A N(1)-C(5) bridged chiral bicyclic imidazole with a morpholine framework was synthesized from an enantiopure 2-amino alcohol. The resultant imidazole reacted with various electrophiles, including primary and secondary alkyl halides, benzyne, and an electron-deficient aryl halide, to give the corresponding imidazolium salts. Some of the imidazolium salts were found to have potential as the precursor of a chiral N-heterocyclic carbene catalyst; by the direct annulation of an enal and a ketone through the intermediacy of a homoenolate and an activated carboxylate, the target lactone was obtained in an enantiomerically enriched form (up to 66% ee).  相似文献

Giant liposomes, or giant vesicles, are cell-size (approximately 5-100 microm) compartments enclosed with phospholipid bilayers, and have often been used in biological research. They are usually generated using hydration methods, "electroformation" and "gentle hydration (or natural swelling)", in which dry lamellar films of phospholipids are hydrated with aqueous solutions. In gentle hydration, however, giant liposomes are difficult to prepare from an electrostatically neutral phospholipid because lipid lamellae cannot repel each other. In this study, we demonstrate the efficient formation of giant liposomes using the gentle hydration of neutral phospholipid (dioleoyl phosphatidylcholine, DOPC) dry films doped with nonelectrolytic monosaccharides (glucose, mannose, and fructose). A mixture of DOPC and such a sugar in an organic solvent (chloroform/methanol) was evaporated to form the films, which were then hydrated with distilled water or Tris buffers containing sodium chloride. Under these conditions, giant liposomes spontaneously formed rapidly and assumed a swollen cell-sized spherical shape with low lamellarity, whereas giant liposomes from pure DOPC films had multilamellar lipid layers, miscellaneous shapes and smaller sizes. This observation indicates that giant unilamellar vesicles (GUVs) of DOPC can be obtained efficiently through the gentle hydration of sugar-containing lipid dry films because repulsion between lipid lamellae is enhanced by the osmosis induced by dissolved sugar.  相似文献

Well‐defined alkoxysilane oligomers containing a cagelike carbosiloxane core were synthesized and used as novel building blocks for the formation of siloxane‐based hybrid networks. These oligomers were synthesized from the cagelike trimer derived from bis(triethoxysilyl)methane by silylation with mono‐, di‐, and triethoxychlorosilanes ((EtO)nMe3?nSiCl, n=1, 2, and 3). Hybrid xerogels were prepared by hydrolysis and polycondensation of these oligomers under acidic conditions. The structures of the products varied depending on the number of alkoxy groups (n). When n=2 and 3, microporous xerogels (BET surface areas of 820 and 510 m2 g?1, respectively) were obtained, whereas a nonporous xerogel was obtained when n=1. 29Si NMR spectroscopic analysis suggested that partial rearrangement of the siloxane networks, which accompanied the cleavage of the Si–O–Si linkages, occurred during the polycondensation processes. By using an amphiphilic triblock copolymer surfactant as a structure‐directing agent, hybrid thin films with a 2D hexagonal mesostructure were obtained when n=2 and 3. These results provide important insight into the rational synthesis of molecularly designed hybrid materials by sol–gel chemistry.  相似文献

Mere chemical generation of ceramide and related double-chain lipids in the membrane of small unilamellar vesicles (SUVs) induces fusion of the vesicles. The lipids can be successfully prepared by dehydrocondensation between single-chain lipids (fatty acids and sphingosine or its analogues) in a lipid bilayer of the SUV by using a combination of 2-chloro-4,6-dimethoxy-1,3,5-triazine and amphiphilic tertiary amine catalysts, a process that can be compared to a successive enzyme model system for a fatty acyl-CoA synthetase followed by acyltransferase. The SUV spontaneously undergoes membrane fusion upon this internal chemical stimulation by the artificial enzyme system.  相似文献

Thermal degradation behavior of poly(4-hydroxybutyric acid) (P(4HB)) was investigated by thermogravimetric and pyrolysis-gas chromatography mass spectrometric analyses under both isothermal and non-isothermal conditions. Based on the thermogravimetric analysis, it was found that two distinct processes occurred at temperatures below and above 350 °C during the non-isothermal degradation of P(4HB) samples depending on both the molecular weight and the heating rate. From 1H NMR analysis of the residual P(4HB) molecules after isothermal degradations at different temperatures, it was confirmed that the ω-hydroxyl chain-end was remained unchanged in the residual P(4HB) molecules at temperatures below 300 °C, while the ω-chain-end of P(4HB) molecules was converted to 3-butenoyl units at temperatures above 300 °C. In contrast, the majority of the volatile products evolved during thermal degradation of P(4HB) was γ-butyrolactone regardless of the degradation temperature. From these results, it is concluded that during the thermal degradation of P(4HB), the selective formation of γ-butyrolactone via unzipping reaction from the ω-hydroxyl chain-end predominantly occurs at temperatures below 300 °C. At temperatures above 300 °C, both the cis-elimination reaction of 4HB unit and the formation of cyclic macromolecules of P(4HB) via intramolecular transesterification take place in addition to unzipping reaction from the ω-hydroxyl chain-end. Finally, the primary reaction of thermal degradation of P(4HB) at temperatures above 350 °C progresses by the cyclic rupture via intramolecular transesterification of P(4HB) molecules with a release of γ-butyrolactone as volatile product. Moreover, we carried out the thermal degradation tests for copolymer of 93 mol% of 4HB with 7 mol% of 3-hydroxybutyric acid (3HB) to examine the effect of 3HB units on thermal stability of P(4HB).  相似文献

