A rapid, selective and sensitive procedure for the analysis of Polycyclic Aromatic Hydrocarbons (PAHs) in biological organisms has been developed and validated. The freeze-dried tissues were digested in ethanolic KOH. After solvent partitioning, the extract was purified on alumina and silica micro-columns and finally analyzed by gas chromatography coupled to mass spectrometry (GC-MS). The final extract was free of most of the endogenous compounds. A quality assurance/quality control (QA/QC) was carried out. The absolute loss of the target compounds at different stages of the analytical procedure and during all of the procedure was estimated (losses<45%). The recoveries of PAHs were estimated using a certified standard compound solution (recoveries>90%) and spiked mussels (recoveries=100±6%). For validation a certified reference material mussel tissue (recoveries= 90±16%) was analyzed.  相似文献

Anionic graft copolymers were synthesized through grafting of poly(ethylene glycol) monomethyl ether (MPEG) onto terpolymers containing succicinic anhydride groups. The backbone polymers were prepared through radical terpolymerization of maleic anhydride, styrene, and one of the following monomers: methyl methacrylate, ethylhexyl methacrylate, and diethyl fumarate. MPEG of different molecular weights were grafted onto the backbone through reactions with the cyclic anhydride groups. In this reaction one carboxylic acid group is formed together with each ester bond. The molecular weights of MPEG were found to influence the rate of the grafting reaction and the final degree of conversion. The graft copolymers were characterized by IR, GPC, and 1H-NMR. Thermal properties were examined by DSC. Graft copolymers containing 50% w/w of MPEG 2000 grafts were found to be almost completely amorphous, presumably because of crosslinking, and hydrogen bonding between carboxylic acid groups in the backbone and the ether oxygens in MPEG grafts. © 1995 John Wiley & Sons, Inc.  相似文献

The reduction of a single-layer FeO film grown on Pt(111) by CO at elevated pressures and temperatures has been studied through an interplay of scanning tunneling microscopy, ambient-pressure X-ray photoelectron spectroscopy, and density functional theory calculations. Exposure of the FeO thin film to CO at pressures between 1 and 30 Torr and temperatures between 500 and 530 K leads to formation of a honeycomb-structured Fe(3)O(2) film with hollow sites occupied by single Pt atoms extracted from the substrate surface. The formation of these adatoms is driven by an increase in CO adsorption energy. In addition, the structure incorporates undercoordinated Fe centers, which are proposed to have substantial effects on the catalytic properties of the surface.  相似文献

New heterobimetallic tetranuclear complexes of formula [Fe(III){B(pz)(4)}(CN)(2)(μ-CN)Mn(II)(bpy)(2)](2)(ClO(4))(2)·CH(3)CN (1), [Fe(III){HB(pz)(3)}(CN)(2)(μ-CN)Ni(II)(dmphen)(2)](2)(ClO(4))(2)·2CH(3)OH (2a), [Fe(III){B(pz)(4)}(CN)(2)(μ-CN)Ni(II)(dmphen)(2)](2)(ClO(4))(2)·2CH(3)OH (2b), [Fe(III){HB(pz)(3)}(CN)(2)(μ-CN)Co(II)(dmphen)(2)](2)(ClO(4))(2)·2CH(3)OH (3a), and [Fe(III){B(pz)(4)}(CN)(2)(μ-CN)Co(II)(dmphen)(2)](2)(ClO(4))(2)·2CH(3)OH (3b), [HB(pz)(3)(-) = hydrotris(1-pyrazolyl)borate, B(Pz)(4)(-) = tetrakis(1-pyrazolyl)borate, dmphen = 2,9-dimethyl-1,10-phenanthroline, bpy = 2,2'-bipyridine] have been synthesized and structurally and magnetically characterized. Complexes 1-3b have been prepared by following a rational route based on the self-assembly of the tricyanometalate precursor fac-[Fe(III)(L)(CN)(3)](-) (L = tridentate anionic ligand) and cationic preformed complexes [M(II)(L')(2)(H(2)O)(2)](2+) (L' = bidentate α-diimine type ligand), this last species having four blocked coordination sites and two labile ones located in cis positions. The structures of 1-3b consist of cationic tetranuclear Fe(III)(2)M(II)(2) square complexes [M = Mn (1), Ni (2a and 2b), Co (3a and 3b)] where corners are defined by the metal ions and the edges by the Fe-CN-M units. The charge is balanced by free perchlorate anions. The [Fe(L)(CN)(3)](-) complex in 1-3b acts as a ligand through two cyanide groups toward two divalent metal complexes. The magnetic properties of 1-3b have been investigated in the temperature range 2-300 K. A moderately strong antiferromagnetic interaction between the low-spin Fe(III) (S = 1/2) and high-spin Mn(II) (S = 5/2) ions has been found for 1 leading to an S = 4 ground state (J(1) = -6.2 and J(2) = -2.7 cm(-1)), whereas a moderately strong ferromagnetic interaction between the low-spin Fe(III) (S = 1/2) and high-spin Ni(II) (S = 1) and Co(II) (S = 3/2) ions has been found for complexes 2a-3b with S = 3 (2a and 2b) and S = 4 (3a and 3b) ground spin states [J(1) = +21.4 cm(-1) and J(2) = +19.4 cm(-1) (2a); J(1) = +17.0 cm(-1) and J(2) = +12.5 cm(-1) (2b); J(1) = +5.4 cm(-1) and J(2) = +11.1 cm(-1) (3a); J(1) = +8.1 cm(-1) and J(2) = +11.0 cm(-1) (3b)] [the exchange Hamiltonian being of the type H? = -J(S?(i)·S?(j))]. Density functional theory (DFT) calculations have been used to substantiate the nature and magnitude of the exchange magnetic coupling observed in 1-3b and also to analyze the dependence of the exchange magnetic coupling on the structural parameters of the Fe-C-N-M skeleton.  相似文献

