The aim of this study was to detect the cleaved form of serine proteinase inhibitor (serpin), squamous cell carcinoma (SCC) antigen-1 in normal and malignant squamous epithelial tissues, which implies the presence of its target proteinase. The cleaved SCC antigen-1 in normal squamous epithelium was identified as a single spot with pI 6.35 and M(r) 40,000 by two-dimensional electrophoresis (2-DE) combined with immunoblotting. Interestingly, the cleaved form showed different biochemical properties in heat stability or immunoreactivity with a monoclonal antibody for SCC antigen (Mab 426) compared to intact SCC antigen-1. Furthermore, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis of tissue extracts showed an abundant 40 kDa band of cleaved SCC antigen-1 in tumor tissue compared to normal tissue. Among the potential target proteinase of SCC antigen-1, immunoblotting analyses revealed that cathepsin L2 was remarkably overexpressed in tumor tissue, while cathepsin L was expressed in both normal and tumor tissues. These findings indicate that SCC antigen-1 interacts with specific endogenous proteinases such as cathepsins L and L2 in physiological and pathological states of squamous epithelium.  相似文献

PCR experiments using DNA primers forming mismatch pairing with template lambda DNA at the 3' end were carried out in order to develop allele-specific primers capable of detecting SNP in genomes without generating pseudopositive amplification products, and thus avoiding the so-called pseudopositive problem. Detectable amounts of PCR products were obtained when primers forming a single or two mismatch pairings at the 3' end were used. In particular, 3' terminal A/C or T/C (primer/template) mismatches tended to allow PCR amplification to proceed, resulting in pseudopositive results in many cases. While less PCR product was observed for primers forming three terminal mismatch pairings, target DNA sequences were efficiently amplified by primers forming two mismatch pairings next to the terminal G/C base pairing. These results indicate that selecting a primer having a 3' terminal nucleotide that recognizes the SNP nucleotide and the next two nucleotides that form mismatch pairings with the template sequence can be used as an allele-specific primer that eliminates the pseudopositive problem. Trials with the human ABO genes demonstrated that this primer design is also useful for detecting a single base pair difference in gene sequences with a signal-to-noise ratio of at least 45.  相似文献

We have prepared four light‐emitting polymers bearing a chromophore composed of carbazole and fluorene by the Suzuki coupling polycondensation. Two nonconjugated polymers (P3CzBFXy and P2CzFXy) had a chromophore tethered by the p‐xylylene spacer, whose connection point between carbazole and fluorene in addition to the number of fluorene unit was systematically changed to investigate the emission wavelength and intensity. The red‐shifted absorption and emission maximum wavelengths together with the improved fluorescence quantum yield of polymers P3CzBFXy and P2CzFXy indicate that the increment of the number of para‐connected benzene rings included in the chromophore effectively extends the conjugation length. The fact that polymer P3CzBFXy has longer wavelength absorption and emission spectra also indicates the interaction of the carbazole nitrogen lone pair with the oligophenylene moiety. Other two polymers P3CzFPy and P3CzFPym having the heterocycle directly bound to the carbazole nitrogen were prepared to know the character of the carbazole nitrogen lone pair and their influence on the fluorescence behavior. The fluorescence spectra of polymer P3CzFPym bearing the pyrimidine ring gradually red‐shifted in conjunction with the decrease of fluorescence quantum yield on going from toluene solution to CHCl3 solution because of the intramolecular charge transfer at the excited state. © 2010 Wiley Periodicals, Inc. The dynamic properties of polyethylene glycol (PEG) molecules on the solid–liquid interface oscillating at MHz were investigated using the quartz crystal microbalance (QCM). The number-average molecular weights (Mn) of the PEG molecules were systematically varied over 4 orders of magnitude. This study makes it clear that the series-resonant frequency shift, ΔF, of the QCM against the square root of the density–viscosity product of the PEG solution is linear and has the intercept. Moreover, systematical analysis reveals that the ΔF slope rapidly decreases with Mn and that the ΔF intercept becomes constant above 4.0 × 103 g mol−1. As a result, those reveal that the resonant length of the PEG molecule moving with the oscillating plate of 9 MHz is 54.2 Å. We also find that the behaviors of ΔF due to Mn are mainly caused by the length of the PEG molecule.  相似文献

A G-quadruplex is a four-stranded DNA structure featuring stacked guanine tetrads, G-quartets. Formation of a G-quadruplex in telomere DNA can inhibit telomerase activity; therefore, development of G-quadruplex-ligands, which induce and/or stabilize G-quadruplexes, has become an area of great interest. Phthalocyanine derivatives have substantial potential as high-affinity G-quadruplex-ligands because these planar chromophores are similar in size and shape to the G-quartets. Here, we focus on the latest findings on phthalocyanine derivatives as G-quadruplex-ligands, and discuss the mechanisms by which phthalocyanines bind to G-quadruplexes with high affinity and selectivity. We also discuss potential biomedical and organic electronic applications of phthalocyanines that are dependent on their photophysical properties.  相似文献

We have synthesized novel σπ conjugated polymers with an alternating organosilanylene and π‐electron system, intending to utilize them for hole‐transporting materials of electroluminescent (EL) devices. 3,6‐Di(lithioethynyl)carbazoles were co‐polymerized with organodichlorosilanes to give the corresponding polymers with molecular weights of MW = 2000–5000. Another type of polymer with a thienylene unit was also synthesized by the nickel‐catalyzed reaction of the di‐Grignard reagent of 1,2‐bis[2‐(5‐bromothienyl)]tetraethyldisilane with 3,6‐dibromocarbazole, the molecular weight being Mn = 3100. The EL devices with a double‐layer system composed of tris(8‐quinolinolato)aluminum(III) and the present polymers as the emitting‐electron‐transporting and hole‐transporting layers, respectively, emit green EL with a maximum intensity of the order of 103 cd m?2. Of these, the device with the thienylene–carbazole polymers exhibited the highest luminance of 1480 cd m?2. Copyright © 2001 John Wiley & Sons, Ltd.  相似文献

This paper proposes interferometric 3-D imaging based on retrieving the space variant cross-spectral density. The cross-spectral density across an arbitrary transverse plane can be retrieved by using the propagation law described in the Fourier domain. The 3-D intensity distribution is reconstructed from a series of the cross-spectral densities across planes within the slab geometry considered. Results of an experiment conducted by incorporating two point sources are also reported. The point spread function of the proposed interferometric imaging system, and its similarity to an ordinary imaging system are discussed. To detect spatially incoherent sources, we also propose an incoherence gating method based on the present interferometric imaging technique. The second-order differential of the series of retrieved cross-spectral densities with respect to a two-point separation enable us to estimate field correlations over the 3-D field, and locations of primary sources are clearly determined.  相似文献

A novel scheme to achieve broadband cascaded third-harmonic generation by use of two-dimensional quasi-phase-matching gratings is proposed. Intrinsic group-velocity mismatch is compensated by the pulse front tilt in noncollinear interaction geometry. The presented scheme enables broadband third-harmonic generation in a single device of long interaction length, which reduces the operation intensity dramatically.  相似文献
