辽宁省位于中国的东北地区,主要生产粮食和饲料.对辽宁省农户的保险意愿调查问卷进行了统计分析.结果表明:性别、受教育程度、收入、灾害损失和对保险的了解程度对农户在农业保险意愿上有影响.女性相对于男性,更不愿意参加农业保险;教育程度高和贫穷的农户不愿意参保.以往受灾损失大的农户也不愿意参保,这样,如果政府不能合理的设定保险品种和保险价格可能会出现逆向选择问题.  相似文献

This paper demonstrates the development of an analytical method for detecting steroid hormones by coupling HPLC to electrochemical detection, using a nickel‐modified glassy carbon electrode. The method was evaluated in terms of sensitivity, linear dynamic range, limit of detection, and response stability. The developed method exhibited good figures of merit for the steroid hormones studied with no evidence of electrode fouling. As an example, the limit of detection (S/N=3) for E3 was 0.10 µM and the response precision (n=5) was 0.6 %. The application of the method for the analysis of a real river water sample is demonstrated.  相似文献

The coupled reference interaction site model-hyper-netted chain (RISM-HNC)/ simulation methodology determines solvation free energies as a function of the set of all radial distribution functions of solvent atoms about atomic solute sites. These functions are determined from molecular dynamics (MD) or Monte Carlo (MC) simulations rather than from solving the RISM and HNC equations iteratively. Previous applications of the method showed that it can predict relative free energies of solvation for small solutes accurately. However, the errors scale with the system size. In this study, we propose the use of the hard-sphere free energy as the reference and a linear response approximation to improve the performance, i.e., accuracy and robustness, of the method, particularly removing the size dependency of the error. The details of the new formalism are presented. To validate the proposed formalism, solvation free energies of N-methylacetamide and methylamine are computed using the new RISM-HNC-based expressions in addition to a linear response expression, which are compared to previous thermodynamic integration and thermodynamic perturbation results performed with the same force field. Additionally, free energies of solvation for cyclohexane, pyridine, benzene and derivatives, and other small organic molecules are calculated and compared to experimental values.  相似文献

Although peptide‐based therapeutics are finding increasing application in the clinic, extensive structural modification is typically required to prevent their rapid degradation by proteases in the blood. We have evaluated the ability of erythrocytes to serve as reservoirs, protective shields (against proteases), and light‐triggered launch pads for peptides. We designed lipidated peptides that are anchored to the surface of red blood cells, which furnishes a protease‐resistant environment. A photocleavable moiety is inserted between the lipid anchor and the peptide backbone, thereby enabling light‐triggered peptide release from erythrocytes. We have shown that a cell‐permeable peptide, a hormone (melanocyte stimulating hormone), and a blood‐clotting agent can be anchored to erythrocytes, protected from proteases, and photolytically released to create the desired biological effect.  相似文献

Chimeric antigen receptor T (CAR‐T) cells have demonstrated promising results against hematological malignancies, but have encountered significant challenges in translation to solid tumors. To overcome these hurdles, we have developed a switchable CAR‐T cell platform in which the activity of the engineered cell is controlled by dosage of an antibody‐based switch. Herein, we apply this approach to Her2‐expressing breast cancers by engineering switch molecules through site‐specific incorporation of FITC or grafting of a peptide neo‐epitope (PNE) into the anti‐Her2 antibody trastuzumab (clone 4D5). We demonstrate that both switch formats can be readily optimized to redirect CAR‐T cells (specific for the corresponding FITC or PNE) to Her2‐expressing tumor cells, and afford dose‐titratable activation of CAR‐T cells ex vivo and complete clearance of the tumor in rodent xenograft models. This strategy may facilitate the application of immunotherapy to solid tumors by affording comparable efficacy with improved safety owing to switch‐based control of the CAR‐T response.  相似文献

