An optically and thermally responsive boron dipyrromethene (BODIPY) dye, namely, meso‐2‐(9,10‐dihydro‐9,10‐ethanoanthracene‐11,12‐dione) (DK)‐linked, bicyclo[2.2.2]octadiene (BCOD)‐fused BODIPY ( BCOD‐DK ), was synthesized. The weakly luminous structure of BCOD‐DK can be changed quantitatively to that of the strongly fluorescent BODIPY BCOD‐Ant by optical excitation at the DK unit, which induces double decarbonylation of the DK unit to give an anthracene unit. The solvent effect on the fluorescence properties of BCOD‐DK suggests that the dramatic change in fluorescence intensity is controlled by intramolecular electron transfer from the BODIPY moiety to the meso‐DK substituent. BCOD‐DK is converted to meso‐ DK benzene‐fused BODIPY ( Benzo‐DK ) by heating at 220 °C with 64–70 nm redshift of absorption and fluorescence peaks without changing the fluorescence quantum yield of ΦF=0.08 in dichloromethane. Benzo‐DK can be converted to strongly fluorescent meso ‐ anthracene benzene‐fused BODIPY Benzo‐Ant by optical excitation. Thus, BCOD‐DK can show four different optical performances simply by irradiation and heating, and hence may be applicable for optical data storage and security data encryption.  相似文献

The addition of iron or boron and/or nitrogen, up to 20?%, to amorphous carbon with a density of about 2.0?gm/cm3 was studied using density functional theory. The bulk cohesive energy decreases with increasing iron, nitrogen, or boron concentration. The decrease is largest for iron and smallest for boron. The trends in the bulk moduli are consistent with the cohesive energies. The optical properties (absorbance and reflectivity) of the samples with nitrogen and/or boron added are very similar to those of the original amorphous carbon. Addition of iron results in larger, energy dependent, changes when compared with either boron or nitrogen. The effect of dopants on low-density amorphous carbon shows some differences with those for higher density amorphous carbon.  相似文献

A potent inhibitor for Vibrio cholerae neuraminidase (VCNA) was developed by using a novel two‐step strategy, a target amino acid validation using mechanism‐based labeling information, and a potent inhibitor search using a focused library. The labeling information suggested the hidden dynamics of a loop structure of VCNA, which can be a potential target of the novel inhibitor. A focused library composed of 187 compounds was prepared from a 9‐azide derivative of 2,3‐dehydro‐N‐acetylneuraminic acid (DANA) to interrupt the function of the loop of the labeled residues. Inhibitor 3c showed potent inhibition properties and was the strongest inhibitor with FANA, a N‐trifluoroacetyl derivative of DANA. Validation studies of the inhibitor with a detergent and a Lineweaver–Burk plot suggested that the 9‐substitution group would interact hydrophobically with the target loop moiety, adding a noncompetitive inhibition property to the DANA skeleton. This information enabled us to design compound 4 having the combined structure of 3c and FANA. Compound 4 showed the most potent inhibition (Ki=73 nM , mixed inhibition) of VCNA with high selectivity among the tested viral, bacterial, and mammal neuraminidases.  相似文献

Let Ω be a 2-dimensional bounded domain, symmetric with respect to the x2-axis. The boundary has several connected components, intersecting the x2-axis. The boundary value is symmetric with respect to the x2-axis satisfying the general outflow condition. The existence of the symmetric solution to the steady Navier–Stokes equations was established by Amick [2] and Fujita [4]. Fujita [4] proved a key lemma concerning the solenoidal extension of the boundary value by virtual drain method. We describe the potential of microchip electrophoresis with a Hitachi SV1210, which can be used to evaluate the integrity of total RNA, for the analysis of mRNA expression. The ribonuclease (RNase) protection assay was performed by using microchip electrophoresis with cyanine 5 (Cy5) labeled 248-base antisense RNA probe (riboprobe) encoding adipose-type fatty acid binding protein (A-FABP) as the riboprobe. The fluorescence intensity corresponding to the protected RNA fragment increased in a dose-dependent manner with respect to the complementary strand RNA. Results were obtained in 120 s, and the same amount of Cy5-labeled antisense riboprobe as used in the conventional method can be used. Furthermore, 8 times more sensitive detection of mRNA by microchip electrophoresis could be obtained. An obvious increase in the mRNA expression of A-FABP, which is known as a differentiation marker of adipocytes, occurred during the adipocyte differentiation of 3T3-L1 cells. These results clearly indicate the potential of microchip electrophoresis for the analysis of mRNA expression in cells.  相似文献

(1) Background: The obesity epidemic has been drastically progressing in both children and adults worldwide. Pharmacotherapy is considered necessary for its treatment. However, many anti-obesity drugs have been withdrawn from the market due to their adverse effects. Instead, natural products (NPs) have been studied as a source for drug discovery for obesity, with the goal of limiting the adverse effects. Zebrafish are ideal model animals for in vivo testing of anti-obesity NPs, and disease models of several types of obesity have been developed. However, the evidence for zebrafish as an anti-obesity drug screening model are still limited. (2) Methods: We performed anti-adipogenic testing using the juvenile zebrafish obesogenic test (ZOT) and mouse 3T3-L1 preadipocytes using the focused NP library containing 38 NPs and compared their results. (3) Results: Seven and eleven NPs reduced lipid accumulation in zebrafish visceral fat tissues and mouse adipocytes, respectively. Of these, five NPs suppressed lipid accumulation in both zebrafish and 3T3-L1 adipocytes. We confirmed that these five NPs (globin-digested peptides, green tea extract, red pepper extract, nobiletin, and Moringa leaf powder) exerted anti-obesity effects in diet-induced obese adult zebrafish. (4) Conclusions: ZOT using juvenile fish can be a high-throughput alternative to ZOT using adult zebrafish and can be applied for in vivo screening to discover novel therapeutics for visceral obesity and potentially also other disorders.  相似文献

Positron emission tomography is a noninvasive method for monitoring drug (or diagnostic) behavior and its localization on the target molecules in the living systems, including the human body, using a short‐lived positron‐emitting radionuclide. New methodologies for introducing representative short‐lived radionuclides, 11C and 18F, into the carbon frameworks of biologically active organic compounds have been established by developing rapid C‐[11C]methylations and C‐[18F]fluoromethylations using rapid Pd0‐mediated cross‐coupling reactions between [11C]methyl iodide (sp3‐hybridized carbon) and an excess amount of organotributylstannane or organoboronic acid ester having sp2(phenyl, heteroaromatic, or alkenyl), sp(alkynyl), or sp3(benzyl and cinnamyl)‐hybridized carbons; and [18F]fluoromethyl halide (iodide or bromide) and an organoboronic acid ester, respectively. These rapid reactions provide a firm foundation for an efficient and general synthesis of short‐lived 11C‐ or 18F‐labeled PET molecular probes to promote in vivo molecular imaging studies.  相似文献

A straightforward synthetic method for the construction of benzofuro[2,3‐b]pyrrol‐2‐ones by a novel domino reaction through a radical addition/[3,3]‐sigmatropic rearrangement/cyclization/lactamization cascade has been developed. The domino reaction of O‐phenyl‐conjugated oxime ether with an alkyl radical allows the construction of two heterocycles with three stereogenic centers as a result of the formation of two C?C bonds, a C?O bond, and a C?N bond in a single operation, leading to pyrrolidine‐fused dihydrobenzofurans, which are not easily accessible by existing synthetic methods. Furthermore, asymmetric synthesis of benzofuro[2,3‐b]pyrrol‐2‐one derivatives through a diastereoselective radical addition reaction to a chiral oxime ether was also developed.  相似文献
