The leaves of a tropical plant, Mitragyna speciosa KORTH (Rubiaceae), have been traditionally used as a substitute for opium. Phytochemical studies of the constituents of the plant growing in Thailand and Malaysia have led to the isolation of several 9-methoxy-Corynanthe-type monoterpenoid indole alkaloids, including new natural products. The structures of the new compounds were elucidated by spectroscopic and/or synthetic methods. The potent opioid agonistic activities of mitragynine, the major constituent of this plant, and its analogues were found in in vitro and in vivo experiments and the mechanisms underlying the analgesic activity were clarified. The essential structural features of mitragynines, which differ from those of morphine and are responsible for the analgesic activity, were elucidated by pharmacological evaluation of the natural and synthetic derivatives. Among the mitragynine derivatives, 7-hydroxymitragynine, a minor constituent of M. speciosa, was found to exhibit potent antinociceptive activity in mice.  相似文献

We have investigated terahertz emission due to dynamical electron transport in wide-miniband GaAs/Al(0.3)Ga0.7As superlattices. By noting that the time-domain THz emission spectroscopy inherently measures the step-response of the electron system to the bias electric field, the obtained THz spectra were compared with the high-frequency conductivities predicted for miniband transport. Excellent agreement between theory and experiment strongly supports that the THz gain due to Bloch oscillating electrons persists at least up to 1.7 THz. It was also found that Zener tunneling into the second miniband sets the high-frequency limit to the THz gain for the samples studied here.  相似文献

We examined the proton relaxation times in vitro in various neurological diseases using experimental and clinical materials, and consequently obtained significant results for making a fundamental analysis of magnetic resonance imaging (MRI) as followings. 1) In the brain edema and cerebral infarction, T1 prolonged and T2 separated into two components, one fast and one slow. Prolongation of T1 referred to the volume of increased water in tissue. The slow component of T2 reflects both the volume and the content of increased edema fluid in tissue. 2) In the edematous brain tissue with the damaged Blood-Brain-Barrier (BBB), the slow component of T2 became shorter after the injection of Mn-EDTA. Paramagnetic ion could be used as an indicator to demonstrate the destruction of BBB in the brain. 3) After the i.v. injection of glycerol, the slow component of T2 became shorter in the edematous brain with the concomitant decrease of water content. The effects of therapeutic drug could be evaluated by the measurement of proton relaxation times. 4) Almost all tumor tissue showed a longer T1 and T2 values than the normal rat brain, and many of them showed two components in T2. It was difficult to determine the histology of tumor tissue by the relaxation time alone because of an overlap of T1 and T2 values occurred among various types of brain tumors. 5) In vivo T1 values of various brain tumor were calculated from the data of MRIs by zero-crossing method, and they were compared with the in vitro T1 values which were measured immediately after the surgical operation. Though the absolute value did not coincide with each other due to differences in magnetic field strength, the tendency of the changes was the same among all kinds of tumors. It is concluded that the fundamental analysis of proton relaxation times is essentially important not only for the study of pathophysiology in many diseases but also for the interpretation of clinical MRI.  相似文献

The supramolecular chemistry of acyclic oligopyrrole derivatives mainly reported by the author’s group in the last four years has been summarized in this review. The author has demonstrated the “first step” to construct the new materials and concepts based on the new molecular systems consisting of pyrrole rings, which form the complexes, assemblies, and organized structures, by using noncovalent interactions such as metal coordination, hydrogen bonding, and π–π interaction. Acyclic π-conjugated oligopyrroles have exhibited not only host–guest binding behaviors in solutions but also the formation of, for example, (i) metal coordination polymers to give emissive colloidal spheres, (ii) solid-state assemblies of acyclic π-conjugated anion receptors and their anion complexes, (iii) anion-responsive supramolecular gels from the receptors with aliphatic chains, and (iv) solvent-assisted organized structures like vesicles derived from amphiphilic anion receptors.  相似文献

Bidipyrrin‐bridged macrocycles, prepared from NiII‐bridged dipyrrin‐based nanorings by intramolecular oxidative biaryl coupling reactions, yielded [2+4]‐type ZnII‐assisted stable twisted‐ring dimers comprising two double helices. These [2+4]‐type metal complexes can be optically resolved by chiral HPLC and exhibit tunable electronic and optical properties as a result of spring‐like motions. The double helices behave as glue to connect two macrocycles and as the screws of hinges to form thermally responsive synchronized spring systems.  相似文献