A new time-optimal rheometry technique is presented. It consists in applying to the material a continuous exponential frequency sweep and analyzing its response by means of Fourier transforms. The properties of this method are that it takes the least possible time to perform the linear viscoelastic measurement in a given frequency range, and also presents an optimal signal to noise ratio in Fourier space. After validation against classical methods, it is used to characterize the gelation of a dental alginate. Properly time-resolved measurements of the frequency dependent viscoelastic modulii are presented. This allows to evaluate rigorously the gel point, using the Winter and Chambon criterion. Analysis of the data shows that a fractal networks forms inside the material after a lag time of about 350s, with a subsequent fast evolution (less than two minutes) towards the final structure.  相似文献

Tumors grown on animals and treated with magnetic drug targeting and magnetic hyperthermia have been analyzed by microcomputed X-ray tomography to study the three-dimensional nanoparticle distribution. The measurements have been performed in two laboratories, with a polychromatic X-ray cone beam as well as with monochromatic parallel beam. Due to the poor resolution in the first case, the distribution of the magnetic nanoparticles can be studied only qualitatively. With the polychromatic beam semi-quantitative results can be achieved. In this paper, the results from both methods are presented and compared.  相似文献

Electron microscopy has proven to be a reliable and essential tool to determine morphological alterations and target organelles in the investigation of new drugs for Chagas disease. In this review, we focused on evaluating different agents that induce death of Trypanosoma cruzi, i.e. lysophospholipids analogues, naphthoquinones and derivatives, cytoskeletal inhibitors and natural products. Apoptosis-like presents as morphological characteristics DNA fragmentation, membrane blebbing and apoptotic body formation. Autophagy involves autophagosome formation, with the appearance of membranes surrounding organelles and cytosolic structures. Necrosis causes the loss of osmotic balance, an increase of cytoplasmic vacuolization and plasma membrane disruption. Mitochondrion appears as a central checkpoint in both apoptosis and necrosis. Our evidences of ultrastructural changes to T. cruzi treated with the different classes of compounds point to dramatic mitochondrial alterations and similar autophagic phenotypes. Lysophospholipid analogues interfere in the lipid biosynthesis in epimastigotes, altering the amount of both phospholipids and sterols, and consequently the physical properties of the membrane. Naphthoquinone derivatives led to a strong DNA fragmentation in trypomastigotes and to the release of cysteine proteases from reservosomes to cytosol in epimastigotes, starting a proteolytic process which results in parasite death. The susceptibility of reservosomes was also observed in parasites treated with propolis, suggesting impairment of lipid metabolism, compromising membrane fluidity and leading to lysis. The cytoskeletal agents blocked mitosis of epimastigotes, arresting cell cycle and impairing the parasite proliferation. The variety of drug stimuli converge to the same pathway of death suggests an intense cross-talking between the three types of PCD in the protozoa.  相似文献

Toxoplasma gondii is an apicomplexan parasite infecting a broad host range, including humans. The parasite invades host cell by active penetration with the participation of its secretory organelles proteins during this process. Until now, only a limited number of secretory proteins have been discovered, and the effectors molecules involved in parasite invasion and survival are not well understood. Osteopontin (OPN) is a multifunctional glycophosphoprotein, secreted by different cell types, which is involved in various physiological and pathological events including cell signaling and survival. For the first time we demonstrated in this work by immunofluorescence and immunoelectron microscopy approaches the localization of an OPN-like protein in dense granules of extracellular T. gondii tachyzoites. Western blotting and RT-PCR confirmed this protein expression by the parasites. Our results also showed, after macrophage invasion, an intense positive labeling for OPN-like protein at the sub-apical portion of tachyzoites, the site of dense granules secretion, and the localization of this protein at the parasitophorous vacuole membrane. These data suggest that dense granules secrete an OPN-like protein, and we speculate that this protein participates during the parasite interaction process with host cells and parasitophorous vacuole formation.  相似文献
