The direct measurement of the circular dichroism (CD) spectra of liquid/liquid interface has been achieved for the first time by the centrifugal liquid membrane (CLM) method combined with a conventional CD spectropolarimetry. In the sample chamber of the CD spectropolarimeter, a cylindrical glass cell containing small amounts of organic and aqueous phases was rotated at ca. 7000 rpm to generate a two-phase liquid membrane with a high specific interfacial area. The CD spectra of the J-aggregate of protonated 5,10,15,20-tetraphenylporphyrin formed at the toluene/sulfuric acid interface in the rotating cell have been measured. In proportion to the environmental pollution problems caused by organotin compounds, the genotoxicities of tin compounds in the environments have become of interest so as to estimate their safety in recent years. In this work, isolated λ-DNA (double-strand DNA) was incubated with inorganic tin(II) and tin(IV) and five organotin compounds [n-butyltin trichloride, di(n-butyltin) dichloride, methyltin trichloride, dimethyltin dichloride and trimethyltin chloride] in reaction systems both with and without hydrogen peroxide (H2O2) content. The tin compounds tested in this study did not induce DNA breakage in the absence of hydrogen peroxide. Divalent inorganic tin (SnCl2) and tetravalent inorganic tin (SnCl4) caused DNA breakage in the presence of hydrogen peroxide (10 mM), and the DNA damage activity of inorganic tin was much more potent in divalent inorganic tin (SnCl2) than in tetravalent inorganic tin (SnCl4). Divalent inorganic tin (SnCl2) induced DNA breakage in a concentration-dependent fashion at concentrations greater than 0.1 mM of SnCl2 in the presence of hydrogen peroxide (10 mM). DNA breakage was not caused by n-butyltin compounds and methyltin compounds either in the presence or in the absence of hydrogen peroxide. 相似文献

A new type of atomic absorption spectrometry using the Zeeman effect of sample materials is proposed. A magnetic field was applied to the sample vapor in the direction perpendicular to the propagation of light emitted from an atomic spectral source. Absorption of radiation polarized perpendicular and parallel to the magnetic field was observed alternatively. The absorption difference was proportional to the true atomic absorption, and was not interfered with by any other molecular absorption and light scattering, i.e., background absorption. The background absorption could be monitored at exactly the same wavelength as an atomic absorption line. Suitable magnetic field strength was found for each line of the various elements. 相似文献

The construction of a liquid-membrane type periodate ion-selective electrode and its application to the potentiometric titration of alpha-diols and alpha-amino-alcohols are described. The ion-pair of periodate anion with Capriquat (tri-n-octylmethylammonium chloride), is easily extracted into nitrobenzene, and this extract is employed as a liquid ion-exchange membrane. The calibration curve shows Nernstian response towards periodate ion over the concentration range from 10(-1)M to 10(-7)M with a slope of 60 mV/pIO(-)(4). Selectivity coefficients with respect to various ions were evaluated. The electrode potential was independent of pH in the range 2.5-7.5. Some alpha-diols and monoethanolamine were successfully titrated potentiometrically with the aid of the present electrode. 相似文献

The novel ring transformation reactions were found in the reactions of 1,3,7,9-tetra-alkyl-8,9-dihydroxanthines and acetylenic compounds. The reaction of the dihydroxanthine with DMAD gave a propellane type compound and with methyl propiolate afforded the similar type compound and a pyrimido[4,5-b]diazepine derivative. Myoglobin will be a good scaffold for engineering a function into proteins. To modulate the physiological function of myoglobin, almost all approaches have been demonstrated by site-directed mutagenesis, however, there are few studies which show a significant improvement in the function. In contrast, we focused on the replacement of heme in the protein with an artificial prosthetic group. Recently, we prepared a novel myoglobin reconstituted with an iron porphycene as a structural isomer of mesoheme. The bluish colored reconstituted myoglobin is relatively stable and the deoxymyoglobin reversibly binds ligands. Interestingly, the O2 affinity of the reconstituted myoglobin, 1.1 x 109 M-1, is a significant 1,400-fold higher than that of the native myoglobin. Furthermore, the unfavorable autoxidation kinetics show 7-fold decrease in rate for the reconstituted myoglobin relative to the native myoglobin, indicating the stable oxy-form against autoxidation. The net results come from the slow dissociation of the O2 ligand in the reconstituted myoglobin, koff = 0.11 s-1, because of the formation of strong hydrogen bond between His64 and negatively charged dioxygen. The present study indicates that the replacement of native heme with an artificially created prosthetic group will give us a unique function into a hemoprotein. 相似文献

The sexual development of the fungus Phycomyces is inhibited by light. The action spectra for this photoinhibitory effect were determined for 48 h continuous exposure between 350 and 700 nm wavelengths during the mating process. Effective wavelengths were shorter than 490 nm, but the most effective wavelengths depended on the stage of sexual development. In early stages of progametangium formation, the major peaks appeared near 360 nm with small shoulders at 410 nm, but in later stages, after gametangium formation, only single peaks were detected in the UVA range (350–390 nm). Low-fluence irradiation in the later stage, however, revealed inhibitory effectiveness at 370–410 nm, implying the existence of a dual photoresponse and multiple regulatiory systems in the mating process of Phycomyces. 相似文献