Hydrogen atom abstraction reactions have been implicated in oxygenation reactions catalyzed by copper monooxygenases such as peptidylglycine alpha-hydroxylating monooxygenase (PHM) and dopamine beta-monooxygenase (DbetaM). We have investigated mononuclear copper(I) and copper(II) complexes with bis[(6-neopentylamino-2-pyridyl)methyl][(2-pyridyl)methyl]amine (BNPA) as functional models for these enzymes. The reaction of [Cu(II)(bnpa)]2+ with H2O2, affords a quasi-stable mononuclear copper(II)-hydroperoxo complex, [Cu(II)(bnpa)(OOH)]+ (4) which is stabilized by hydrophobic interactions and hydrogen bonds in the vicinity of the copper(II) ion. On the other hand, the reaction of [Cu(I)(bnpa)]+ (1) with O2 generates a trans-mu-1,2-peroxo dicopper(II) complex [Cu(II)2(bnpa)2(O2(2-]2+ (2). Interestingly, the same reactions carried out in the presence of exogenous substrates such as TEMPO-H, produce a mononuclear copper(II)-hydroperoxo complex 4. Under these conditions, the H-atom abstraction reaction proceeds via the mononuclear copper(II)-superoxo intermediate [Cu(II)(bnpa)(O2-)]+ (3), as confirmed from indirect observations using a spin trap reagent. Reactions with several substrates having different bond dissociation energies (BDE) indicate that, under our experimental conditions the H-atom abstraction reaction proceeds for substrates with a weak X-H bond (BDE < 72.6 kcal mol(-1)). These investigations indicate that the copper(II)-hydroperoxo complex is a useful tool for elucidation of H-atom abstraction reaction mechanisms for exogenous substrates. The useful functionality of the complex has been achieved via careful control of experimental conditions and the choice of appropriate ligands for the complex.  相似文献

Retro-allylation of homoallyl alcohol by rhodium catalysis occurs to generate allylrhodium species. This allylrhodium reacts with aldehydes to give the corresponding secondary alcohols in situ. Biologically important and structurally unique marine natural products avarone (1), avarol (2), neoavarone (3), neoavarol (4) and aureol (5), were efficiently synthesized in a unified manner starting from (+)-5-methyl-Wieland-Miescher ketone 10. The synthesis involved the following crucial steps: i) Sequential BF(3)Et(2)O-induced rearrangement/cyclization reaction of 2 and 4 to produce 5 with complete stereoselectivity in high yield (2 --> 5 and 4 --> 5); ii) strategic salcomine oxidation of the phenolic compounds 6 and 8 to derive the corresponding quinones 1 and 3 (6 --> 1 and 8 --> 3); and iii) Birch reductive alkylation of 10 with bromide 11 to construct the requisite carbon framework 12 (10 + 11 --> 12). An in vitro cytotoxicity assay of compounds 1-5 against human histiocytic lymphoma cells U937 determined the order of cytotoxic potency (3 > 1 > 5 > 2 > 4) and some novel aspects of structure-activity relationships.  相似文献

Effects of trimethylsilyl, trimethylgermyl, and trimethylstannyl substituents attached to fused aromatic hydrocarbons such as pyrene, anthracene, phenanthrene, and naphthalene were studied in terms of UV absorption and fluorescence properties in aerated cyclohexane solutions. Absorption maxima of trimethylsilyl-, trimethylgermyl-, and trimethylstannyl-substituted aromatic hydrocarbons shifted to longer wavelengths than those of unsubstituted ones. Absorption maxima of mono-, bis-, tris-, and tetrakis(trimethylsilyl)pyrenes shifted to longer wavelength consecutively at intervals of 10 nm. Fluorescence intensities and fluorescence lifetimes of trimethylsilyl-substituted aromatic hydrocarbons were larger and longer than those of unsubstituted ones, and they decreased in the order of Me3SiAr > Me3GeAr > Me3SnAr. Fluorescence intensity of 1,3,6,8-tetrakis(trimethylsilyl)pyrene was largest among those of a series of mono-, bis-, tris-, and tetrakis(trimethylsilyl)pyrenes under aerated conditions.  相似文献
