionization mode. Good linearity was obtained over the concentration range of 10?C200 ng mL?1. The limits of detection were 0.49 and 1.05 ng mL?1 for hydralazine in mouse plasma and brain, respectively. The limits of quantitation were 1.5 and 3.18 ng mL?1 for hydralazine in mouse plasma and brain, respectively. Sample analysis time was 6 min including sample separation. The method was successfully applied to a pharmacokinetic study following intraperitoneal injection of hydralazine in BALB/C mice at the dose of 20 mg kg?1.  相似文献
