Mycalolides are tris-oxazole macrolides isolated from the marine sponge Mycale sp., which shows cytotoxic, antifungal, and actin-depolymerizing activities. To develop an efficient synthetic route of mycalolides and to evaluate its functional mechanism of biological activities, tris-oxazole macrolactone analogs of mycalolides were synthesized through the use of ring-closing metathesis (RCM). The presence/absence of protecting groups at C3, solvent polarity, and reaction temperature significantly affected the stereoselectivity of RCM (E/Z=2.5/1.0–1.0/2.5). The 19E- and 19Z-stereoisomers both exhibited moderate cytotoxicity against tumor cells, but neither showed significant actin-depolymerizing properties or antimycotic activity against pathogenic fungi. Thus, both the side-chain (actin-binding) moiety and the macrolactone moiety were suggested to be essential for the potent biological activities of the parent molecules. 相似文献

Abstract Single halogen atom (i. e. I, Br, Cl and F) substituted calix[4]pyrroles, compounds 2, 3, 4 and 5, were synthesized. Studies of these systems reveal that replacement of a single β-pyrrolic hydrogen atom can increase the anion binding ability of calix[4]pyrroles for a variety of anions (e. g. Cl?, Br?, H2PO4 ? and HSO? 4) relative to normal non-halogen substituted calix[4]pyrrole 1. In the case of chloride anion, the expected relative affinity sequence of 5 > 4 > 3 > 2 was observed. This was not found to be true for Br?, H2PO? 4, and HSO? 4. Here, the chlorine substituted calix[4]pyrrole 4 was found to display a slightly higher affinity in the case of each anion than the fluorine-bearing derivative 5. This was rationalized in terms of intermolecular NH … F hydrogen bonding interactions being present in CD2Cl2 solutions of 5. Support for this latter conclusion came from concentration and temperature-dependent NMR spectroscopic studies. A matched set of mono halogen substituted calix[4]pyrroles was used to study in detail, the extent to which halogen substituents may be used to fine-tune the anion binding properties of calix[4]pyrroles. 相似文献

We describe the HPLC separation and identification of N@C60 and N2@C60. These species were observed after eleven sequential HPLC separations. Their retention times are in the same range as those of the other noninteractive endohedral species of C60, such as noble gas endohedral C60. The separation factors of these endohedrals were evaluated by using a mixture of hexane/toluene as eluent. We note that this is the first evidence for the N2@C60 molecule existing in the form of endohedral C60 complex. 相似文献

The C42-C52 part of ciguatoxin CTX3C (1) was synthesized from tri-O-acetyl d-glucal. The synthetic segment had a tetrahydropyran ring corresponding to the ‘C49-reduced’ L-ring of 1, designed to avoid side reactions due to acid-labile C49 acetal carbon during acidic reductive conditions planned in further synthesis toward 1. The vicinal dimethyl part at C47-C48 was constructed by a stepwise conjugate addition/methylation procedure. The C50-C52 unit was installed by Grignard addition of the C3 unit followed by spirocyclization and reductive cleavage of the spirocyclic acetal. In recent experimental studies of infrared (IR) spectra of hydrated phenol clusters, the Mikami and Ebata group at Tohoku University attributed apparently two very different spectra to PhOH(H2O)4. The first spectrum has a wide transparent region between 3500 and 3700 cm-1, which they named a window region. The second spectrum has several absorption bands in this window region. Our previous study revealed that the first spectrum was assigned to the isomers which have a single-ring structure of a hydrogen-bonding network of OH's of phenol and waters. The experimentalists suggested that the second spectrum might be identified to a proton-transferred phenol(SINGLEBOND)water cluster. In the present article, the geometrical structures of proton-transferred hydrated phenol clusters were determined with the ab initio molecular orbital method and their IR spectra were calculated. The proton-transferred phenol(SINGLEBOND)water cluster is at a stable local minimum, but the energy is much higher than that of the most stable nontransferred cluster. The effect of preferential wetting of one of the constituent block chains and corresponding block copolymer morphologies to a carbon substrate is studied from a molecular level. The single chain distribution of the block copolymer was estimated as a function of the distance from the substrate by a combined method of transmission electron microtomography (TEMT) and self-consistent field (SCF) simulation. The former provides three-dimensional (3D) morphological information of cylindrical microdomains near the surface, while the latter utilizes the 3D morphology to quantitatively determine the interaction between the block chains and substrate, which is further used to estimate the single chain distribution of one of the block chains, i.e., the subchain, of the matrix. It was found that the subchains in the vicinity of the wetting layer are substantially compressed, while the radius of gyration of the subchain at a distance L (L is the interlayer distance of the cylindrical microdomains from the substrate) has already reached the same value as that in the bulk, indicating that the propagation of the surface interaction is limited to one layer. The methodology developed in this study can be used not only to estimate the surface effect on polymer chains for a variety of different surfaces, but also to provide a means to understand complicated block copolymer morphologies from a molecular level. 相似文献
