Water-compatible molecularly imprinted polymers (MIPs) were prepared in water-methanol systems for selective extraction and separation of ciprofloxacin from human urine samples. Molecular recognition properties, binding capability, and chromatographic applications of the MIPs were evaluated and the results revealed the obtained MIPs have high affinity for ciprofloxacin in aqueous environment and the selectivity can be easily controlled by adjusting the pH of mobile phase. After centrifugation, the urine samples were directly injected into the MIPs column and ciprofloxacin could be selectively retained on the column while other biological matrixes were quickly washed out. Good linearity was obtained from 0.1 to 100 mg L(-1) (r=0.999) with the relative standard deviations less than 3.6%. The limit of detection of the method was 0.03 microg/mL and the recoveries were more than 94.5% at three different concentrations. Moreover, by increasing the injection volume, the detection limits of the method could be improved more than 100-folds.  相似文献

Two new liquid chromatographic chiral stationary phases based on diastereomeric chiral crown ethers incorporating two different chiral units such as optically active 3,3'-diphenyl-1,1'-binaphthyl and tartaric acid unit were prepared. Between the two CSPs, one was much superior to the other especially in the resolution of tocainide and its analogues (for example, in the resolution of tocainide the separation factor, alpha, was 4.26 vs. 1.00 on the two CSPs). From these results, the two chiral units composing the two diastereomeric chiral crown ether moieties of the stationary phases were expected to show "matched" or "mismatched" effect on the chiral recognition according to their stereochemistry. The different chiral recognition abilities of the two CSPs were rationalized by the different three-dimensional structures of the two diastereomeric chiral crown ethers.  相似文献

As a part of our ongoing search for a safe and efficient anti-tumor vaccine, we attempted to determine whether the molecular nature of certain tumor antigens would influence immune responses against tumor cells. As compared with freeze-thawed or formaldehyde-fixed tumor antigens, heat-denatured tumor antigens elicited profound anti-tumor immune responses and greatly inhibited the growth of live tumor cells. The heat-denatured tumor antigens induced a substantial increase in the anti-tumor CTL response in the absence of any adjuvant material. This response appears to be initiated by strong activation of the antigen-presenting cells, which may recognize heat-denatured protein antigens. Upon recognition of the heat-denatured tumor antigens, macrophages and dendritic cells were found to acutely upregulate the expression of co-stimulatory molecules such as B7.2, as well as the secretion of inflammatory cytokines such as IL-12 and TNF-alpha. The results of this study indicate that heat-denatured tumor extracts might elicit protective anti-tumor adaptive immune responses and also raise the possibility that a safe and efficient adjuvant-free tumor vaccine might be developed in conjunction with a dendritic cell-based tumor vaccine.  相似文献

A unique benzannulation strategy for regioselective de novo synthesis of densely functionalized phenols is described. Through metal-mediated formal [2+2+1+1] cycloaddition of two different alkynes and two molecules of CO, a series of densely functionalized phenols were obtained. The benzannulation strategy allows efficient regioselective installation up to five different substituents on a phenol ring. The resulting phenols have a substitution pattern different from those obtained from Dötz and Danheiser benzannulations.  相似文献

We have examined both self-assembly and confinement effect in room-temperature ionic liquid (RTIL)-aluminum hydroxide hybrids (RAHs) to attain a fundamental understanding of special phenomena in nanoscale spaces as well as to design functional nanomaterials for practical applications. Phase-controlled one-dimensional (1D) RAHs were synthesized through a simple ionothermal process. The RAHs were hierarchically transformed in terms of the molecular structures, morphologies, and phases of the materials during the ionothermal process with respect to the concentration of RTIL. In addition to the hierarchical transformation, the RTIL/aluminum hydroxide nanohybrids revealed unexpected physical behaviors, including thermal transition variation of the RTIL in confined environments and a phase transition from nanosolid to nanoliquid affected by changes of the melting points. More importantly, intermolecular interaction induced-self-assembly and confinement effect of RTILs inside an integrated hybrid system, which have not been clearly explained to date, were analyzed by 2D infrared correlation spectroscopy (2D IR COS); dynamic behaviors of RTILs, i.e., sequentially spatial reorientation and kinetically conformational changes, were attributed to the interactions between RTILs and aluminum hydroxides. 2D IR COS offers a new way to interpret highly complex, veiled systems such as the formation mechanism of nanoparticles, biomineralization, self/supramolecular assembly, and nanoconfinement.  相似文献

Two new phenyl glycosides, mangliesides A and B (1, 2), a new ionol glycoside, manglieside C (3), two new lignan glycosides, mangliesides D and E (4, 5), were isolated from the leaves of Manglietia phuthoensis, along with two known lignans, 3-methoxymagnolol (6) and obovatol (7). Their structures were established by means of 1D and 2D NMR, electrospray ionization (ESI)-MS and HR-ESI-MS experiments. Among them, compounds 2 and 5 significantly (p<0.05) increased the growth and differentiation of osteoblastic MC3T3-E1 cells in vitro.  相似文献

The structure of a model liquid crystalline fluid confined between two microstructured substrates is studied through Monte Carlo simulations. A simple model for a structured substrate, similar in spirit to those used for rough walls of walls with grafted polymers, is introduced. It is found that varying the structure of the substrate, a transition in the alignment of the confined fluid, from parallel to perpendicular, is induced. For particular substrate structures, it is possible to induce tilted alignment in the confined fluid, the tilt angle being temperature dependent.  相似文献