Chitosan, a biopolymer found in the exoskeletons of shellfish, has been shown to be antibacterial, biodegradable, osteoconductive, and has the ability to promote organized bone formation. These properties make chitosan an ideal material for use as a bioactive coating on medical implant materials. In this study, coatings made from 86.4% de-acetylated chitosan were bound to implant-quality titanium. The chitosan films were bound through a three-step process that involved the deposition of 3-aminopropyltriethoxysilane (APTES) in toluene, followed by a reaction between the amine end of APTES with gluteraldehyde, and finally, a reaction between the aldehyde end of gluteraldehyde and chitosan. Two different metal treatments were examined to determine if major differences in the ability to bind chitosan could be seen. X-ray photoelectron spectroscopy (XPS) was used to examine the surface of the titanium metal and to study the individual reaction steps. The changes to the titanium surface were consistent with the anticipated reaction steps, with significant changes in the amounts of nitrogen, silicon, and titanium that were present. It was demonstrated that more APTES was bound to the piranha-treated titanium surface as compared to the passivated titanium surface, based on the amounts of titanium, carbon, nitrogen, and silicon that were present. The metal treatments did not affect the chemistry of the chitosan films. Using toluene to bond APTES on titanium surfaces, rather than aqueous solutions, prevented the formation of unwanted polysiloxanes and increased the amount of silane on the surface for forming bonds to the chitosan films. Qualitatively, the films were more strongly attached to the titanium surfaces after using toluene, which could withstand the ultrahigh vacuum environment of XPS, as compared to the aqueous solutions, which were removed from the titanium surface when exposed to the ultrahigh vacuum environment of XPS.  相似文献

A donor-acceptor system is presented in which the electron-transfer rates can be sensitively controlled by means of excitation wavelength and temperature. The electron donor is a butadiyne-linked zinc porphyrin dimer that is connected to a C(60) electron acceptor. The broad distribution of conformations allowed by the butadiyne linker makes it possible to selectively excite perpendicular or planar donor conformers and thereby prepare separate initial states with driving forces for electron transfer that differ by almost 0.2 eV. This, as well as significant differences in electronic coupling, leads to distinctly different rate constants for electron transfer, which in consequence can be controlled by changing excitation wavelength. By extending the system with a secondary donor (ferrocene), a second, long-range charge-separated state can be formed. This system has been used to test the influence of conformational heterogeneity on electron transfer mediated by the porphyrin dimer in the ground state. It was found that if the dimer is forced to a planar conformation by means of a bidentate ligand, the charge recombination rate increased by an order of magnitude relative to the unconstrained system. This illustrates how control of conformation of a molecular wire can affect its behaviour.  相似文献

A compact hand-held heated fluorometric instrument for performing real-time isothermal nucleic acid amplification and detection is described. The optoelectronic instrument combines a Printed Circuit Board/Micro Electro Mechanical Systems (PCB/MEMS) reaction detection/chamber containing an integrated resistive heater with attached miniature LED light source and photo-detector and a disposable glass waveguide capillary to enable a mini-fluorometer. The fluorometer is fabricated and assembled in planar geometry, rolled into a tubular format and packaged with custom control electronics to form the hand-held reactor. Positive or negative results for each reaction are displayed to the user using an LED interface. Reaction data is stored in FLASH memory for retrieval via an in-built USB connection. Operating on one disposable 3 V lithium battery >12, 60 min reactions can be performed. Maximum dimensions of the system are 150 mm (h) × 48 mm (d) × 40 mm (w), the total instrument weight (with battery) is 140 g. The system produces comparable results to laboratory instrumentation when performing a real-time nucleic acid sequence-based amplification (NASBA) reaction, and also displayed comparable precision, accuracy and resolution to laboratory-based real-time nucleic acid amplification instrumentation. A good linear response (R2 = 0.948) to fluorescein gradients ranging from 0.5 to 10 μM was also obtained from the instrument indicating that it may be utilized for other fluorometric assays. This instrument enables an inexpensive, compact approach to in-field genetic screening, providing results comparable to laboratory equipment with rapid user feedback as to the status of the reaction.  相似文献

Based on work of Atkin and Swinnerton-Dyer on partition rank difference functions, and more recent work of Lovejoy and Osburn, Mao has proved several inequalities between partition ranks modulo 10, and additional results modulo 6 and 10 for the \(M_2\) rank of partitions without repeated odd parts. Mao conjectured some additional inequalities. We prove some of Mao’s rank inequality conjectures for both the rank and the \(M_2\) rank modulo 10 using elementary methods.  相似文献
