We used nearest‐neighbor searches in chemical space to improve the activity of the antimicrobial peptide dendrimer (AMPD) G3KL and identified dendrimer T7 , which has an expanded activity range against Gram‐negative pathogenic bacteria including Klebsiellae pneumoniae, increased serum stability, and promising activity in an in vivo infection model against a multidrug‐resistant strain of Acinetobacter baumannii. Imaging, spectroscopic studies, and a structural model from molecular dynamics simulations suggest that T7 acts through membrane disruption. These experiments provide the first example of using virtual screening in the field of dendrimers and show that dendrimer size does not limit the activity of AMPDs.  相似文献

This study was related to the investigation of the chemical fixation of carbon dioxide to a copolymer bearing epoxide and the application of the cyclic carbonate group containing copolymer‐to‐polymer blends. In the synthesis of poly[(2‐oxo‐1,3‐dioxolane‐4‐yl) methyl methacrylate‐co‐ethyl acrylate] [poly(DOMA‐co‐EA)] from poly(glycidyl methacrylate‐co‐ethyl acrylate) [poly(GMA‐co‐EA)] and CO2, quaternary ammonium salts showed good catalytic activity. The films of poly(DOMA‐co‐EA) with poly(methyl methacrylate) (PMMA) or poly(vinyl chloride) (PVC) blends were cast from N,N′‐dimethylformamide solution. The miscibility of the blends of poly(DOMA‐co‐EA) with PMMA or PVC have been investigated both by DSC and visual inspection of the blends. The optical clarity test and DSC analysis showed that poly(DOMA‐co‐EA) containing blends were miscible over the whole composition range. The miscibility behaviors were discussed in terms of Fourier transform infrared spectra and interaction parameters based on the binary interaction model. © 2001 John Wiley & Sons, Inc. The surface-enhanced Raman scatterings of dimethyl sulfide, diethyl sulfide and dimethyl disulfide have been investigated in silver sol. The dimethyl disulfide molecule decomposes on silver to the corresponding mercaptide implying facile cleavage of its S---S bond. The C---S bond in dialkyl monosulfide appears not to cleave on silver. For diethyl sulfide, the C2 conformation seems to be favorable on silver than other conformations.  相似文献

The studies with proteins are necessary to understand the biological effects of atmospheric pressure plasma (APP). Among proteins, those with transient metal ions play key roles in many biological events and they are very sensitive to environmental redox states. Iron-containing hemoglobin (Hb) is investigated in this study, after APP treatments under two environmental gas conditions of pure N2 and N2?+?O2 mixture. Monitoring the intensity change for absorption spectra could lead to a quantitative assessment of the effect of discharge plasma on Hb. Redox states of Hb are classified into five states including O2-bound Hb (oxy-Hb), deoxy-Hb, met-Hb, NO-bound Hb (NO-Hb), and hemichrome. Chemically generated reactive species and some scavengers are applied to understand the chemical reactions. Our experimental results confirm the complex chemical reactions of APP and suggest the possible use of Hb as a model protein for the visualization of APP biological effects.  相似文献

We demonstrate that the microwave-assisted protein enzymatic digestion (MAPED) method can be successfully applied to the mass spectrometric characterization of proteins captured on the affinity surfaces of protein chips. The microwave-assisted on-chip tryptic digestion method was developed using a domestic microwave, completing the on-chip proteolysis reaction in minutes, whereas the previous on-chip digestion methods by incubation took hours of incubation time. For the model protein chips, antibody-presenting surfaces were prepared, where anti-α-tubulin1 and antibovine serum albumin (BSA) were immobilized on self-assembled monolayers. The resulting digestion efficiency, displaying sequence coverages of 30 and 14% for α-tubulin1 and BSA, respectively, was comparable to the previous time-consuming incubation studies. It allowed the characterization of immunosensed proteins by MASCOT search using peptide mass fingerprinting. In an example of this method for protein chip applications, BSA naturally involved in fetal bovine serum was unambiguously identified on a model protein chip by imaging mass spectrometry. This work shows that biomass spectrometry techniques can be implemented for surface mass spectrometry and biochip applications. Along with recent advances in imaging mass spectrometry, this technique will provide a new opportunity for high-speed, and thus high-throughput in the future, label-free mass spectrometric assays using protein arrays.  相似文献

For the first time, employment of nicotinamide coenzyme NAD analogs has overcome the limitations of NAD in electrochemical regeneration. It has been shown that NAD analogs, APAD and PAAD, were electrochemically reduced more efficiently than original NAD and that the stability of their reduced products was also much higher than NADH.  相似文献

Polypyrrole (PPy) and poly(pyrrole-2,6-dimethyl-β-cyclodextrin) [P(Py-β-DMCD)] films prepared by potential cycling in aqueous acidic solutions on indium tin oxide (ITO)-coated glass and gold electrodes were studied by in situ UV-vis and Raman spectroscopy. Characteristic UV-vis and Raman bands were identified and their dependencies on the electrode potential have been discussed. Spectroelectrochemical results reveal differences both in the position of the spectral bands and their potential dependence for PPy and P(Py-β-DMCD) films indicating interactions between polymer chains and CDs during electropolymerization process. The films were also characterized by cyclic voltammetry and FT-IR spectroscopy.  相似文献

The effect of alcohol washing on the anatase-rutile transition of precipitated titanium oxide was investigated using X-ray powder diffraction, Fourier-transform IR spectroscopy, and thermogravimetry. Alcohol (butanol) rinsing accelerated the anatase-rutile transition of precipitated titanium oxide powder so that the onset temperature of transition decreased drastically from 800 degrees C for water-washed powder to 550 degrees C for alcohol-rinsed powder. Alternation of transition kinetics and mechanisms by rinsing media could be confirmed from the analysis of temperature and time dependence of rutile content. The attributability of the chemical state of anatase after crystallization, which contained H(2)O, OH, and organic residues, to the change of transition kinetics with alcohol rinsing will be discussed. Two mechanisms, the effect of residual organics and/or H(2)O(OH), could be suggested on the basis of analysis of the difference between chemical states of water-washed anatase and alcohol-rinsed powder.
