Based on techniques for single molecule imaging and nanomanipulation by optical tweezers, we have developed a new technique that allows simultaneous measurement of individual ATPase and mechanical reactions from a single myosin molecule during force generation. We show how the ATPase reaction couples to the mechanical reaction directly at the single molecule level. The results show that the myosin head can produce force even after releasing the bound nucleotide, probably ADP, suggesting that the chemical energy driven by ATP hydrolysis can be hysteretically stored in the myosin molecule. This view does not support a widely accepted hypothesis in which the force generation is tightly coupled to ligand dissociation.This paper was originally presented as an invited paper at the seventh Meeting on Near Field Optics, which was held on July 1, 1998 at Nagoya University, Nagoya, organized by the Research Group on Near Field Optics of the Optical Society of Japan, an affiliate of the Japan Society of Applied Physics.  相似文献

Bubble detectors which are commonly used as neutron detectors have been demonstrated through this study to be good detectors for registration of high energy heavy ion tracks. Large size bubble detectors made in China Institute of Atomic Energy were irradiated to heavy ions Ar and C up to 650 MeV/u and 400 MeV/u, respectively. Very clear features of stringy tracks of high energy heavy ions and their fragmentations are manifested and distinguishable. A single track created by a specific high energy heavy ion is composed of a line of bubbles, which is visible by naked eyes and retained for months wihhout reduction in size. The creation of heavy ion tracks in bubble detectors is governed by a threshold whose essence is approximately a critical value of energy loss rate (dE/dX)c similar to that of etch track detectors. Ranges of heavy ions in bubble detectors are apparent and predictable by existing formulas. Identification of high energy heavy ions and the applications to heavy ion physics, cosmic rays, exotic particles and cancer therapy monitoring are obviously promising. The experimental and theoretical aspects of high energy heavy ion tracks in bubble detectors as well as the expectable applications are presented and discussed.  相似文献

The purpose of the present paper is to demonstrate the potential of generalized two-dimensional (2D) near-infrared (NIR) correlation spectroscopy in studies of hydration and structure of proteins. We describe here two examples. The first example is concerned with heat denaturation process of ovalbumin in aqueous solutions and the second one deals with isomerizations with varying pH of serum albumin. New insight has been obtained into the hydration and unfolding process of secondary structures of ovalbumin and serum albumin by studying temperature- or pH-dependent correlation patterns in 2D synchronous and asynchronous spectra. Generalized 2D NIR correlation spectroscopy emphasizes spectral features not readily observable in conventional one-dimensional spectra, enabling to extract subtle but valuable structural changes concerning with the protein denaturation.  相似文献

It has been found that small mutations of certain genes are the definitive origin of many heritable disorders and cancers with striking development of recent molecular biology. Such new findings have taken close-up of the importance of gene mutation assays based on the difference of DNA base sequences in diagnostic or medical field Capillary electrophoresis can be a good candidate for an ideal method on such gene analysis, because the methods can be performed with trace amount of samples, high resolution and shorter running time. We have established an effect of oligonucleotide, which was introduced onto capillary inner surface, on the recognition of an overall sequence of sample DNA fragments as an affinity ligand.  相似文献

Infrared (IR) spectra and wide-angle X-ray diffraction (WAXD) patterns of poly(3-hydroxybutyrate) (PHB) and its copolyester, poly(3-hydroxybutyrate-co-3-hydroxyhexanoate), P(HB-co-HHx) (HHx = 12 mol%) were measured over a temperature range of 20 °C to higher temperatures (PHB, 185 °C, HHx = 12 mol%, 140 °C) to explore their structure change and thermal behavior and molecular interaction. The WAXD measurements revealed that the a lattice parameter increases significantly with temperature, while the b lattice parameter varies a little in the crystalline PHB and P(HB-co-HHx). It seems that the intermolecular interaction between the CO group of one helical structure and the CH3 group of the other helical structure decreases along the a axis of crystalline PHB and P(HB-co-HHx) with temperature. The temperature-dependent IR spectral variations were analyzed for the CH stretching, CO stretching, CH3 deformation, and C O C stretching variation regions, and bands characteristic of crystalline and amorphous parts were identified in each region. It was found from the anomalous frequencies of the CH3 asymmetric stretching and CO stretching bands of PHB and P(HB-co-HHx) and the X-ray crystallographic structure of PHB that there is an intermolecular interaction (C H…OC hydrogen bond) between the CO group and the CH3 group combining two helical structures in PHB and P(HB-co-HHx). In this review paper we discuss the role of the C H…OC hydrogen bonding and the crystal and lamella structure of PHB and P(HB-co-HHx) (HHx = 12 mol %) in comparison with the structure of Nylon.  相似文献

Fourier transform infrared (FTIR) spectroscopic measurements have been undertaken to estimate the conformational energies of poly(acrylic acid) (PAA) cast films in the temperature range of 40–130°C. The temperature dependence of the IR spectra in the C=O stretching region has been analyzed to yield the side-chain and backbone conformational energies. The estimated energies are close to those previously obtained by polarized Raman spectroscopic measurements for PAA solutions. Combining the FTIR value of conformational energy with the simplified rotational isomeric state (RIS) model proposed in the Raman analysis provides a persistence length in accordance with earlier SAXS experiments. Some natural plant extract formulations (NPEFs, also referred to as essential oils) used in organic farming have been shown to contain synthetic pesticides. We obtained samples of four NPEFs (Muso, Hekiro, Kensogen-Ten, and Nurse Green) that were contaminated with the synthetic pyrethroid cypermethrin, and we used gas chromatography coupled with combustion, cryofocusing, and isotope ratio mass spectrometry to determine the stable carbon isotope ratios (δ13C) for the cypermethrin in the four NPEF samples, as well as in ten cypermethrin reagents and two commercial pesticide formulations (Agrothrin emulsion and Agrothrin water-dispersible powder). Our goal was to identify the source of the cypermethrin in the NPEFs. Cryofocusing markedly sharpened the cypermethrin peak and thus improved the accuracy and precision of the determined δ13C values. The δ13C values (±?SD) of the 16 cypermethrin samples ranged from ?28.3?±?0.2 to ?24.5?±?0.2?‰. Surprisingly, the four NPEFs showed similar δ13C values (?26.8 to ?27.3?‰), suggesting that the cypermethrin in all the samples came from the same source (either the same chemical reaction or the same primary material). This possibility was supported by previously published results. In addition, the δ13C values of the two commercial pesticides were similar to the values for the NPEFs, suggesting that the commercial pesticides had been diluted and sold as NPEFs.  相似文献

This review paper reports near-infrared (NIR) imaging studies using a newly-developed NIR camera, Compovision. Compovision can measure a significantly wide area of 150 mm×250 mm at high speed of between 2 and 5 s. It enables a wide spectral region measurement in the 1 000~2 350 nm range at 6 nm intervals. We investigated the potential of Compovision in the applications to industrial problems such as the evaluation of pharmaceutical tablets and polymers. Our studies have demonstrated that NIR imaging based on Compovision can solve several issues such as long acquisition times and relatively low sensitivity of detection. NIR imaging with Compovision is strongly expected to be applied not only to pharmaceutical tablet monitoring and polymer characterization but also to various applications such as those to food products, biomedical substances and organic and inorganic materials.  相似文献
